    Report description: The NNI was launched in 2001 to provide high quality childcare in the most disadvantaged neighbourhoods of England, to help parents into employment, reduce child poverty and boost children’s development. By 2005 45,000 new childcare places had been created in approximately 1,400 neighbourhood nurseries. This report brings together the findings of the four individual strands of the National Evaluation of Neighbourhood Nurseries Initiative as shown above and makes a number of recommendations. The report shows the rationale for the government’s strategy in targeting disadvantaged neighbourhoods and in focusing on high quality childcare to provide the link between raising parental employment and income and improving children’s life chances

    On the use of Parylene C polymer as substrate for peripheral nerve electrodes

    Parylene C is a highly flexible polymer used in several biomedical implants. Since previous studies have reported valuable biocompatible and manufacturing characteristics for brain and intraneural implants, we tested its suitability as a substrate for peripheral nerve electrodes. We evaluated 1-year-aged in vitro samples, where no chemical differences were observed and only a slight deviation on Young's modulus was found. The foreign body reaction (FBR) to longitudinal Parylene C devices implanted in the rat sciatic nerve for 8 months was characterized. After 2 weeks, a capsule was formed around the device, which continued increasing up to 16 and 32 weeks. Histological analyses revealed two cell types implicated in the FBR: macrophages, in contact with the device, and fibroblasts, localized in the outermost zone after 8 weeks. Molecular analysis of implanted nerves comparing Parylene C and polyimide devices revealed a peak of inflammatory cytokines after 1 day of implant, returning to low levels thereafter. Only an increase of CCL2 and CCL3 was found at chronic time-points for both materials. Although no molecular differences in the FBR to both polymers were found, the thick tissue capsule formed around Parylene C puts some concern on its use as a scaffold for intraneural electrodes

    Long-term functionality of transversal intraneural electrodes is improved by dexamethasone treatment

Among the different designs, intraneural electrodes implanted in peripheral nerves represent a good strategy to stimulate nerve fibers to send sensory feedback and to record nerve signals to control the prosthetic limb. However, intraneural electrodes, as any device implanted in the body, induce a foreign body reaction (FBR) that results in the tissue encapsulation of the device. The FBR causes a progressive decline of the electrode functionality over time due to the physical separation between the electrode active sites and the axons to interface. Modulation of the inflammatory response has arisen as a good strategy to reduce the FBR and maintain electrode functionality. In this study transversal intraneural multi-channel electrodes (TIMEs) were implanted in the rat sciatic nerve and tested for 3 months to evaluate stimulation and recording capabilities under chronic administration of dexamethasone. Dexamethasone treatment significantly reduced the threshold for evoking muscle responses during the follow-up compared to saline-treated animals, without affecting the selectivity of stimulation. However, dexamethasone treatment did not improve the signal-to-noise ratio of the recorded neural signals. Dexamethasone treatment allowed to maintain more working active sites along time than saline treatment. Thus, systemic administration of dexamethasone appears as a useful treatment in chronically implanted animals with neural electrodes as it increases the number of functioning contacts of the implanted TIME and reduces the intensity needed to stimulate the nerve

    Nutrient-rich submarine groundwater discharge increases algal carbon uptake in a tropical reef ecosystem

    Submarine groundwater discharge (SGD) in high volcanic islands can be an important source of freshwater and nutrients to coral reefs. High inorganic nutrient content is generally thought to augment primary production in coastal systems but when this is delivered via a freshwater vector as is the case with SGD in this study, the effects on productivity are unclear. In the current literature, there is limited evidence for a direct association between SGD and primary productivity of reefs. To elucidate the response of primary productivity to SGD, we conducted spatially and temporally explicit in situ benthic chamber experiments on a reef flat along a gradient of SGD. We found significant quadratic relationships between C-uptake and SGD for both phytoplankton and the most abundant macroalga, Gracilaria salicornia, with uptake maxima at SGD-derived salinities of ~21−22 (24.5−26.6 μmol NO3-L−1). These results suggest a physiological tradeoff between salinity tolerance and nutrient availability for reef primary producers. Spatially explicit modeling of reefs with SGD and without SGD indicate reef-scale G. salicornia and phytoplankton C-uptake decreased by 82% and 36% in the absence of SGD, respectively. Thus, nutrient-rich and low salinity SGD has significant effects on algal C-uptake in reef systems

    SN 1988Z: Spectro-photometric catalogue and energy estimates

    We present a spectro-photometric catalogue of the evolution of supernova 1988Z which combines new and published observations in the radio, optical and X-ray bands, with the aim of offering a comprehensive view of the evolution of this object and deriving the total energy radiated since discovery. The major contribution to the total radiated energy comes at optical to X-ray frequencies, with a total emission of at least 2×10512 \times 10^{51} erg (for Ho=50 km/s) in 8.5 years. A model-dependent extrapolation of this value indicates that the total radiated energy may be as high as 105210^{52} erg. The high value of the radiated energy supports a scenario in which most of the kinetic energy of the ejecta is thermalized and radiated in a short interaction with a dense circumstellar medium of nearly constant density.     Identification and characterization of Rhipicephalus (Boophilus) microplus candidate protective antigens for the control of cattle tick infestations

    The cattle ticks, Rhipicephalus (Boophilus) spp., affect cattle production in tropical and subtropical regions of the world. Tick vaccines constitute a cost-effective and environmentally friendly alternative to tick control. The recombinant Rhipicephalus microplus Bm86 antigen has been shown to protect cattle against tick infestations. However, variable efficacy of Bm86-based vaccines against geographic tick strains has encouraged the research for additional tick-protective antigens. Herein, we describe the analysis of R. microplus glutathione-S transferase, ubiquitin (UBQ), selenoprotein W, elongation factor-1 alpha, and subolesin (SUB) complementary DNAs (cDNAs) by RNA interference (RNAi) in R. microplus and Rhipicephalus annulatus. Candidate protective antigens were selected for vaccination experiments based on the effect of gene knockdown on tick mortality, feeding, and fertility. Two cDNA clones encoding for UBQ and SUB were used for cattle vaccination and infestation with R. microplus and R. annulatus. Control groups were immunized with recombinant Bm86 or adjuvant/saline. The highest vaccine efficacy for the control of tick infestations was obtained for Bm86. Although with low immunogenic response, the results with the SUB vaccine encourage further investigations on the use of recombinant subolesin alone or in combination with other antigens for the control of cattle tick infestations. The UBQ peptide showed low immunogenicity, and the results of the vaccination trial were inconclusive to assess the protective efficacy of this antigen. These experiments showed that RNAi could be used for the selection of candidate tick-protective antigens. However, vaccination trials are necessary to evaluate the effect of recombinant antigens in the control of tick infestations, a process that requires efficient recombinant protein production and formulation systems
