Le message de Luther est diffusĂ© dâabord par la rĂ©impression dâun certain nombre de ses Ă©crits. Entre 1519 et 1525, des textes de Luther sont publiĂ©s 168 fois Ă Strasbourg, ainsi que dâautres Ă©manant dâauteurs proches de Luther tels que ceux Bucer. Un seul Ă©diteur, GrĂŒninger, rĂ©siste Ă la vague luthĂ©rienne. TouchĂ©s par ces publications, des clercs mais aussi des laĂŻcs se rallient au mouvement Ă©vangĂ©lique. Mais câest surtout Ă travers la prĂ©dication dâun certain nombre de pasteurs adhĂ©rant aux idĂ©es de Luther, tels que Matthieu Zell, que la grande majoritĂ© des fidĂšles va ĂȘtre convaincue par le message luthĂ©rien, dans les villes dâabord, puis Ă la campagne. Tout en reprenant les conceptions des rĂ©formateurs de Wittenberg au sujet de la justification par la foi, de lâautoritĂ© de lâĂcriture sainte, de la messe et du sacerdoce de tous les fidĂšles, les prĂ©dicateurs en place ainsi que dâautres, autoproclamĂ©s, tels que ClĂ©ment Ziegler, mettent surtout lâaccent sur les fruits de la foi, en particulier lâamour et exigent la suppression dâun certain nombre dâinstitutions et de pratiques. De larges couches de la population des villes se rallient au mouvement ; lâopposition, reprĂ©sentĂ©e en particulier par de nombreux clercs dont le fougueux franciscain Thomas Murner, va ĂȘtre marginalisĂ©e. Ă la campagne, le message luthĂ©rien est souvent associĂ© aux revendications paysannes qui vont conduire au soulĂšvement de 1525. De maniĂšre gĂ©nĂ©rale, la rĂ©ception du mouvement Ă©vangĂ©lique entraĂźne des changements qui concernent le culte, le statut des pasteurs, la fermeture des couvents et une plus grande implication des autoritĂ©s civiles dans lâorganisation et la vie des Ăglises. The same goes for authors closely related to Luther, such as Bucer. One publisher only, GrĂŒninger, could withstand the wave of Lutheranism. Showing an interest for such publications, both clerics and lay people joined the evangelical movement. But is is mainly the sermons of Luther partisans, such as Matthieu Zell, that managed to convince most of the faithful, first in cities, then in smaller places. While propa-gating the doctrine of the Wittenberg theologians concerning the justification through faith, the authority of the Bible, the mass and the priestly ministry of any faithful, the official clergymen or self-appointed ones like ClĂ©ment Ziegler would underscore the benefits of faith, mainly that of love, and required that certain institutions and religious practices be abandoned. The movement rallied a high percentage of the urban population; the opponents, in fact many clerics, among them the passionate Francis-can Thomas Murner, were to be marginalized. In the countryside the Lutheran mes-sage would frequently merge with the peasantsâ claims which were to foster the 1525 revolt. As a rule, the way the evangelical movement was received was to entail changes concerning the form of worship, the status of the clergy the closing down of convents and a more deliberate involvement of civil authorities in the organisation of Church life. Breite Schichten der stĂ€dtischen Bevölkerung schlieĂen sich der Bewegung an; die Opposition, ĂŒberwiegend durch Kleriker vertreten, wie dem ungestĂŒmen Franziskaner Thomas Murner, wird zur Randerscheinung. Auf dem Land wird die lutherische Botschaft oft mit den Forderungen der Bauern verbunden, was zu den AufstĂ€nden von 1525 fĂŒhren wird. Das Annehmen des evangelischen Glaubens hat verschiedene VerĂ€nderungen zur Folge. Therapeutic targeting of ependymoma as informed by oncogenic enhancer profiling
Genomic sequencing has driven precision-based oncology therapy; however, the genetic drivers of many malignancies remain unknown or non-targetable, so alternative approaches to the identification of therapeutic leads are necessary. Ependymomas are chemotherapy-resistant brain tumours, which, despite genomic sequencing, lack effective molecular targets. Intracranial ependymomas are segregated on the basis of anatomical location (supratentorial region or posterior fossa) and further divided into distinct molecular subgroups that reflect differences in the age of onset, gender predominance and response to therapy1,2,3. The most common and aggressive subgroup, posterior fossa ependymoma group A (PF-EPN-A), occurs in young children and appears to lack recurrent somatic mutations2. Conversely, posterior fossa ependymoma group B (PF-EPN-B) tumours display frequent large-scale copy number gains and losses but have favourable clinical outcomes1,3. More than 70% of supratentorial ependymomas are defined by highly recurrent gene fusions in the NF-ÎșB subunit gene RELA (ST-EPN-RELA), and a smaller number involve fusion of the gene encoding the transcriptional activator YAP1 (ST-EPN-YAP1)1,3,4. Subependymomas, a distinct histologic variant, can also be found within the supratetorial and posterior fossa compartments, and account for the majority of tumours in the molecular subgroups ST-EPN-SE and PF-EPN-SE. Here we describe mapping of active chromatin landscapes in 42 primary ependymomas in two non-overlapping primary ependymoma cohorts, with the goal of identifying essential super-enhancer-associated genes on which tumour cells depend. Enhancer regions revealed putative oncogenes, molecular targets and pathways; inhibition of these targets with small molecule inhibitors or short hairpin RNA diminished the proliferation of patient-derived neurospheres and increased survival in mouse models of ependymomas.
