    Novel insights into the genomic basis of citrus canker based on the genome sequences of two strains of Xanthomonas fuscans subsp. aurantifolii

    Background: Citrus canker is a disease that has severe economic impact on the citrus industry worldwide. There are three types of canker, called A, B, and C. The three types have different phenotypes and affect different citrus species. The causative agent for type A is Xanthomonas citri subsp. citri, whose genome sequence was made available in 2002. Xanthomonas fuscans subsp. aurantifolii strain B causes canker B and Xanthomonas fuscans subsp. aurantifolii strain C causes canker C. Results: We have sequenced the genomes of strains B and C to draft status. We have compared their genomic content to X. citri subsp. citri and to other Xanthomonas genomes, with special emphasis on type III secreted effector repertoires. In addition to pthA, already known to be present in all three citrus canker strains, two additional effector genes, xopE3 and xopAI, are also present in all three strains and are both located on the same putative genomic island. These two effector genes, along with one other effector-like gene in the same region, are thus good candidates for being pathogenicity factors on citrus. Numerous gene content differences also exist between the three cankers strains, which can be correlated with their different virulence and host range. Particular attention was placed on the analysis of genes involved in biofilm formation and quorum sensing, type IV secretion, flagellum synthesis and motility, lipopolysacharide synthesis, and on the gene xacPNP, which codes for a natriuretic protein. Conclusion: We have uncovered numerous commonalities and differences in gene content between the genomes of the pathogenic agents causing citrus canker A, B, and C and other Xanthomonas genomes. Molecular genetics can now be employed to determine the role of these genes in plant-microbe interactions.     Genetic diversity of Xanthomonas axonopodis pv. citri based on plasmid profile and pulsed field gel electrophoresis

    Xanthomonas axonopodis pv. citri strains that cause disease in citrus were investigated by pulsed field and plasmid profile analysis. For the first method, genomic DNA was digested by the rare-cutting enzymes Xba I and Vsp I. The strains evaluated were collected in seven different States of Brazil and in Argentina, Bolivia, Paraguay and Uruguay. Genetic variability was found among strains of X. axonopodis pv. citri from different geographical areas Argentina, Bolivia and Uruguay, with similarities varying from 0.62 to 0.83. However, the strains collected in Brazil, despite being from different States, have shown a genetic similarity ranging from 0.83 to 1.00. Cluster analysis showed a relationship between genomic similarity and geographical origin of the strains. Plasmids were observed in all strains, with a total of five different plasmids, with sizes between 57.7 and 83.0 kilobases. The 72.6 kb plasmid was the most frequent, present in 15 out of 22 strains, while the 68.1 kb plasmid was observed in two strains only. Although the plasmid diversity detected in the present study was not very great, the X. axonopodis pv. citri strains evaluated showed a considerable degree of diversity with regard to this extrachromosomal genetic element

