    The goals of this study were to determine the impact of sub-therapeutic chlortetracycline in market swine diets on 1) the prevalence and antimicrobial resistance of Salmonella enterica 2) antimicrobial resistance of the aerobic Gram negative fecal flora. There was no significant difference in the prevalence or antimicrobial resistance of S. enterica isolates. For the gram-negative fecal flora, there was a statistically significant difference (p\u3c0.05) between treatment groups for the frequency of antimicrobial resistance in the gram negative flora with pigs receiving chlortetracycline having a greater frequency of isolates resistant tetracycline, gentamicin, and ceftriaxone, and a lesser proportion of isolates resistant to ampicillin

    Educational Priorities for Children with Cri-Du-Chat Syndrome

    There are few data on the educational needs of children with cri-du-chat syndrome: a neurodevelopmental disorder that affects learning and development. We therefore designed an Internet survey to identify parents’ educational priorities in relation to children’s level of need/ability. The survey listed 54 skills/behaviors (e.g., toileting, expresses wants and needs, and tantrums) representing 10 adaptive behavior domains (e.g., self-care, communication, and problem behavior). Parents rated their child’s current level of ability/performance with respect to each skill/behavior and indicated the extent to which training/treatment was a priority. Fifty-four surveys were completed during the 3-month data collection period. Parents identified nine high priority skills/behaviors. Results supported the view that parent priorities are often based on the child’s deficits and emergent skills, rather than on child strengths. Implications for educational practice include the need for competence to develop high priority skills/behaviors and the value of assessing children’s deficits and emergent skills to inform the content of individualized education plans

    Meteorological and evaluation datasets for snow modelling at 10 reference sites: description of in situ and bias-corrected reanalysis data

    International audienceAbstract. This paper describes in situ meteorological forcing and evaluation data, and bias-corrected reanalysis forcing data, for cold regions' modelling at 10 sites. The long-term datasets (one maritime, one arctic, three boreal, and five mid-latitude alpine) are the reference sites chosen for evaluating models participating in the Earth System Model-Snow Model Intercomparison Project. Periods covered by the in situ data vary between 7 and 20 years of hourly meteorological data, with evaluation data (snow depth, snow water equivalent, albedo, soil temperature, and surface temperature) available at varying temporal intervals. Thirty-year (1980–2010) time series have been extracted from a global gridded surface meteorology dataset (Global Soil Wetness Project Phase 3) for the grid cells containing the reference sites, interpolated to 1 h time steps and bias-corrected. Although the correction was applied to all sites, it was most important for mountain sites hundreds of metres higher than the grid elevations and for which uncorrected air temperatures were too high and snowfall amounts too low. The discussion considers the importance of data sharing to the identification of errors and how the publication of these datasets contributes to good practice, consistency, and reproducibility in geosciences. The Supplement provides information on instrumentation, an estimate of the percentages of missing values, and gap-filling methods at each site. It is hoped that these datasets will be used as benchmarks for future model development and that their ease of use and availability will help model developers quantify model uncertainties and reduce model errors.     Incidence and Tracking of Escherichia coli O157:H7 in a Major Produce Production Region in California

    Fresh vegetables have become associated with outbreaks caused by Escherichia coli O157:H7 (EcO157). Between 1995–2006, 22 produce outbreaks were documented in the United States, with nearly half traced to lettuce or spinach grown in California. Outbreaks between 2002 and 2006 induced investigations of possible sources of pre-harvest contamination on implicated farms in the Salinas and San Juan valleys of California, and a survey of the Salinas watershed. EcO157 was isolated at least once from 15 of 22 different watershed sites over a 19 month period. The incidence of EcO157 increased significantly when heavy rain caused an increased flow rate in the rivers. Approximately 1000 EcO157 isolates obtained from cultures of>100 individual samples were typed using Multi-Locus Variable-number-tandem-repeat Analysis (MLVA) to assist in identifying potential fate and transport of EcO157 in this region. A subset of these environmental isolates were typed by Pulse Field Gel Electrophoresis (PFGE) in order to make comparisons with human clinical isolates associated with outbreak and sporadic illness. Recurrence of identical and closely related EcO157 strains from specific locations in the Salinas and San Juan valleys suggests that transport of the pathogen is usually restricted. In a preliminary study, EcO157 was detected in water at multiple locations in a low-flow creek only within 135 meters of a point source. However, possible transport up to 32 km was detected during periods of higher water flow associated with flooding. During the 2006 baby spinach outbreak investigation, transport was also detected where water was unlikely to be involved. These results indicate that contamination of the environment is a dynamic process involving multiple sources and methods of transport. Intensive studies of the sources, incidence, fate and transport of EcO157 near produce production are required to determine the mechanisms of pre-harvest contamination and potential risks for human illness

