PLANTS WITH USEFUL TRAITS AND RELATED METHODS

An investigation of latent fingermark residues and their development on porous substrates using physical developer and nile red
University of Technology Sydney. Faculty of Science.The detection of fingermarks on porous surfaces that have been wet is currently limited to only one routine technique, Physical Developer (PD). PD is a well-established fingermark development method that has a plethora of issues associated with its use, principally based around the preparation and relative instability of the working solution and lack of contrast on dark substrates as it is non-luminescent. While these issues have been mostly addressed at an operational level, in-depth chemical remedies for the PD process cannot be devised because the mechanisms and specific targets of PD remain largely unknown. This highlights the need to understand the fundamental chemistry of the technique to recognise what may be improved and what complementary detection methods could be applied to ensure that the maximum number of available fingermarks are developed in casework.
This research has also considered nile red (NR), a technique known to develop fingermarks on porous substrates that have been wet that remain undeveloped by treatment with PD. An investigation of the optimisation of the NR working solution was undertaken. An oil-in-water microemulsion containing NR was prepared using the solvent-diffusion method. The optimized NR solution was demonstrated to be effective in developing fresh latent fingermarks. The working solution is prepared with the slow addition of a NR in dichloromethane stock solution to a dual surfactant solution to create a lactescent dual organic-aqueous phase intermediate. Stirring promotes the evaporation of the solvent, resulting in a transparent NR microemulsion. The solution contains less hazardous solvents than the previously published formulation, with an extended shelf life and at a much lower cost. The optimized microemulsion was shown to outperform a previously reported aqueous nile blue working solution on both natural and groomed fingermarks, with shorter exposure times for image capture after development.
The storage, ageing and concentration of the components of a PD working solution was evaluated to determine the most effective formulation and protocol for the use of PD. A more thorough understanding of the working solution components of PD has allowed a refinement of the development method and the formulation itself, resulting in increased development success and a better understanding of when fingermarks will and will not be effectively developed by the technique. A revised PD working solution formulation and development protocol has been established that has been successfully utilised by undergraduate forensic science students.
An investigation of the chemical targets of PD contained in latent fingermark deposits through reactivity assessments of various lipid, eccrine and lipid–eccrine mixtures in the residue was undertaken. Experimental results showed that silver deposition from the PD working solution occurs in the presence of an emulsion of both lipid and eccrine constituents. PD was interestingly shown to also be reactive towards emulsions of oleic acid and water, indicating that the silver deposition may occur as a result of nucleation sites at the emulsion phase boundary, or as a result of desorption of constituents from the substrate that promote silver reduction. These results explain the anomalies seen with the selective development by PD of a proportion of fingermarks on a substrate, and the development of other fingermarks by other techniques when used in sequence, such as Oil Red O (ORO) or NR. A consideration of the various processes that emulsions undergo also helps to rationalise the fluctuating developmental effectiveness of PD on ageing fingermarks.
A better understanding of the PD technique permits a more informed selection of alternative or complementary detection methods. This research provides further insight into not only the application of the PD technique but, more importantly, into the reasons behind the observed development. This increased understanding highlights the need to sequence PD with a lipid sensitive technique, such as NR, for the development of latent fingermarks on porous substrates that have been wet as the two techniques have discrete and complementary targets. It also emphasizes the need to develop a better understanding of latent fingermark residue–substrate interactions
A 3-player protocol preventing persistence in strategic contention with limited feedback
In this paper, we study contention resolution protocols from a game-theoretic
perspective. In a recent work, we considered acknowledgment-based protocols,
where a user gets feedback from the channel only when she attempts
transmission. In this case she will learn whether her transmission was
successful or not. One of the main results of ESA2016 was that no
acknowledgment-based protocol can be in equilibrium. In fact, it seems that
many natural acknowledgment-based protocols fail to prevent users from
unilaterally switching to persistent protocols that always transmit with
probability 1. It is therefore natural to ask how powerful a protocol must be
so that it can beat persistent deviators.