    Performance of 'Okitsu' Satsuma Mandarin on nine rootstocks

    Mandarins have become increasingly valued as citrus fruits for the fresh market due to the easy peeling, attractive flavor, and health and nutritional properties. Plant growth and yield, and characteristics of fruits of 'Okitsu' Satsuma mandarin (Citrus unshiu Marc.) trees grafted on nine rootstocks were evaluated in Londrina, northern Paraná, Brazil. The rootstocks were: 'Rangpur' lime (Citrus limonia Osb.); 'Cleopatra' (Citrus reshni hort. ex Tanaka) and 'Sunki' mandarins (Citrus sunki hort. ex Tanaka); 'C-13' [Citrus sinensis × Poncirus trifoliata (L.) Raf.] and 'Carrizo' citranges [C. sinensis × P. trifoliata (L.) Raf.]; 'Volkamer' lemon (Citrus volkameriana V. Ten. & Pasq.); trifoliate orange [P. trifoliata (L.) Raf.; 'Caipira DAC' sweet orange [C. sinensis (L.) Osb.] and 'Swingle' citrumelo [Citrus paradisi Macfad. cv. Duncan × P. trifoliata (L.) Raf.]. The highest plant growth was for the trees on 'Cleopatra' mandarin and 'Caipira DAC' sweet orange. In contrast, the smallest size was for the trees on 'Volkamer' lemon and trifoliate orange. The largest difference between the trunk diameter below and above the grafting point was induced by 'Swingle' citrumelo. Trees of 'Okitsu' Satsuma mandarin on 'Swingle' citrumelo presented the highest yield, while 'C-13', 'Carrizo', 'Sunki', and 'Swingle' induced the largest fruit masses. With regard to fruit characteristics, 'Carrizo' and trifoliate orange induced the best ratio and juice content.     aBstraCt -'Pêra' sweet orange is the most important citrus cultivar in Brazil, but it is highly susceptible to Citrus tristeza virus (CTV), the causal agent of citrus tristeza disease. In this study, we evaluated the severity of citrus tristeza disease and its relation with the agronomic performance of seven cultivars and six selections of 'Pêra' sweet orange under field conditions in the North and Northwest regions of the State of Paraná, Brazil. The citrus tristeza disease severity was determined based on symptoms of stem pitting. The vegetative growth assessed in trees of all four locations for the 2012 season, and in two field plots for the 2011, 2012 and 2013 harvests, respectively. The cultivar 'IPR 153' (Bianchi) had the best performance concerning stem pitting severity, while Selection 27 was among the ones with the highest rate of vegetative growth. Further, Selections 27 and 12, and the cultivar 'Vimusa' had the largest fruit yields. Based on correlation analysis, stem pitting symptoms severity was negatively correlated with vegetative growth and fruit yield.     O objetivo do trabalho foi a caracterização de acessos de laranja-doce [Citrus sinensis (L.) Osbeck] do Banco Ativo de Germoplasma de Citros (BAG Citros) do IAPAR, em Londrina-PR. Foram estudados os acessos I-02 'Piralima'; I-03 'Barão'; I-11 'Natal', I-16 'Hamlin', I-17 'Seleta-Vermelha', I-60 'Natal', enxertados sobre limão- 'Cravo' (Citrus limonia Osbeck). As plantas foram conduzidas em espaçamento de 7,0 m x 7,0 m e sem irrigação. Os dados de produção (de 1983 a 1997) e as características físico-químicas dos frutos (de 1984 a 2000), como massa (MF), sólidos solúveis totais (SST), acidez titulável total (ATT), ratio (SST/ATT), rendimento em suco (Suco) e índice tecnológico (IT) foram submetidos à análise de variância, complementada pelo teste de Tukey, a 5% de probabilidade. O acesso I-16 'Hamlin' foi o mais produtivo (218,1 kg por planta) e diferiu-se dos demais. Para a massa do fruto, destacaram-se os acessos I-17 'Seleta-Vermelha' (208,8 g) e I-11 'Natal' (142,0 g), sem diferença entre si. Com relação às características químicas dos frutos, os acessos apresentaram resultados semelhantes, dentro dos padrões considerados adequados para as cultivares, exceto o acesso I-17, 'Seleta- Vermelha', que teve índice tecnológico menor que 2,0 kg.caixa-1 de SST.<br>The aim of this work was to characterize sweet orange [Citrus sinensis (L.) Osbeck] accessions from the IAPAR Active Citrus Germplasm Bank (AGB Citrus) in Londrina, Paraná, Brazil. The accessions studied were: I-02 'Piralima', I-03 'Barão', I-11 'Natal', I-16 'Hamlin', I-17 'Seleta Vermelha', I-60 'Natal', I-67 budded on Rangpur lime (Citrus limonia Osbeck). The plants had 7.0 m x 7.0 m spacing, and were carried out without irrigation. Data of yield (from 1983 to 1997) and fruit physical-chemical characteristics (from 1984 to 2000), like fruit mass (FM), total soluble solids (TSS), total titratable acidity (TTA), ratio (TSS/TTA), juice content and technological index (TI) were submitted to ANOVA and the means were compared by the Tukey´s test (5%). I-16 'Hamlin' accession had the highest yield (218.1 kg per plant), and it was different from the others. Accessions I-17 'Seleta Vermelha' and I-11 'Natal' had the highest fruit mass (209 and 142 g, respectively), with no difference between them. The accessions had similar results regarding fruit chemical characteristics, and had acceptable values regarding quality standards, except I-17 'Seleta Vermelha', which had TI lower than 2.0 kg.box-1 of SST
