Building the cosmic distance scale: from Hipparcos to Gaia
Hipparcos, the first ever experiment of global astrometry, was launched by
ESA in 1989 and its results published in 1997 (Perryman et al., Astron.
Astrophys. 323, L49, 1997; Perryman & ESA (eds), The Hipparcos and Tycho
catalogues, ESA SP-1200, 1997). A new reduction was later performed using an
improved satellite attitude reconstruction leading to an improved accuracy for
stars brighter than 9th magnitude (van Leeuwen & Fantino, Astron. Astrophys.
439, 791, 2005; van Leeuwen, Astron. Astrophys. 474, 653, 2007).
The Hipparcos Catalogue provided an extended dataset of very accurate
astrometric data (positions, trigonometric parallaxes and proper motions),
enlarging by two orders of magnitude the quantity and quality of distance
determinations and luminosity calibrations. The availability of more than 20000
stars with a trigonometric parallax known to better than 10% opened the way to
a drastic revision of our 3-D knowledge of the solar neighbourhood and to a
renewal of the calibration of many distance indicators and age estimations. The
prospects opened by Gaia, the next ESA cornerstone, planned for launch in June
2013 (Perryman et al., Astron. Astrophys. 369, 339, 2001), are still much more
dramatic: a billion objects with systematic and quasi simultaneous astrometric,
spectrophotometric and spectroscopic observations, about 150 million stars with
expected distances to better than 10%, all over the Galaxy. All stellar
distance indicators, in very large numbers, will be directly measured,
providing a direct calibration of their luminosity and making possible detailed
studies of the impacts of various effects linked to chemical element
abundances, age or cluster membership. With the help of simulations of the data
expected from Gaia, obtained from the mission simulator developed by DPAC, we

Differential roles of the Drosophila EMT-inducing transcription factors Snail and Serpent in driving primary tumour growth.
Several transcription factors have been identified that activate an epithelial-to-mesenchymal transition (EMT), which endows cells with the capacity to break through basement membranes and migrate away from their site of origin. A key program in development, in recent years it has been shown to be a crucial driver of tumour invasion and metastasis. However, several of these EMT-inducing transcription factors are often expressed long before the initiation of the invasion-metastasis cascade as well as in non-invasive tumours. Increasing evidence suggests that they may promote primary tumour growth, but their precise role in this process remains to be elucidated. To investigate this issue we have focused our studies on two Drosophila transcription factors, the classic EMT inducer Snail and the Drosophila orthologue of hGATAs4/6, Serpent, which drives an alternative mechanism of EMT; both Snail and GATA are specifically expressed in a number of human cancers, particularly at the invasive front and in metastasis. Thus, we recreated conditions of Snail and of Serpent high expression in the fly imaginal wing disc and analysed their effect. While either Snail or Serpent induced a profound loss of epithelial polarity and tissue organisation, Serpent but not Snail also induced an increase in the size of wing discs. Furthermore, the Serpent-induced tumour-like tissues were able to grow extensively when transplanted into the abdomen of adult hosts. We found the differences between Snail and Serpent to correlate with the genetic program they elicit; while activation of either results in an increase in the expression of Yorki target genes, Serpent additionally activates the Ras signalling pathway. These results provide insight into how transcription factors that induce EMT can also promote primary tumour growth, and how in some cases such as GATA factors a ‘multi hit’ effect may be achieved through the aberrant activation of just a single gene
ECMO for COVID-19 patients in Europe and Israel
Since March 15th, 2020, 177 centres from Europe and Israel have joined the study, routinely reporting on the ECMO support they provide to COVID-19 patients. The mean annual number of cases treated with ECMO in the participating centres before the pandemic (2019) was 55. The number of COVID-19 patients has increased rapidly each week reaching 1531 treated patients as of September 14th. The greatest number of cases has been reported from France (n = 385), UK (n = 193), Germany (n = 176), Spain (n = 166), and Italy (n = 136) .The mean age of treated patients was 52.6 years (range 16–80), 79% were male. The ECMO configuration used was VV in 91% of cases, VA in 5% and other in 4%. The mean PaO2 before ECMO implantation was 65 mmHg. The mean duration of ECMO support thus far has been 18 days and the mean ICU length of stay of these patients was 33 days. As of the 14th September, overall 841 patients have been weaned from ECMO
support, 601 died during ECMO support, 71 died after withdrawal of ECMO, 79 are still receiving ECMO support and for 10 patients status n.a. . Our preliminary data suggest that patients placed
on ECMO with severe refractory respiratory or cardiac failure secondary to COVID-19 have a reasonable (55%) chance of survival. Further extensive data analysis is expected to provide invaluable information on the demographics, severity of illness, indications and different ECMO management strategies in these patients
Extracorporeal Membrane Oxygenation for Severe Acute Respiratory Distress Syndrome associated with COVID-19: An Emulated Target Trial Analysis.
