Conduction in jammed systems of tetrahedra
Control of transport processes in composite microstructures is critical to
the development of high performance functional materials for a variety of
energy storage applications. The fundamental process of conduction and its
control through the manipulation of granular composite attributes (e.g., grain
shape) are the subject of this work. We show that athermally jammed packings of
tetrahedra with ultra-short range order exhibit fundamentally different
pathways for conduction than those in dense sphere packings. Highly resistive
granular constrictions and few face-face contacts between grains result in
short-range distortions from the mean temperature field. As a consequence,
'granular' or differential effective medium theory predicts the conductivity of
this media within 10% at the jamming point; in contrast, strong enhancement of
transport near interparticle contacts in packed-sphere composites results in
conductivity divergence at the jamming onset. The results are expected to be
particularly relevant to the development of nanomaterials, where nanoparticle
building blocks can exhibit a variety of faceted shapes.Comment: 9 pages, 10 figure
Variable-cell method for stress-controlled jamming of athermal, frictionless grains
A new method is introduced to simulate jamming of polyhedral grains under
controlled stress that incorporates global degrees of freedom through the
metric tensor of a periodic cell containing grains. Jamming under
hydrostatic/isotropic stress and athermal conditions leads to a precise
definition of the ideal jamming point at zero shear stress. The structures of
tetrahedra jammed hydrostatically exhibit less translational order and lower
jamming-point density than previously described `maximally random jammed' hard
tetrahedra. Under the same conditions, cubes jam with negligible nematic order.
Grains with octahedral symmetry jam in the large-system limit with an abundance
of face-face contacts in the absence of nematic order. For sufficiently large
face-face contact number, percolating clusters form that span the entire
simulation box. The response of hydrostatically jammed tetrahedra and cubes to
shear-stress perturbation is also demonstrated with the variable-cell method.Comment: 10 pages, 8 figure
Isostaticity of Constraints in Jammed Systems of Soft Frictionless Platonic Solids
The average number of constraints per particle in
mechanically stable systems of Platonic solids (except cubes) approaches the
isostatic limit at the jamming point (), though
average number of contacts are hypostatic. By introducing angular alignment
metrics to classify the degree of constraint imposed by each contact,
constraints are shown to arise as a direct result of local orientational order
reflected in edge-face and face-face alignment angle distributions. With

Investigating Habits: Strategies,Technologies and Models
Understanding habits at a biological level requires a combination of behavioral observations and measures of ongoing neural activity. Theoretical frameworks as well as definitions of habitual behaviors emerging from classic behavioral research have been enriched by new approaches taking account of the identification of brain regions and circuits related to habitual behavior. Together, this combination of experimental and theoretical work has provided key insights into how brain circuits underlying action-learning and action-selection are organized, and how a balance between behavioral flexibility and fixity is achieved. New methods to monitor and manipulate neural activity in real time are allowing us to have a first look under the hood of a habit as it is formed and expressed. Here we discuss ideas emerging from such approaches. We pay special attention to the unexpected findings that have arisen from our own experiments suggesting that habitual behaviors likely require the simultaneous activity of multiple distinct components, or operators, seen as responsible for the contrasting dynamics of neural activity in both cortico-limbic and sensorimotor circuits recorded concurrently during different stages of habit learning. The neural dynamics identified thus far do not fully meet expectations derived from traditional models of the structure of habits, and the behavioral measures of habits that we have made also are not fully aligned with these models. Trypanosoma brucei PRMT1 Is a Nucleic Acid Binding Protein with a Role in Energy Metabolism and the Starvation Stress Response.
