A Pan1/End3/Sla1 complex links Arp2/3-mediated actin assembly to sites of clathrin-mediated endocytosis.
More than 60 highly conserved proteins appear sequentially at sites of clathrin-mediated endocytosis in yeast and mammals. The yeast Eps15-related proteins Pan1 and End3 and the CIN85-related protein Sla1 are known to interact with each other in vitro, and they all appear after endocytic-site initiation but before endocytic actin assembly, which facilitates membrane invagination/scission. Here we used live-cell imaging in parallel with genetics and biochemistry to explore comprehensively the dynamic interactions and functions of Pan1, End3, and Sla1. Our results indicate that Pan1 and End3 associate in a stable manner and appear at endocytic sites before Sla1. The End3 C-terminus is necessary and sufficient for its cortical localization via interaction with Pan1, whereas the End3 N-terminus plays a crucial role in Sla1 recruitment. We systematically examined the dynamic behaviors of endocytic proteins in cells in which Pan1 and End3 were simultaneously eliminated, using the auxin-inducible degron system. The results lead us to propose that endocytic-site initiation and actin assembly are separable processes linked by a Pan1/End3/Sla1 complex. Finally, our study provides mechanistic insights into how Pan1 and End3 function with Sla1 to coordinate cargo capture with actin assembly
Automatic Classification and Speaker Identification of African Elephant (\u3cem\u3eLoxodonta africana\u3c/em\u3e) Vocalizations
A hidden Markov model (HMM) system is presented for automatically classifying African elephant vocalizations. The development of the system is motivated by successful models from human speech analysis and recognition. Classification features include frequency-shifted Mel-frequency cepstral coefficients (MFCCs) and log energy, spectrally motivated features which are commonly used in human speech processing. Experiments, including vocalization type classification and speaker identification, are performed on vocalizations collected from captive elephants in a naturalistic environment. The system classified vocalizations with accuracies of 94.3% and 82.5% for type classification and speaker identification classification experiments, respectively. Classification accuracy, statistical significance tests on the model parameters, and qualitative analysis support the effectiveness and robustness of this approach for vocalization analysis in nonhuman species
The Thermogravimetric Behaviour of Metal Diethyldithiocarbamates at reduced pressure
Snapshot Samplings of the Bitcoin Transaction Network and Analysis of Cryptocurrency Growth
The purpose of this work was to perform a network analysis on the rapidly
growing bitcoin transaction network. Using a web-socket API, we collected data
on all transactions occurring during a six hour window. Sender and receiver
addresses as well as the amount of bitcoin exchanged were record. Graphs were
generated, using R and Gephi, in which nodes represent addresses and edges
represent the exchange of bitcoin. The six hour data set was subsetted into a
one and two hour sampling snapshot of the network. We performed comparisons and
analysis on all subsets of the data in an effort to determine the minimum
sampling length that represented the network as a whole. Our results suggest
that the six hour sampling was the minimum limit with respect to sampling time
needed to accurately characterize the bitcoin transaction network.Anonymity is
a desired feature of the blockchain and bitcoin network however, it limited us
in our analysis and conclusions we drew from our results were mostly inferred.
Future work is needed and being done to gather more comprehensive data so that
the bitcoin transaction network can be better analyzed.Comment: 8 pages, 8 figures, 2 table
Simulating spin-charge separation with light
In this work we show that stationary light-matter excitations generated
inside a hollow one-dimensional waveguide filled with atoms, can be made to
generate a photonic two-component Lieb Liniger model. We explain how to prepare
and drive the atomic system to a strongly interacting regime where spin-charge
separation could be possible. We then proceed by explaining how to measure the
corresponding effective spin and charge densities and velocities through
standard optical methods based in measuring dynamically the emitted photon
intensities or by analyzing the photon spectrum. The relevant interactions
exhibit the necessary tunability both to generate and efficiently observe spin
charge separation with current technology.Comment: 4 pages. Comments welcom
Voicing values: Laying foundations for ageing people to participate in design
© 2016 ACM. This paper discusses Participatory Design workshops that sought to enable ageing people to articulate their core values in relation to their experiences of ageing. Our motivations were to better understand how ageing people decide whether or not to adopt and use particular technologies, and to gain insights into the kinds of technologies that might support their aspirations as they age. We contribute to current understandings of ageing people's values, including a range of values that were most important to our participants, insights into how these values are expressed and experienced in everyday lives, the interrelatedness of values in action, and how the three social dimensions of self, friends and family, as well as community influence the expression of values. The workshops demonstrated how engaged ageing people are with others and the broader communities they inhabit. We reflect on the processes, methods and tools that were useful when supporting people to voice their values and how this approach can support the participation of ageing people in design
Development and validation of the crew-station system-integration research facility
The various issues associated with the use of integrated flight management systems in aircraft were discussed. To address these issues a fixed base integrated flight research (IFR) simulation of a helicopter was developed to support experiments that contribute to the understanding of design criteria for rotorcraft cockpits incorporating advanced integrated flight management systems. A validation experiment was conducted that demonstrates the main features of the facility and the capability to conduct crew/system integration research
