    Pengembangan Modul Matematika Materi Aritmatika Sosial Berbasis Pendekatan Saintifik Untuk Siswa Kelas VII SMP

    Penelitian ini bertujuan untuk menghasilkan modul matematika dengan pendekatan saintifik pada materi Aritmatika Sosial di kelas VII SMP Negeri I Bajawa Utara yang memenuhi kriteria valid dan praktis. Jenis penelitian ini adalah penelitian yang mengacu pada model pengembangan ADDIE yaitu analysis, design, development, implementation, dan evaluation. Validasi modul dilakukan oleh dua orang ahli pendidikan bidang Matematika. Kepraktisan modul diuji pada lima peserta didik kelas VII SMP Negeri I semester genap tahun ajaran 2019/2020. Instrumen penelitian ini adalah lembar validasi modul, lembar kepraktisaan modul, semua data yang dikumpulkan dianalisis kevalidan dan kepraktisan. Hasil penelitian menunjukan bahwa, modul matematika dengan pendekatan saintifik pada materi aritmatika sosial di kelas VII SMP Negeri I Bajawa Utara termaksud dalam kategori valid dari aspek materi dan desain dengan skor rata-rata 4,15,  modul matematika dengan pendekatan saintifik pada materi aritmatika sosial di kelas VII SMP Negeri I Bajawa Utara termasuk dalam kategori sangat praktis dengan skor rata-rata 4,7. Modul sudah memuat langkah-langkah pendekatan saintifik yang membuat aktivitas guru dan peserta didik  berkriteria baik, modul sudah membuat peserta didik melaksanakan proses pembelajaran sesuai dengan kaidah pendekatan saintifik sehingga peserta didik memperoleh hasil belajar yang tinggi

MSMEs are a sector consisting of small and medium scale businesses, both in terms of production, turnover and number of employees. They are often the backbone of the local economy, playing a role in creating jobs, supporting economic growth, and strengthening a country's economic resilience. The method used in this research is digital marketing assistance to increase turnover in Sleman Regency MSMEs.     Desa Kotagana merupakan sebuah desa yang ada di Kecamatan Mauponggo, Kabupaten Nagekeo, Provinsi Nusa Tenggara Timur Indonesia. Kegiatan Kuliah Kerja Nyata (KKN) yang dilaksanakan mulai 5 Desember 2022 sampai 5 Januari 2023, Berdasarkan data yang diperoleh Desa Kotagana memiliki kondisi, potensi, dan permasalahan yang berbeda-beda seperti adanya permasalahan yang dirasakan siswa SD dan TK setelah dampak Covid-19, sehingga  siswa-siswi kembali melakukan proses belajar mengajar di sekolah secara aktif dan juga mendapat pendampingan belajar khusus di rumah dari mahasiswa KKN STKIP Citra Bakti. Kuliah Kerja Nyata (KKN) merupakan sebagai program pengabdian kepada masyarakat dilaksanakan sebagai salah satu perwujudan tridharma perguruan tinggi. KKN adalah media yang digunakan untuk mentransfer IPTEKS yang dikaji dan dikembangkan di Pusat Penelitian dan Pengabdian Kepada Masyarakat STKIP Citra Bakti. Berdasarkan hal tersebut mahasiswa KKN STKIP Citra Bakti Ngada yang tidak terlepas dari fungsinya sebagai mahasiswa pendidikan, diharapkan mampu menyalurkan ilmu atau pengetahuan yang diperoleh dari bangku perkuliahan kepada orang lain. Teknik pengumpulan data yang digunakan dalam artikel ini ialah pendekatan partisipatif (Participatory Approach), untuk memberikan gambaran tentang kegiatan pendampingan belajar dalam meningkatkan minat belajar siswa sekolah dasar dari tingkat TK sampai SD di Desa Kotagana Kecamatan Mauponggo Kabupaten Nagekeo.     The evolutionary history of the stearoyl-CoA desaturase gene family in vertebrates

    <p/> <p>Background</p> <p>Stearoyl-CoA desaturases (SCDs) are key enzymes involved in <it>de novo </it>monounsaturated fatty acid synthesis. They catalyze the desaturation of saturated fatty acyl-CoA substrates at the delta-9 position, generating essential components of phospholipids, triglycerides, cholesterol esters and wax esters. Despite being crucial for interpreting SCDs roles across species, the evolutionary history of the SCD gene family in vertebrates has yet to be elucidated, in particular their isoform diversity, origin and function. This work aims to contribute to this fundamental effort.</p> <p>Results</p> <p>We show here, through comparative genomics and phylogenetics that the SCD gene family underwent an unexpectedly complex history of duplication and loss events. Paralogy analysis hints that SCD1 and SCD5 genes emerged as part of the whole genome duplications (2R) that occurred at the stem of the vertebrate lineage. The SCD1 gene family expanded in rodents with the parallel loss of SCD5 in the Muridae family. The SCD1 gene expansion is also observed in the Lagomorpha although without the SCD5 loss. In the amphibian <it>Xenopus tropicalis </it>we find a single SCD1 gene but not SCD5, though this could be due to genome incompleteness. In the analysed teleost species no SCD5 is found, while the surrounding SCD5-less locus is conserved in comparison to tetrapods. In addition, the teleost SCD1 gene repertoire expanded to two copies as a result of the teleost specific genome duplication (3R). Finally, we describe clear orthologues of SCD1 and SCD5 in the chondrichthian, <it>Scyliorhinus canicula</it>, a representative of the oldest extant jawed vertebrate clade. Expression analysis in <it>S. canicula </it>shows that whilst SCD1 is ubiquitous, SCD5 is mainly expressed in the brain, a pattern which might indicate an evolutionary conserved function.</p> <p>Conclusion</p> <p>We conclude that the SCD1 and SCD5 genes emerged as part of the 2R genome duplications. We propose that the evolutionary conserved gene expression between distinct lineages underpins the importance of SCD activity in the brain (and probably the pancreas), in a yet to be defined role. We argue that an expression independent of an external stimulus, such as diet induced activity, emerged as a novel function in vertebrate ancestry allocated to the SCD5 isoform in various tissues (e.g. brain and pancreas), and it was selectively maintained throughout vertebrate evolution.</p

