Rather than treating overburden, the mixing auger injects water and/or air as a drilling fluid until the top of the treatment zone is reached. Reagent grout is then injected as the mixing auger moves through the treatment zone. For CCR impoundments, Discrete ISS offers the ability to produce a low permeability treated zone underneath the CCR material, hydraulically isolating it from groundwater. Forgen conducted pilot testing at a CCR impoundment in the Summer of 2022 under the direction of Silar Services Incorporated (SSi), the discrete ISS technology patent holder. In three areas of the impoundment, Forgen successfully drilled through 25 to 45 feet of CCR material and stabilized a 4 to 5-foot thick layer of ash and underlying native soil. A rosette of 7 columns was installed in each of the 3 areas. Logs were generated for each column by the drill rig monitoring system, including the inclination of the Kelly bar/drill shaft (via x and y inclinometers) and the horizontal location (northing and easting) of the mixing head tip logged continuously throughout the advancement of the auger at each column location. The logs allowed Forgen to demonstrate that complete overlap was achieved between adjacent columns. Combined with the QC results for collected wet grab samples and post-treatment core samples, Forgen was able to demonstrate that the treatment zone was emplaced at the planned top and bottom elevations and that the hydraulic conductivity of the treated material was less than 1x10-6 cm/s. The pilot study results indicate that a low permeability layer can be installed underneath the CCR material in the impoundment
Processing liquid metal for conformable electronics
Future generations of robots, electronics, and assistive medical devices will include systems that are soft, elastically deformable, and may adapt their functionality in unstructured environments. This will require soft active materials for power circuits and sensing of deformation and contact pressure. Liquid-embedded elastomer electronics offer one solution as key elements of highly deformable and soft robotic systems. Several designs for stretchable conductors and soft sensory skins (including strain, pressure, and curvature sensors) based on a liquid-embedded-elastomer approach have been developed. Many of these fluidâelastomer composites utilize liquid metal alloys due to their high conductivities and inherent compliance. Understanding how these alloys can be processed for high-yield manufacturability is critical to the development of parallel processing technology, which is needed to create more complex and low-cost systems. This discussion will highlight surface interactions between droplets of galliumâindium alloys and elastomeric substrates, and the implementation of this study to selective patterning, direct-writing, and inkjet printing of hyperelastic electronic components
Synthesis and Biological Activities of a 3â˛-Azido Analogue of Doxorubicin Against Drug-Resistant Cancer Cells
Doxorubicin (DOX), an anthracycline antibiotic, is one of the most active anticancer chemotherapeutic agents. The clinical use of DOX, however, is limited by the dose-dependant P-glycoprotein (P-gp)-mediated resistance. Herein, a 3â˛-azido analogue of DOX (ADOX) was prepared from daunorubicin (DNR). ADOX exhibited potent antitumor activities in drug-sensitive (MCF-7 and K562) and drug-resistant cell lines (MCF-7/DNR, K562/DOX), respectively. The drug resistance index (DRI) values of ADOX were much lower than that of DOX. The cytotoxicity experiments of ADOX or DOX against K562/DOX, with or without P-gp inhibitor, indicated that ADOX circumvents resistance by abolishing the P-gp recognition. This conclusion was further supported by drug influx/efflux flow cytometry experiments, as well as by molecular docking of ADOX to P-gp. In vivo animal tests, ADOX exhibited higher activity and less toxicity than DOX. The current data warranted ADOX for additional pre-clinical evaluations for new drug development
Rapid Diagnosis of Smear-Negative Tuberculosis Using Immunology and Microbiology with Induced Sputum in HIV-Infected and Uninfected Individuals
