    A knowledge-based weighting framework to boost the power of genome-wide association studies

    Background: We are moving to second-wave analysis of genome-wide association studies (GWAS), characterized by comprehensive bioinformatical and statistical evaluation of genetic associations. Existing biological knowledge is very valuable for GWAS, which may help improve their detection power particularly for disease susceptibility loci of moderate effect size. However, a challenging question is how to utilize available resources that are very heterogeneous to quantitatively evaluate the statistic significances. Methodology/Principal Findings: We present a novel knowledge-based weighting framework to boost power of the GWAS and insightfully strengthen their explorative performance for follow-up replication and deep sequencing. Built upon diverse integrated biological knowledge, this framework directly models both the prior functional information and the association significances emerging from GWAS to optimally highlight single nucleotide polymorphisms (SNPs) for subsequent replication. In the theoretical calculation and computer simulation, it shows great potential to achieve extra over 15% power to identify an association signal of moderate strength or to use hundreds of whole-genome subjects fewer to approach similar power. In a case study on late-onset Alzheimer disease (LOAD) for a proof of principle, it highlighted some genes, which showed positive association with LOAD in previous independent studies, and two important LOAD related pathways. These genes and pathways could be originally ignored due to involved SNPs only having moderate association significance. Conclusions/Significance: With user-friendly implementation in an open-source Java package, this powerful framework will provide an important complementary solution to identify more true susceptibility loci with modest or even small effect size in current GWAS for complex diseases. © 2010 Li et al.published_or_final_versio

    Specific neuroprotective effects of manual stimulation of real acupoints versus non-acupoints in rats after middle cerebral artery occlusion

    The objective of this study was to investigate the effectiveness and specific effects of acupuncture on ischemic-induced damage in rats after permanent middle cerebral artery occlusion. Cerebral ischemia was induced by middle cerebral artery occlusion in male Wistar rats. The rats were divided into the following 4 groups: normal controls, ischemic, real acupuncture-treated (Shuigou, DU26), and non-acupoint-treated groups. On the third postoperative day, neurological deficit scores, cerebral blood flow, infarction volume, and neuronal cell death counts were measured. In the real acupuncture-treated group, the neurological deficit scores and cerebral blood flow were improved (p < 0.05) and the infarction volume and neuronal cell death counts were reduced (p < 0.01) compared to the ischemic and non-acupoint-treated groups. The present study demonstrated that real acupuncture was effective against focal ischemia-induced damage in rats after middle cerebral artery occlusion, and the effects were specifically related to the right needling location.Key words: specificity, real acupoint, non-acupoint, middle cerebral artery occlusion, animal experimentatio

    Biosensor Based on Ultrasmall MoS2 Nanoparticles for Electrochemical Detection of H2O2 Released by Cells at the Nanomolar Level

    Monodispersed surfactant-free MoS2 nanoparticles with sizes of less than 2 nm were prepared from bulk MoS2 by simple ultrasonication and gradient centrifugation. The ultrasmall MoS2 nanoparticles expose a large fraction of edge sites, along with their high surface area, which lead to attractive electrocatalytic activity for reduction of H2O2. An extremely sensitive H2O2 biosensor based on MoS2 nanoparticles with a real determination limit as low as 2.5 nM and wide linear range of 5 orders of magnitude was constructed. On the basis of this biosensor, the trace amount of H2O2 released from Raw 264.7 cells was successfully recorded, and an efficient glucose biosensor was also fabricated.     Postnatal expression of myostain (MSTN) and myogenin (MYoG) genes in Hu sheep of China

    The study of candidate genes is an important tool to identify genes associated with economic traits. Skeletal muscle development is an important physiological process in meat animals, and it directly affects meat production. The expression of myostain (MSTN) and myogenin (MYoG) genes in longissimus dorsi, during the early growth stage of Hu sheep, was studied by semi-quantitative Reverse transcription polymerase chain reaction (RT-PCR). The results demonstrate that age and gender were playing a very important role in the expression of sheep muscle. MSTN and MYOG genes showed similar variation pattern for the male and female. The expression level of the MSTN and MYoG genes all showed a positive correlation with live weight, carcass weight and meat percentage, but only showed a significant relationship with meat percentage.     Expression of a diverse array of Ca<sup>2+</sup>-activated K<sup>+</sup> channels (SK1/3, IK1, BK) that functionally couple to the mechanosensitive TRPV4 channel in the collecting duct system of kidney