    Effect of dietary fatty acids on expression of lipogenic enzymes and fatty acid profile in tissues of bulls

    This study investigated the effects of dietary linolenic acid (C18:3n-3) v. linoleic acid (C18:2n-6) on fatty acid composition and protein expression of key lipogenic enzymes, acetyl-CoA carboxylase (ACC), stearoyl-CoA desaturase (SCD) and delta 6 desaturase (6d) in longissimus muscle and subcutaneous adipose tissue of bulls. Supplementation of the diet with C18:3n-3 was accompanied by an increased level of n-3 fatty acids in muscle which resulted in decrease of n-6/n-3 ratio. The diet enriched with n-3 polyunsaturated fatty acids (PUFAs) significantly inhibited SCD protein expression in muscle and subcutaneous adipose tissue, and reduced the 6d expression in muscle. There was no significant effect of the diet on ACC protein expression. Inhibition of the 6d expression was associated with a decrease in n-6 PUFA level in muscles, whereas repression of SCD protein was related to a lower oleic acid (C18:1 cis-9) content in the adipose tissue. Expression of ACC, SCD and 6d proteins was found to be relatively higher in subcutaneous adipose tissue when compared with longissimus muscle. It is suggested that dietary manipulation of fatty acid composition in ruminants is mediated, at least partially, through the regulation of lipogenic enzymes expression and that regulation of the bovine lipogenic enzymes expression is tissue specific. © 2010 The Animal Consortium

    Elevated Stearoyl-CoA Desaturase in Brains of Patients with Alzheimer's Disease

    The molecular bases of Alzheimer's disease (AD) remain unclear. We used a lipidomic approach to identify lipid abnormalities in the brains of subjects with AD (N = 37) compared to age-matched controls (N = 17). The analyses revealed statistically detectable elevations in levels of non-esterified monounsaturated fatty acids (MUFAs) and mead acid (20:3n-9) in mid-frontal cortex, temporal cortex and hippocampus of AD patients. Further studies showed that brain mRNAs encoding for isoforms of the rate-limiting enzyme in MUFAs biosynthesis, stearoyl-CoA desaturase (SCD-1, SCD-5a and SCD-5b), were elevated in subjects with AD. The monounsaturated/saturated fatty acid ratio (‘desaturation index’) – displayed a strong negative correlation with measures of cognition: the Mini Mental State Examination test (r = −0.80; P = 0.0001) and the Boston Naming test (r = −0.57; P = 0.0071). Our results reveal a previously unrecognized role for the lipogenic enzyme SCD in AD

    StearoylCoA Desaturase-5: A Novel Regulator of Neuronal Cell Proliferation and Differentiation

    Recent studies have demonstrated that human stearoylCoA desaturase-1 (SCD1), a Δ9-desaturase that converts saturated fatty acids (SFA) into monounsaturated fatty acids, controls the rate of lipogenesis, cell proliferation and tumorigenic capacity in cancer cells. However, the biological function of stearoylCoA desaturase-5 (SCD5), a second isoform of human SCD that is highly expressed in brain, as well as its potential role in human disease, remains unknown. In this study we report that the constitutive overexpression of human SCD5 in mouse Neuro2a cells, a widely used cell model of neuronal growth and differentiation, displayed a greater n-7 MUFA-to-SFA ratio in cell lipids compared to empty-vector transfected cells (controls). De novo synthesis of phosphatidylcholine and cholesterolesters was increased whereas phosphatidylethanolamine and triacylglycerol formation was reduced in SCD5-expressing cells with respect to their controls, suggesting a differential use of SCD5 products for lipogenic reactions. We also observed that SCD5 expression markedly accelerated the rate of cell proliferation and suppressed the induction of neurite outgrowth, a typical marker of neuronal differentiation, by retinoic acid indicating that the desaturase plays a key role in the mechanisms of cell division and differentiation. Critical signal transduction pathways that are known to modulate these processes, such epidermal growth factor receptor (EGFR)Akt/ERK and Wnt, were affected by SCD5 expression. Epidermal growth factor-induced phosphorylation of EGFR, Akt and ERK was markedly blunted in SCD5-expressing cells. Furthermore, the activity of canonical Wnt was reduced whereas the non-canonical Wnt was increased by the presence of SCD5 activity. Finally, SCD5 expression increased the secretion of recombinant Wnt5a, a non-canonical Wnt, whereas it reduced the cellular and secreted levels of canonical Wnt7b. Our data suggest that, by a coordinated modulation of key lipogenic pathways and transduction signaling cascades, SCD5 participates in the regulation of neuronal cell growth and differentiation