Rationale and Objectives. Blood-based studies have demonstrated the potential of immunological assays to detect tuberculosis. However lung fluid sampling may prove superior as it enables simultaneous microbiological detection of mycobacteria to be performed. Until now this has only been possible using the expensive and invasive technique of broncho-alveolar lavage. We sought to evaluate an immunoassay using non-invasive induced-sputum to diagnose active tuberculosis. Methods and Results. Prospective cohort study of forty-two spontaneous sputum smear-negative or sputum non-producing adults under investigation for tuberculosis. CD4 lymphocytes specific to purified-protein-derivative of Mycobacterium tuberculosis actively synthesising interferon-gamma were measured by flow cytometry and final diagnosis compared to immunoassay using a cut-off of 0.5%. Sixteen subjects (38%) were HIV-infected (median CD4 count [range] = 332 cells/mu l [103748]). Thirty-eight (90%) were BCG-vaccinated. In 27 subjects diagnosed with active tuberculosis, the median [range] percentage of interferon-gamma synthetic CD4+ lymphocytes was 2.77% [0-23.93%] versus 0% [0-2.10%] in 15 negative for active infection (p<0.0001). Sensitivity and specificity of the immunoassay versus final diagnosis of active tuberculosis were 89% (24 of 27) and 80% (12 of 15) respectively. The 3 positive assays in the latter group occurred in subjects diagnosed with quiescent/latent tuberculosis. Assay performance was unaffected by HIV-status, BCG-vaccination or disease site. Combining this approach with traditional microbiological methods increased the diagnostic yield to 93% (25 of 27) alongside acid-fast bacilli smear and 96% (26 of 27) alongside tuberculosis culture. Conclusions. These data demonstrate for the first time that a rapid immunological assay to diagnose active tuberculosis can be performed successfully in combination with microbiological methods on a single induced-sputum sample
A cross-sectional investigation of regional patterns of diet and cardio-metabolic risk in India
<p>Abstract</p> <p>Background</p> <p>The role of diet in India's rapidly progressing chronic disease epidemic is unclear; moreover, diet may vary considerably across North-South regions.</p> <p>Methods</p> <p>The India Health Study was a multicenter study of men and women aged 35-69, who provided diet, lifestyle, and medical histories, as well as blood pressure, fasting blood, urine, and anthropometric measurements. In each region (Delhi, n = 824; Mumbai, n = 743; Trivandrum, n = 2,247), we identified two dietary patterns with factor analysis. In multiple logistic regression models adjusted for age, gender, education, income, marital status, religion, physical activity, tobacco, alcohol, and total energy intake, we investigated associations between regional dietary patterns and abdominal adiposity, hypertension, diabetes, and dyslipidemia.</p> <p>Results</p> <p>Across the regions, more than 80% of the participants met the criteria for abdominal adiposity and 10 to 28% of participants were considered diabetic. In Delhi, the "fruit and dairy" dietary pattern was positively associated with abdominal adiposity [highest versus lowest tertile, multivariate-adjusted OR and 95% CI: 2.32 (1.03-5.23); P<sub>trend </sub>= 0.008] and hypertension [2.20 (1.47-3.31); P<sub>trend </sub>< 0.0001]. In Trivandrum, the "pulses and rice" pattern was inversely related to diabetes [0.70 (0.51-0.95); P<sub>trend </sub>= 0.03] and the "snacks and sweets" pattern was positively associated with abdominal adiposity [2.05 (1.34-3.14); P<sub>trend </sub>= 0.03]. In Mumbai, the "fruit and vegetable" pattern was inversely associated with hypertension [0.63 (0.40-0.99); P<sub>trend </sub>= 0.05] and the "snack and meat" pattern appeared to be positively associated with abdominal adiposity.</p> <p>Conclusions</p> <p>Cardio-metabolic risk factors were highly prevalent in this population. Across all regions, we found little evidence of a Westernized diet; however, dietary patterns characterized by animal products, fried snacks, or sweets appeared to be positively associated with abdominal adiposity. Conversely, more traditional diets in the Southern regions were inversely related to diabetes and hypertension. Continued investigation of diet, as well as other environmental and biological factors, will be needed to better understand the risk profile in this population and potential means of prevention.</p