RATIONALE: Whether COVID patients may benefit from extracorporeal membrane oxygenation (ECMO) compared with conventional invasive mechanical ventilation (IMV) remains unknown. OBJECTIVES: To estimate the effect of ECMO on 90-Day mortality vs IMV only Methods: Among 4,244 critically ill adult patients with COVID-19 included in a multicenter cohort study, we emulated a target trial comparing the treatment strategies of initiating ECMO vs. no ECMO within 7 days of IMV in patients with severe acute respiratory distress syndrome (PaO2/FiO2 <80 or PaCO2 ≥60 mmHg). We controlled for confounding using a multivariable Cox model based on predefined variables. MAIN RESULTS: 1,235 patients met the full eligibility criteria for the emulated trial, among whom 164 patients initiated ECMO. The ECMO strategy had a higher survival probability at Day-7 from the onset of eligibility criteria (87% vs 83%, risk difference: 4%, 95% CI 0;9%) which decreased during follow-up (survival at Day-90: 63% vs 65%, risk difference: -2%, 95% CI -10;5%). However, ECMO was associated with higher survival when performed in high-volume ECMO centers or in regions where a specific ECMO network organization was set up to handle high demand, and when initiated within the first 4 days of MV and in profoundly hypoxemic patients. CONCLUSIONS: In an emulated trial based on a nationwide COVID-19 cohort, we found differential survival over time of an ECMO compared with a no-ECMO strategy. However, ECMO was consistently associated with better outcomes when performed in high-volume centers and in regions with ECMO capacities specifically organized to handle high demand. Socializing One Health: an innovative strategy to investigate social and behavioral risks of emerging viral threats
In an effort to strengthen global capacity to prevent, detect, and control infectious diseases in animals and people, the United States Agency for International Development’s (USAID) Emerging Pandemic Threats (EPT) PREDICT project funded development of regional, national, and local One Health capacities for early disease detection, rapid response, disease control, and risk reduction. From the outset, the EPT approach was inclusive of social science research methods designed to understand the contexts and behaviors of communities living and working at human-animal-environment interfaces considered high-risk for virus emergence. Using qualitative and quantitative approaches, PREDICT behavioral research aimed to identify and assess a range of socio-cultural behaviors that could be influential in zoonotic disease emergence, amplification, and transmission. This broad approach to behavioral risk characterization enabled us to identify and characterize human activities that could be linked to the transmission dynamics of new and emerging viruses. This paper provides a discussion of implementation of a social science approach within a zoonotic surveillance framework. We conducted in-depth ethnographic interviews and focus groups to better understand the individual- and community-level knowledge, attitudes, and practices that potentially put participants at risk for zoonotic disease transmission from the animals they live and work with, across 6 interface domains. When we asked highly-exposed individuals (ie. bushmeat hunters, wildlife or guano farmers) about the risk they perceived in their occupational activities, most did not perceive it to be risky, whether because it was normalized by years (or generations) of doing such an activity, or due to lack of information about potential risks. Integrating the social sciences allows investigations of the specific human activities that are hypothesized to drive disease emergence, amplification, and transmission, in order to better substantiate behavioral disease drivers, along with the social dimensions of infection and transmission dynamics. Understanding these dynamics is critical to achieving health security--the protection from threats to health-- which requires investments in both collective and individual health security. Involving behavioral sciences into zoonotic disease surveillance allowed us to push toward fuller community integration and engagement and toward dialogue and implementation of recommendations for disease prevention and improved health security
Synthèse totale de produits naturels actifs sur le SNC (nouvelles stratégies en série tétrahydropyridinique et application en synthèse totale)
Ce mémoire présente notre contribution à la synthèse totale de molécules actives sur le système nerveux central ainsi que les quelques outils méthodologiques qui nous ont été nécessaires. La première partie décrit les différentes synthèses totales réalisées. Dans un premier temps nous avons concentré nos efforts sur la synthèse de quelques alcaloïdes mineurs du tabac isolés de Nicotiana tabacum tels que l'anabasine, l'anatabine et leurs dérivés N-méthylés. Le savoir-faire acquis a été mis en application pour la première synthèse énantiosélective du SIB-1508Y, composé en phase II d'étude clinique pour le traitement de la maladie de Parkinson. Nous nous sommes également intéressés à la synthèse totale de la lobeline et de quelques-uns de ses analogues structuraux isolés de Lobelia inflata tels que la sédamine et l'allosédamine. Dans une seconde partie nous exposons deux méthodologies mises au point lors de nos recherches. Le premier chapitre présente un nouvel accès à des alcools homoallyliques hétéroaromatiques chiraux par réduction énantiosélective des cétones correspondantes. La compréhension des mécanismes moléculaires impliqués dans la formation des plaquettes (mégacaryopoïèse) a beaucoup progressé ces dernières années, grâce notamment à l’étude de certaines maladies génétiques provoquant des anomalies de ce processus physiologique. Parmi les nombreux récepteurs membranaires présents à la surface des plaquettes, l’intégrine αIIbβ3 en est le principal : elle permet aux plaquettes de s’agréger lors de la survenue d’une brèche vasculaire. Pourtant, la présence d’un taux normal de plaquettes chez les patients ayant un déficit constitutionnel en cette protéine a très longtemps fait évoquer que celle-ci n’était pas impliquée dans la mégacaryopoïèse. Récemment, de nouvelles observations cliniques sont venues contredire cette hypothèse en apportant de nouvelles données sur son importance dans la production normale des plaquettes