    The voltage- and Ca2+-activated, large conductance K+ channel (BK, maxi-K) is expressed in the collecting duct system of kidney where it underlies flow- and Ca2+-dependent K+ excretion. To determine if other Ca2+-activated K+ channels (KCa) may participate in this process, mouse kidney and the K+-secreting mouse cortical collecting duct (CCD) cell line, mCCDcl1, were assessed for TRPV4 and KCa channel expression and cross-talk. qPCR mRNA analysis and immunocytochemical staining demonstrated TRPV4 and KCa expression in mCCDcl1 cells and kidney connecting tubule (CNT) and CCD. Three subfamilies of KCa channels were revealed: the high Ca2+-binding affinity small-conductance SK channels, SK1and SK3, the intermediate conductance channel, IK1, and the low Ca2+-binding affinity, BK channel (BKα subunit). Apparent expression levels varied in CNT/CCD where analysis of CCD principal cells (PC) and intercalated cells (IC) demonstrated differential staining: SK1:PCIC, IK1:PC>IC, BKα:PC = IC, and TRPV4:PC>IC. Patch clamp analysis and fluorescence Ca2+ imaging of mCCDcl1 cells demonstrated potent TRPV4-mediated Ca2+ entry and strong functional cross-talk between TRPV4 and KCa channels. TRPV4-mediated Ca2+ influx activated each KCa channel, as evidenced by selective inhibition of KCa channels, with each active KCa channel enhancing Ca2+ entry (due to membrane hyperpolarization). Transepithelial electrical resistance (TEER) analysis of confluent mCCDcl1 cells grown on permeable supports further demonstrated this cross-talk where TRPV4 activation induce a decrease in TEER which was partially restored upon selective inhibition of each KCa channel. It is concluded that SK1/SK3 and IK1 are highly expressed along with BKα in CNT and CCD and are closely coupled to TRPV4 activation as observed in mCCDcl1 cells. The data support a model in CNT/CCD segments where strong cross talk between TRPV4-mediated Ca2+ influx and each KCa channel leads to enhance Ca2+ entry which will support activation of the low Ca2+-binding affinity BK channel to promote BK-mediated K+ secretion

    Impacts of wind forcing on sea level variations in the East China Sea: Local and remote effects

    The regional sea level variation in the East China Sea (ECS) was influenced not only by local factors but also by remote wind from adjoining ocean with the oceanic connectivity influenced by upper-ocean circulation. The satellite altimeter observations showed that from 1993 to 2008, the inter-annual sea level variation in the ECS was negatively related to the strength of Kuroshio. To investigate the relative role of local and remote wind, two sensitive experiments were carried out using the POP model. Model experiments revealed that wind-induced redistributions of water played a significant role in the sea level variation of the ECS. The seasonal variations were induced by both local winds and remote Pacific wind stress with approximately equal contribution. However, on the inter-annual sea level variations, the remote wind forcing over the North Pacific could contribute substantially more than that of local wind which modulated sea level immediately. Remote wind influenced the China Sea in forms of changing of wind stress curl and ocean currents, which influenced the intensity of the Kuroshio, especially during El Nino episodes.     The increasing growth of the seafood processing industries considerably requires more industrial process activities and water consumption. It is estimated that approximately 10–40 m3 of wastewater is generated from those industries for processing one-tonne of raw materials. Due to limitations and regulations in natural resources utilization, a suitable and systematic wastewater treatment plant is very important to meet rigorous discharge standards. As a result of food waste biodegradability, the biological treatment and some extent of swim-bed technology, including a novel acryl-fibre (biofilm) material might be used effectively to meet the effluent discharge criteria.
