    A viral vector model for circuit-specific synucleinopathy

    In Parkinson's disease (PD), pathomechanisms such as aberrant network dysfunctions can be elucidated by conducting multiscale explorations in animal models. However, the lack of specificity in the existing models limits a restricted targeting of individual network elements and characterization of PD as a “circuitopathy”. We therefore developed a cell-type specific viral vector (AAV2/9-CreON-A53T-αSyn) mouse model that allows to induce synucleinopathy within individual circuit elements in vivo. When specifically targeted to dopaminergic (DA) neurons of the substantia nigra pars compacta (SNc), our approach recapitulates the main hallmarks of the disease, namely Lewy-body-like aggregation, progressive cellular and nigrostriatal projections loss, together with locomotor impairment. Our strategy is supported by new state-of-the-art analytical approaches for cell quantification and behavior characterization. Altogether, we provide a novel model of synucleinopathy, which offers new opportunities to study the contribution of individual network elements to disease pathomechanisms.Fil: Lantheaume, Alexia.     Cet ouvrage collectif prĂ©sente les rĂ©sultats d’une enquĂȘte internationale menĂ©e auprĂšs d’environ 7 000 Ă©lĂšves ĂągĂ©s de 11 Ă  19 ans, Ă  qui il a Ă©tĂ© demandĂ© de raconter l’histoire nationale. Des Ă©lĂšves français, suisses, catalans, allemands ont rĂ©pondu Ă  un questionnaire et racontĂ© librement « leur » histoire nationale. Contre les idĂ©es reçues d’un manque de connaissances historiques et de l’absence de tout rĂ©cit commun, l’enquĂȘte dĂ©voile des formes de narration, des organisateurs du rĂ©cit et des contenus partagĂ©s, une vĂ©ritable trame commune. L’analyse de ces formes et contenus prĂ©cise leurs points forts, mais aussi les absences ou les spĂ©cificitĂ©s nationales, et montre comment les savoirs scolaires coexistent avec d’autres sources de connaissances. Sont Ă©tudiĂ©es en particulier les places respectives et les fonctions du politique, de la religion, de la guerre, dans ces rĂ©cits qui tĂ©moignent d’un imaginaire national. Un effet « territorial » est enfin mis en valeur, Ă  la fois moins important que prĂ©vu en France et significatif Ă  l’échelle des pays. Sensibles au contexte, ces rĂ©cits sont la manifestation juvĂ©nile d’une conscience historique en formation, qui s’exprime selon des formes inĂ©dites
