    Structured fibrous carbon-based catalyst for continuous nitrate removal from natural water

    [EN] Bimetallic (Pd–Cu, Pd–Sn) nanoparticles supported on structured fibrous carbons (activated carbon fibers and carbon nanofibers grown on sintered metal fibers) were tested in nitrate removal of natural polluted water by hydrogen (a batch and continuous mode). Dependence of the activity/selectivity on catalyst chemical composition, promoter nature and metal particle size was studied. Sn-modified Pd nanoparticles showed higher N2 selectivity as compared to Cu-modified ones.     Probing of a human proteome microarray with a recombinant pathogen protein reveals a novel mechanism by which hookworms suppress B-cell receptor signaling.

    Na-ASP-2 is an efficacious hookworm vaccine antigen. However, despite elucidation of its crystal structure and studies addressing its immunobiology, the function of Na-ASP-2 has remained elusive. We probed a 9000-protein human proteome microarray with Na-ASP-2 and showed binding to CD79A, a component of the B-cell antigen receptor complex. Na-ASP-2 bound to human B lymphocytes ex vivo and downregulated the transcription of approximately 1000 B-cell messenger RNAs (mRNAs), while only approximately 100 mRNAs were upregulated, compared with control-treated cells. The expression of a range of molecules was affected by Na-ASP-2, including factors involved in leukocyte transendothelial migration pathways and the B-cell signaling receptor pathway. Of note was the downregulated transcription of lyn and pi3k, molecules that are known to interact with CD79A and control B-cell receptor signaling processes. Together, these results highlight a previously unknown interaction between a hookworm-secreted protein and B cells, which has implications for helminth-driven immunomodulation and vaccine development. Further, the novel use of human protein microarrays to identify host-pathogen interactions, coupled with ex vivo binding studies and subsequent analyses of global gene expression in human host cells, demonstrates a new pipeline by which to explore the molecular basis of infectious diseases.This is the author accepted version.     MGMT gene silencing and benefit from temozolomide in glioblastoma.

    BACKGROUND: Epigenetic silencing of the MGMT (O6-methylguanine-DNA methyltransferase) DNA-repair gene by promoter methylation compromises DNA repair and has been associated with longer survival in patients with glioblastoma who receive alkylating agents. METHODS: We tested the relationship between MGMT silencing in the tumor and the survival of patients who were enrolled in a randomized trial comparing radiotherapy alone with radiotherapy combined with concomitant and adjuvant treatment with temozolomide. The methylation status of the MGMT promoter was determined by methylation-specific polymerase-chain-reaction analysis. RESULTS: The MGMT promoter was methylated in 45 percent of 206 assessable cases. Irrespective of treatment, MGMT promoter methylation was an independent favorable prognostic factor (P<0.001 by the log-rank test; hazard ratio, 0.45; 95 percent confidence interval, 0.32 to 0.61). Among patients whose tumor contained a methylated MGMT promoter, a survival benefit was observed in patients treated with temozolomide and radiotherapy; their median survival was 21.7 months (95 percent confidence interval, 17.4 to 30.4), as compared with 15.3 months (95 percent confidence interval, 13.0 to 20.9) among those who were assigned to only radiotherapy (P=0.007 by the log-rank test). In the absence of methylation of the MGMT promoter, there was a smaller and statistically insignificant difference in survival between the treatment groups. CONCLUSIONS: Patients with glioblastoma containing a methylated MGMT promoter benefited from temozolomide, whereas those who did not have a methylated MGMT promoter did not have such a benefit

    The use of Pd catalysts on carbon-based structured materials for the catalytic hydrogenation of bromates in different types of water

    [EN] The aim of this work is to study the activity of new Pd catalysts, supported on two different nano structured carbon materials, for bromate catalytic hydrogenation. The influence of the support has been studied, obtaining the best results with a palladium catalyst supported on carbon nanofibers (CNF) grown in sintered metal fibers (SMF). The results have shown the importance of the catalyst support in order to minimize the mass-transfer limitations ensuring an efficient catalyst use. In this way the most active catalysts are those with a mesoporous structure containing high dispersed Pd nanoparticles. The activity of this catalyst for bromate reduction has been tested in different types of water, namely, distilled water, natural water and industrial wastewater. It has been shown that the catalyst activity depends on the water matrix and bromate reduction rate depends on the hydrogen partial pressure. The potential use of the catalyst has been studied in a continuous reactor.     Recombinant humanised anti-HER2/neu antibody (HerceptinÂź) induces cellular death of glioblastomas

    Glioblastoma multiforme (GBM) remains the most devastating primary tumour in neuro-oncology. Targeting of the human epithelial receptor type 2 (HER2)-neu receptor by specific antibodies is a recent well-established therapy for breast tumours. Human epithelial receptor type 2/neu is a transmembrane tyrosine/kinase receptor that appears to be important for the regulation of cancer growth. Human epithelial receptor type 2/neu is not expressed in the adult central nervous system, but its expression increases with the degree of astrocytoma anaplasia. The specificity of HER2/neu for tumoral astrocytomas leads us to study in vitro treatment of GBM with anti-HER2/neu antibody. We used human GBM cell lines expressing HER2/neu (A172 express HER2/neu more than U251MG) or not (U87MG) and monoclonal humanised antibody against HER2/neu (HerceptinÂź). Human epithelial receptor type 2/neu expression was measured by immunohistochemistry and flow cytometry. Direct antibody effect, complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity were evaluated by different cytometric assays. We have shown, for the first time, the ability of anti-HER2/neu antibodies to induce apoptosis and cellular-dependent cytotoxicity of HER2/neu-expressing GBM cell lines. The results decreased from A172 to U251 and were negative for U87MG, in accordance with the decreasing density of HER2/neu receptors

    Individual adaptability : validation and application of the concept

    A novel composite material based on carbon nanofibers (CNF) grown on sintered metal fibers (SMFInconel) filter was investigated for its favorable properties as catalytic support. The CNF were formed directly over the SMFInconel by thermal (650 degrees C) chemical vapor deposition of ethane-hydrogen mixture. The CNF/SMFInconel composite consists of metal fibers entangled by CNF network of microns thickness and strongly anchored to the metal surface. The properties of the CNF/SMFInconel were controlled by the synthesis conditions. The CNF coating reduced the filter porosity and decreased the material permeability, but the pressure drop during the gas passage through the reactor remained low for CNF fraction < 10 %. Palladium was deposited on CNF previously activated by boiling in hydrogen peroxide. The activity and selectivity of Pd degrees/CNF/SMFInconel catalysts were tested in the selective hydrogenation of acetylene and compared with Pd supported on activated carbon fibers (Pd degrees/ACF). The TOF was one order of magnitude higher for Pd degrees/CNF/SMFInconel as compared to Pd degrees/ACF. This effect was attributed to a strong metal-support interaction of Pd degrees-nanoparticles with the graphitized CNF. The reaction was found to be structure sensitive leading to a decrease of TOF for the Pd degrees-particles < 3 nm

    The authors present a case of a tuberothalamic infarct subsequent to division of the posterior communicating artery for clipping of a high-lying aneurysm of the basilar bifurcation using the pterional approach. In view of this clinical observation and some particular aspects of the microsurgical anatomy of the perforating vessels of the posterior communicating artery, we conclude that interrupting this parent vessel carries a significant risk of infarction