In this paper we consider age-based protocols, which can be described by a
sequence of probabilities of transmitting in each time step. Those
probabilities are given beforehand and do not change based on the transmission
history. We present a 3-player age-based protocol that can prevent users from
unilaterally deviating to a persistent protocol in order to decrease their
expected transmission time. It is worth noting that the answer to this question
does not follow from the results and proof ideas of ESA2016. Our protocol is
non-trivial, in the sense that, when all players use it, finite expected
transmission time is guaranteed. In fact, we show that this protocol is
preferable to any deadline protocol in which, after some fixed time, attempt
transmission with probability 1 in every subsequent step. An advantage of our
protocol is that it is very simple to describe, and users only need a counter
to keep track of time. Whether there exist -player age-based protocols that
do not use counters and can prevent persistence is left as an open problem for

Rainfall and sentinel chicken seroconversions predict human cases of Murray Valley encephalitis in the north of Western Australia
Background Murray Valley encephalitis virus (MVEV) is a flavivirus that occurs in Australia and New Guinea. While clinical cases are uncommon, MVEV can cause severe encephalitis with high mortality. Sentinel chicken surveillance is used at many sites around Australia to provide an early warning system for risk of human infection in areas that have low population density and geographical remoteness. MVEV in Western Australia occurs in areas of low population density and geographical remoteness, resulting in logistical challenges with surveillance systems and few human cases. While epidemiological data has suggested an association between rainfall and MVEV activity in outbreak years, it has not been quantified, and the association between rainfall and sporadic cases is less clear. In this study we analysed 22 years of sentinel chicken and human case data from Western Australia in order to evaluate the effectiveness of sentinel chicken surveillance for MVEV and assess the association between rainfall and MVEV activity. Methods Sentinel chicken seroconversion, human case and rainfall data from the Kimberley and Pilbara regions of Western Australia from 1990 to 2011 were analysed using negative binomial regression. Sentinel chicken seroconversion and human cases were used as dependent variables in the model. The model was then tested against sentinel chicken and rainfall data from 2012 and 2013.Results Sentinel chicken seroconversion preceded all human cases except two in March 1993. Rainfall in the prior three months was significantly associated with both sentinel chicken seroconversion and human cases across the regions of interest. Sentinel chicken seroconversion was also predictive of human cases in the models. The model predicted sentinel chicken seroconversion in the Kimberley but not in the Pilbara, where seroconversions early in 2012 were not predicted. The latter may be due to localised MVEV activity in isolated foci at dams, which do not reflect broader virus activity in the region. Conclusions We showed that rainfall and sentinel chickens provide a useful early warning of MVEV risk to humans across endemic and epidemic areas, and that a combination of the two indicators improves the ability to assess MVEV risk and inform risk management measures
Hierarchical Hough all-sky search for periodic gravitational waves in LIGO S5 data
We describe a new pipeline used to analyze the data from the fifth science
run (S5) of the LIGO detectors to search for continuous gravitational waves
from isolated spinning neutron stars. The method employed is based on the Hough
transform, which is a semi-coherent, computationally efficient, and robust
pattern recognition technique. The Hough transform is used to find signals in
the time-frequency plane of the data whose frequency evolution fits the pattern
produced by the Doppler shift imposed on the signal by the Earth's motion and
the pulsar's spin-down during the observation period. The main differences with
respect to previous Hough all-sky searches are described. These differences
include the use of a two-step hierarchical Hough search, analysis of
coincidences among the candidates produced in the first and second year of S5,

CRISPR-Cas9 screens in human cells and primary neurons identify modifiers of C9ORF72 dipeptide-repeat-protein toxicity.
Hexanucleotide-repeat expansions in the C9ORF72 gene are the most common cause of amyotrophic lateral sclerosis and frontotemporal dementia (c9ALS/FTD). The nucleotide-repeat expansions are translated into dipeptide-repeat (DPR) proteins, which are aggregation prone and may contribute to neurodegeneration. We used the CRISPR-Cas9 system to perform genome-wide gene-knockout screens for suppressors and enhancers of C9ORF72 DPR toxicity in human cells. We validated hits by performing secondary CRISPR-Cas9 screens in primary mouse neurons. We uncovered potent modifiers of DPR toxicity whose gene products function in nucleocytoplasmic transport, the endoplasmic reticulum (ER), proteasome, RNA-processing pathways, and chromatin modification. One modifier, TMX2, modulated the ER-stress signature elicited by C9ORF72 DPRs in neurons and improved survival of human induced motor neurons from patients with C9ORF72 ALS. Together, our results demonstrate the promise of CRISPR-Cas9 screens in defining mechanisms of neurodegenerative diseases
