    Physiological Effects of Tapering and Detraining in World-Class Kayakers

    . Purpose: This study analyzed changes in neuromuscular, body composition, and endurance markers during 4 wk of tapering and subsequent 5 wk of reduced training (RT) or training cessation (TC). Methods: Fourteen world-class kayakers were randomly assigned to either a TC (n = 7) or an RT group (n = 7). One-repetition maximum (1RM) strength, mean concentric velocity with 45% 1RM (V 45% ) in the bench press (BP) and prone bench pull (PBP) exercises, and body composition assessments were conducted at the start (T0) and end (T1) of a 43-wk training program, after tapering for the world championships (T2) and after TC or RT (T3). A graded exercise test on a kayak ergometer for determination of maximal oxygen uptake at T0, T1, and T3 was also performed. Results: After tapering, no significant changes were observed in 1RM or V 45% . TC resulted in significantly greater declines in 1RM strength (j8.9% and j7.8%, P G 0.05, respectively, for BP and PBP) than those observed for RT (j3.9% and j3.4%). Decreases in V 45% in BP and PBP were larger for TC (j12.6% and j10.0%) than for RT (j9.0% and j6.7%). Increases in sum of eight skinfolds were observed after both TC and RT, whereas declines in maximal aerobic power were lower for RT (j5.6%) than for TC (j11.3%). Conclusions: Short-term TC results in large decreases in maximal strength and especially V 45% in highly trained athletes. These results suggest the need of performing a minimal maintenance program to avoid excessive declines in neuromuscular function in cases where a prolonged break from training is required.     Melatonin MT1 and MT2 receptors in the ram reproductive tract

    Some melatonin functions in mammals are exerted through MT1 and MT2 receptors. However, there are no reports of their presence in the reproductive tract of the ram, a seasonal species. Thus, we have investigated their existence in the ram testis, epididymis, accessory glands and ductus deferens. Real-time polymerase chain reaction (qPCR) revealed higher levels of m-RNA for both receptors in the testis, ampulla, seminal vesicles, and vas deferens, than in the other organs of the reproductive tract (p < 0.05). Western blot analyses showed protein bands compatible with the MT1 in the testis and cauda epididymis, and for the MT2 in the cauda epididymis and deferent duct. Immunohistochemistry analyses revealed the presence of MT1 receptors in spermatogonias, spermatocytes, and spermatids, and MT2 receptors in the newly-formed spermatozoa in the testis, whereas both receptors were located in the epithelial cells of the ampulla, seminal vesicles, and ductus deferens. Indirect immunofluorescence showed significant differences in the immunolocation of both receptors in spermatozoa during their transit in the epididymis. In conclusion, it was demonstrated that melatonin receptors are present in the ram reproductive tract. These results open the way for new studies on the molecular mechanism of melatonin and the biological significance of its receptors

    Climate and atmospheric history of the past 420,000 years from the Vostok ice core,