    Using C. elegans to discover therapeutic compounds for ageing-associated neurodegenerative diseases

    Age-associated neurodegenerative disorders such as Alzheimer’s disease are a major public health challenge, due to the demographic increase in the proportion of older individuals in society. However, the relatively few currently approved drugs for these conditions provide only symptomatic relief. A major goal of neurodegeneration research is therefore to identify potential new therapeutic compounds that can slow or even reverse disease progression, either by impacting directly on the neurodegenerative process or by activating endogenous physiological neuroprotective mechanisms that decline with ageing. This requires model systems that can recapitulate key features of human neurodegenerative diseases that are also amenable to compound screening approaches. Mammalian models are very powerful, but are prohibitively expensive for high-throughput drug screens. Given the highly conserved neurological pathways between mammals and invertebrates, Caenorhabditis elegans has emerged as a powerful tool for neuroprotective compound screening. Here we describe how C. elegans has been used to model various human ageing-associated neurodegenerative diseases and provide an extensive list of compounds that have therapeutic activity in these worm models and so may have translational potential

    "The Violent Take It by Force": Heathcliff and the Vitalizing Power of Mayhem in "Wuthering Heights"

    In Wuthering Heights, Emily Bronte employs the character Heathcliff as both a real and mythic being in order to challenge class conventions in Victorian society. She shares this societal contention with other Victorian novelists, but where her contemporaries are typically realistic in their works, Bronte creates a concurrent mythic realm alongside the real in order to allow Heathcliff the space and license to be a Revenant, a symbol used in the folk tradition of the Scots, which I contend was a likely influence on Bronte’s work. Heathcliff’s real nature clashes with this symbolic one, especially when reality will not allow him to be with Catherine, the woman he loves. Her rejection of him serves two central purposes: 1) for the author to spotlight the arbitrary nature of the class system and the decisions individuals make inside it; and 2) for the author to provide a pivot point in the story at which she transforms Heathcliff from a real character to a mythic one. Heathcliff spends the latter half of the novel exacting redemptive punishment on all who have wronged him (and the marginalized he represents), including Catherine herself, a reality he struggles with because he still loves her despite her class-motivated marriage to the hated Edgar Linton. In the end, Heathcliff transgresses his symbolic purpose by going too far in punishing the innocent Hareton, at which point Bronte has him die as unceremoniously as she did Catherine earlier in the novel. Young Hareton and Cathy’s relationship is the fruit of the Revenant Heathcliff’s redeeming work, an ending that, for Bronte, seems to merge more than just the two houses; it seems to also reconcile divergent and conflicting ways of thinking inside the class system

    Rising fluoroquinolone resistance in \u3ci\u3eCampylobacter\u3c/i\u3e isolated from feedlot cattle in the United States

    Antibiotic resistance, particularly to fluoroquinolones and macrolides, in the major foodborne pathogen Campylobacter is considered a serious threat to public health. Although ruminant animals serve as a significant reservoir for Campylobacter, limited information is available on antibiotic-resistant Campylobacter of bovine origin. Here, we analyzed the antimicrobial susceptibilities of 320 C. jejuni and 115 C. coli isolates obtained from feedlot cattle farms in multiple states in the U.S. The results indicate that fluoroquinolone resistance reached to 35.4% in C. jejuni and 74.4% in C. coli, which are significantly higher than those previously reported in the U.S. While all fluoroquinolone resistant (FQR) C. coli isolates examined in this study harbored the single Thr-86-Ile mutation in GyrA, FQR C. jejuni isolates had other mutations in GyrA in addition to the Thr-86-Ile change. Notably, most of the analyzed FQR C. coli isolates had similar PFGE (pulsed field gel electrophoresis) patterns and the same MLST (multilocus sequence typing) sequence type (ST-1068) regardless of their geographic sources and time of isolation, while the analyzed C. jejuni isolates were genetically diverse, suggesting that clonal expansion is involved in dissemination of FQR C. coli but not C. jejuni. These findings reveal the rising prevalence of FQR Campylobacter in the U.S. and provide novel information on the epidemiology of antibiotic-resistant Campylobacter in the ruminant reservoir