Physical and mental health comorbidity is common in people with multiple sclerosis: nationally representative cross-sectional population database analysis
<b>Background</b> Comorbidity in Multiple Sclerosis (MS) is associated with worse health and higher mortality. This study aims to describe clinician recorded comorbidities in people with MS. <p></p>
<b>Methods</b> 39 comorbidities in 3826 people with MS aged ≥25 years were compared against 1,268,859 controls. Results were analysed by age, gender, and socioeconomic status, with unadjusted and adjusted Odds Ratios (ORs) calculated using logistic regression. <p></p>
<b>Results</b> People with MS were more likely to have one (OR 2.44; 95% CI 2.26-2.64), two (OR 1.49; 95% CI 1.38-1.62), three (OR 1.86; 95% CI 1.69-2.04), four or more (OR 1.61; 95% CI 1.47-1.77) non-MS chronic conditions than controls, and greater mental health comorbidity (OR 2.94; 95% CI 2.75-3.14), which increased as the number of physical comorbidities rose. Cardiovascular conditions, including atrial fibrillation (OR 0.49; 95% CI 0.36-0.67), chronic kidney disease (OR 0.51; 95% CI 0.40-0.65), heart failure (OR 0.62; 95% CI 0.45-0.85), coronary heart disease (OR 0.64; 95% CI 0.52-0.71), and hypertension (OR 0.65; 95% CI 0.59-0.72) were significantly less common in people with MS. <p></p>
<b>Conclusion</b> People with MS have excess multiple chronic conditions, with associated increased mental health comorbidity. The low recorded cardiovascular comorbidity warrants further investigation
Long-acting injectable multipurpose prevention technology for prevention of HIV and unplanned pregnancy
Only condoms are proven to protect against both HIV and unplanned pregnancy, however, poor user acceptability and lack of partner cooperation impede effectiveness. We developed an injectable ultra-long-acting, biodegradable, and removable in-situ forming implant (ISFI) as multipurpose prevention technology (MPT). MPT ISFIs co-formulated an antiretroviral (dolutegravir (DTG)) or cabotegravir (CAB)), and a hormonal contraceptive (etonogestrel (ENG) or medroxyprogesterone acetate (MPA)). All formulations were well-tolerated in mice with no signs of chronic local or systemic inflammation. Plasma CAB and DTG concentrations were above 4x PA-IC90 for 90 days with zero-order and diffusion-controlled absorption, respectively, and no differences when co-formulated with either hormone. Plasma ENG and MPA concentrations were quantifiable for 90 days. Complete removal of CAB/MPA ISFIs resulted in MPA concentrations falling below the limit of quantification after 24 h post-removal, but incomplete CAB elimination from plasma. Collectively, we demonstrated the ability to co-formulate antiretrovirals with contraceptives in an ISFI that is well-tolerated with sustained plasma concentrations up to 90 days
Power grip, pinch grip, manual muscle testing or thenar atrophy - which should be assessed as a motor outcome after carpal tunnel decompression? A systematic review
<p>Abstract</p> <p>Background</p> <p>Objective assessment of motor function is frequently used to evaluate outcome after surgical treatment of carpal tunnel syndrome (CTS). However a range of outcome measures are used and there appears to be no consensus on which measure of motor function effectively captures change. The purpose of this systematic review was to identify the methods used to assess motor function in randomized controlled trials of surgical interventions for CTS. A secondary aim was to evaluate which instruments reflect clinical change and are psychometrically robust.</p> <p>Methods</p> <p>The bibliographic databases Medline, AMED and CINAHL were searched for randomized controlled trials of surgical interventions for CTS. Data on instruments used, methods of assessment and results of tests of motor function was extracted by two independent reviewers.</p> <p>Results</p> <p>Twenty-two studies were retrieved which included performance based assessments of motor function. Nineteen studies assessed power grip dynamometry, fourteen studies used both power and pinch grip dynamometry, eight used manual muscle testing and five assessed the presence or absence of thenar atrophy. Several studies used multiple tests of motor function. Two studies included both power and pinch strength and reported descriptive statistics enabling calculation of effect sizes to compare the relative responsiveness of grip and pinch strength within study samples. The study findings suggest that tip pinch is more responsive than lateral pinch or power grip up to 12 weeks following surgery for CTS.</p> <p>Conclusion</p> <p>Although used most frequently and known to be reliable, power and key pinch dynamometry are not the most valid or responsive tools for assessing motor outcome up to 12 weeks following surgery for CTS. Tip pinch dynamometry more specifically targets the thenar musculature and appears to be more responsive. Manual muscle testing, which in theory is most specific to the thenar musculature, may be more sensitive if assessed using a hand held dynamometer – the Rotterdam Intrinsic Handheld Myometer. However further research is needed to evaluate its reliability and responsiveness and establish the most efficient and psychometrically robust method of evaluating motor function following surgery for CTS.</p
