    The Clinical Signifcance of Expression of ERCC1 and PKCalpha in Non-small Cell Lung Cancer

    Background and objective Excision repair cross-complementing 1 (Excision-Repair Cross-Complementing 1, ERCC1), an important member of the DNA repair gene family, plays a key role in nucleotide excision repair and apoptosis of tumor cells. Protein kinase C-Ī± (Protein kinase C, PKCĪ±), an isozyme in protein kinase C family, is an important signaling molecule in signal transduction pathways of tumors, which has been implicated in malignant transformation and proliferation. The aim of this study was to explore the clinical significance of ERCC1 and PKCĪ± in non-small cell lung cancer (NSCLC). Methods The expression of ERCC1 and PKCĪ± were examined by immunohistochemistry (IHC) in the specimens of 51 cases of NSCLC patients tissue and 21 cases of paracancerous tissue. The relationship between detected data and patientsā€² clinical parameters was analyzed by SPSS 13.0 software. Results The positive expression rate of ERCC1 and PKCĪ± in NSCLC tissues was significantly higher than paracancerous tissues (Ī”<0.05). Expression of ERCC1 was closely related to clinical stage and N stage. The positive rate of ERCC1 was higher in III+IV or N1+N2 stage patients compared with I+II or N0 stage (Ī”=0.011, P=0.015). We also found that 5-year survival of negative group of ERCC1 was remarkably higher than that of positive group by Ļ‡2 test (Ī”<0.05). Expression of ERCC1 was positively correlative to PKCĪ± by Spearmanā€²s correlation analysis (r=0.425, P=0.002) in NSCLC. Conclusion The results suggest ERCC1 and PKCĪ± might be correlated with the development of NSCLC. ERCC1 might be related to prognosis of NSCLC. There might be existed a mechanism of coordination or regulation between ERCC1 and PKCĪ±

    Transarterial Embolization with Berenstein Liquid Coils and N-butyl Cyanoacrylate in a Vein of Galen Aneurysmal Malformation: a Case Report

    A 12-week-old baby with a vein of Galen aneurysmal malformation (VGAM) was successfully treated with performing transarterial microcatheter-directed embolization with Berenstein Liquid Coils and n-butyl cyanoacrylate in the feeding arteries. Post-procedure angiography showed a marked decrease of the blood flow into the dilated vein of Galen. Three months later, follow-up angiography showed that the vein of Galen aneurysmal malformation had totally disappeared, and the baby recovered very well without any sequelae. We report here on this interesting case along with a review of the relevant literature, and we aim to enhance physicians' awareness of the treatment for VGAMs

    Transmembrane TM3b of Mechanosensitive Channel MscS Interacts With Cytoplasmic Domain Cyto-Helix

    The mechanosensitive channel MscS functions as an osmolyte emergency release-valve in the event of a sudden decrease in external environmental osmolarity. MscS has served as a paradigm for studying how channel proteins detect and respond to mechanical stimuli. However, the inter-domain interactions and structural rearrangements that occur in the MscS gating process remain largely unknown. Here, we determined the interactions between the transmembrane domain and cytoplasmic domain of MscS. Using in vivo cellular viability, single-channel electrophysiological recording, and cysteine disulfide trapping, we demonstrated that N117 of the TM3b helix and N167 of the Cyto-helix are critical residues that function at the TM3b-Cyto helix interface. In vivo downshock assays showed that double cysteine substitution at N117 and N167 failed to rescue the osmotic-lysis phenotype of cells in acute osmotic downshock. Single-channel recordings demonstrated that cysteine cross-linking of N117C and N167C led to a non-conductive channel. Consistently, coordination of the histidines of N117H and N167H caused a decrease in channel gating. Moreover, cross-linked N117 and N167 altered the gating of the severe gain-of-function mutant L109S. Our results demonstrate that N117ā€“N167 interactions stabilize the inactivation state by an association of TM3b segments with Ī²-domains of the cytoplasmic region, providing further insights into the gating mechanism of the MscS channel

    VLBI Astrometry of Radio Stars to Link Radio and Optical Celestial Reference Frames: Observing Strategies

    The Gaia celestial reference frame (Gaia-CRF) will benefit from a close assessment with independent methods, such as Very Long Baseline Interferometry (VLBI) measurements of radio stars at bright magnitudes. However, obtaining full astrometric parameters for each radio star through VLBI measurements demands a significant amount of observation time. This study proposes an efficient observing strategy that acquires double-epoch VLBI positions to measure the positions and proper motions of radio stars at a reduced cost. The solution for CRF link compatible with individual VLBI position measurements is introduced, and the optimized observing epoch scheduling is discussed. Applying this solution to observational data yields results sensitive to sample increase or decrease, yet they remain consistently in line with the literature at the 1-sigma level. This suggests the potential for improvement with a larger sample size. Simulations for adding observations demonstrate the double-epoch strategy reduces CRF link parameter uncertainties by over 30% compared to the five-parameter strategy.Comment: 9 pages, 4 figures, accepted for publication in the Monthly Notices of the Royal Astronomy Society (MNRAS

    Effects of Different Thawing Methods on the Quality of Micropterus salmoides at Room Temperature

    In order to choose a suitable thawing method for Micropterus salmoides (room temperature thawing, running water thawing, saltwater thawing, ultrasonic static water thawing and ultrasonic running water thawing), this paper took Micropterus salmoides as the research subject. To investigate the effects of different thawing methods on water retention, total volatile base nitrogen (TVB-N), malondialdehyde content, peroxide value, total sulfhydryl content and Ca2+-ATPase activity of Micropterus salmoides. At the same time, the changes of texture properties such as hardness, elasticity and adhesion of fish were analyzed. The results showed that the normal temperature thaw 219 min, longest, fish water retention, lipid and protein oxidation was the most serious. Thawing water and salt water thawing time consuming 35 and 55 min, respectively, was the shorter of the normal temperature thaw, but fish water retention, protein oxidation, quality and structure characteristics of the various quality indexes such as the change was still serious. After ultrasonic static water thawing, the fish had better water retention and texture characteristics, and could effectively alleviate the oxidation of fish protein, but the lipid oxidation was more serious. Ultrasonic flow water thawing could complete the thawing of fish within 24 minutes, which was more efficient. Compared with other thawing methods, it could effectively maintain the water retention and texture characteristics of fish, and effectively delay the oxidation of fish protein and lipid. Ultrasonic thawing of frozen water, therefore, Micropterus salmoides had little effect on quality, was the most suitable thawing method