    Antarctica has allowed the extension of the ice record of atmospheric composition and climate to the past four glacial-interglacial cycles. The succession of changes through each climate cycle and termination was similar, and atmospheric and climate properties oscillated between stable bounds. Interglacial periods differed in temporal evolution and duration. Atmospheric concentrations of carbon dioxide and methane correlate well with Antarctic air-temperature throughout the record. Present-day atmospheric burdens of these two important greenhouse gases seem to have been unprecedented during the past 420,000 years. The late Quaternary period (the past one million years) is punctuated by a series of large glacial-interglacial changes with cycles that last about 100,000 years (ref. 1). Glacial-interglacial climate changes are documented by complementary climate records 1,2 largely derived from deep sea sediments, continental deposits of flora, fauna and loess, and ice cores. These studies have documented the wide range of climate variability on Earth. They have shown that much of the variability occurs with periodicities corresponding to that of the precession, obliquity and eccentricity of the Earth&apos;s orbit 1,3 . But understanding how the climate system responds to this initial orbital forcing is still an important issue in palaeoclimatology, in particular for the generally strong ϳ100,000-year (100-kyr) cycle. Ice cores give access to palaeoclimate series that includes local temperature and precipitation rate, moisture source conditions, wind strength and aerosol fluxes of marine, volcanic, terrestrial, cosmogenic and anthropogenic origin. They are also unique with their entrapped air inclusions in providing direct records of past changes in atmospheric trace-gas composition. The ice-drilling project undertaken in the framework of a long-term collaboration between Russia, the United States and France at the Russian Vostok station in East Antarctica (78Њ S, 106Њ E, elevation 3,488 m, mean temperature −55 ЊC) has already provided a wealth of such information for the past two glacial-interglacial cycles [4][5][6][7][8][9] Here we present a series of detailed Vostok records covering this ϳ400-kyr period. We show that the main features of the more recent Vostok climate cycle resemble those observed in earlier cycles. In particular, we confirm the strong correlation between atmospheric greenhouse-gas concentrations and Antarctic temperature, as well as the strong imprint of obliquity and precession in most of the climate time series. Our records reveal both similarities and differences between the successive interglacial periods. They suggest the lead of Antarctic air temperature, and of atmospheric greenhousegas concentrations, with respect to global ice volume and Greenland air-temperature changes during glacial terminations.     Pathogenetics of alveolar capillary dysplasia with misalignment of pulmonary veins.

    Alveolar capillary dysplasia with misalignment of pulmonary veins (ACDMPV) is a lethal lung developmental disorder caused by heterozygous point mutations or genomic deletion copy-number variants (CNVs) of FOXF1 or its upstream enhancer involving fetal lung-expressed long noncoding RNA genes LINC01081 and LINC01082. Using custom-designed array comparative genomic hybridization, Sanger sequencing, whole exome sequencing (WES), and bioinformatic analyses, we studied 22 new unrelated families (20 postnatal and two prenatal) with clinically diagnosed ACDMPV. We describe novel deletion CNVs at the FOXF1 locus in 13 unrelated ACDMPV patients. Together with the previously reported cases, all 31 genomic deletions in 16q24.1, pathogenic for ACDMPV, for which parental origin was determined, arose de novo with 30 of them occurring on the maternally inherited chromosome 16, strongly implicating genomic imprinting of the FOXF1 locus in human lungs. Surprisingly, we have also identified four ACDMPV families with the pathogenic variants in the FOXF1 locus that arose on paternal chromosome 16. Interestingly, a combination of the severe cardiac defects, including hypoplastic left heart, and single umbilical artery were observed only in children with deletion CNVs involving FOXF1 and its upstream enhancer. Our data demonstrate that genomic imprinting at 16q24.1 plays an important role in variable ACDMPV manifestation likely through long-range regulation of FOXF1 expression, and may be also responsible for key phenotypic features of maternal uniparental disomy 16. Moreover, in one family, WES revealed a de novo missense variant in ESRP1, potentially implicating FGF signaling in the etiology of ACDMPV

    MADS box genes expressed in developing inflorescences of rice and sorghum

    With the aim of elucidating the complex genetic system controlling flower morphogenesis in cereals, we have characterized two rice and two sorghum MADS box genes isolated from cDNA libraries made from developing inflorescences. The rice clones OsMADS24 and OsMADS45, which share high homology with the Arabidopsis AGL2 and AGL4 MADS box genes, are expressed in the floral meristem, in all the primordia, and in mature floral organs. High expression levels have also been found in developing kernels. The sorghum clone SbMADS1 is also homologous to AGL2 and AGL4: expression analysis and mapping data suggest that it is the ortholog of OsMADS24. The pattern of expression of SbMADS2, the other sorghum MADS box gene, suggests that it may play a role as a meristem identity gene, as does AP1 in Arabidopsis, to which it shows considerable homology. The four genes have been mapped on a rice RFLP genetic map: the results are discussed in terms of synteny among cereals