In Trypanosoma brucei and related kinetoplastid parasites, transcription of protein coding genes is largely unregulated. Rather, mRNA binding proteins, which impact processes such as transcript stability and translation efficiency, are the predominant regulators of gene expression. Arginine methylation is a posttranslational modification that preferentially targets RNA binding proteins and is, therefore, likely to have a substantial impact on T. brucei biology. The data presented here demonstrate that cells depleted of T. brucei PRMT1 (TbPRMT1), a major type I protein arginine methyltransferase, exhibit decreased virulence in an animal model. To understand the basis of this phenotype, quantitative global proteomics was employed to measure protein steady-state levels in cells lacking TbPRMT1. The approach revealed striking changes in proteins involved in energy metabolism. Most prominent were a decrease in glycolytic enzyme abundance and an increase in proline degradation pathway components, changes that resemble the metabolic remodeling that occurs during T. brucei life cycle progression. The work describes several RNA binding proteins whose association with mRNA was altered in TbPRMT1-depleted cells, and a large number of TbPRMT1-interacting proteins, thereby highlighting potential TbPRMT1 substrates. Many proteins involved in the T. brucei starvation stress response were found to interact with TbPRMT1, prompting analysis of the response of TbPRMT1-depleted cells to nutrient deprivation. Indeed, depletion of TbPRMT1 strongly hinders the ability of T. brucei to form cytoplasmic mRNA granules under starvation conditions. Finally, this work shows that TbPRMT1 itself binds nucleic acids in vitro and in vivo, a feature completely novel to protein arginine methyltransferases.IMPORTANCETrypanosoma brucei infection causes human African trypanosomiasis, also known as sleeping sickness, a disease with a nearly 100% fatality rate when untreated. Current drugs are expensive, toxic, and highly impractical to administer, prompting the community to explore various unique aspects of T. brucei biology in search of better treatments. In this study, we identified the protein arginine methyltransferase (PRMT), TbPRMT1, as a factor that modulates numerous aspects of T. brucei biology. These include glycolysis and life cycle progression signaling, both of which are being intensely researched toward identification of potential drug targets. Our data will aid research in those fields. Furthermore, we demonstrate for the first time a direct association of a PRMT with nucleic acids, a finding we believe could translate to other organisms, including humans, thereby impacting research in fields as distant as human cancer biology and immune response modulation. In circuits engaging the dorsomedial striatum, sensitivity to goal value is accentuated during learning, whereas outcome sensitivity is thought to be minimal in the dorsolateral striatum and its habit-related corticostriatal circuits. However, a distinct population of projection neurons in the dorsolateral striatum exhibits selective sensitivity to rewards. Here, we evaluated the outcome-related signaling in such neurons as rats performed an instructional T-maze task for two rewards. As the rats formed mazerunning habits and then changed behavior after reward devaluation, we detected outcome-related spike activity in 116 units out of 1,479 recorded units. During initial training, nearly equal numbers of these units fired preferentially either after rewarded runs or after unrewarded runs, and the majority were responsive at only one of two reward locations. With overtraining, as habits formed, firing in nonrewarded trials almost disappeared, and reward-specific firing declined. Thus error-related signaling was lost, and reward signaling became generalized. Following reward devaluation, in an extinction test, postgoal activity was nearly undetectable, despite accurate running. Strikingly, when rewards were then returned, postgoal activity reappeared and recapitulated the original early response pattern, with nearly equal numbers responding to rewarded and unrewarded runs and to single rewards. These findings demonstrate that outcome evaluation in the dorsolateral striatum is highly plastic and tracks stages of behavioral exploration and exploitation. These signals could be a new target for understanding compulsive behaviors that involve changes to dorsal striatum function
Models for Metal Hydride Particle Shape, Packing, and Heat Transfer
A multiphysics modeling approach for heat conduction in metal hydride powders
is presented, including particle shape distribution, size distribution,
granular packing structure, and effective thermal conductivity. A statistical
geometric model is presented that replicates features of particle size and
shape distributions observed experimentally that result from cyclic hydride
decreptitation. The quasi-static dense packing of a sample set of these
particles is simulated via energy-based structural optimization methods. These
particles jam (i.e., solidify) at a density (solid volume fraction) of
0.665+/-0.015 - higher than prior experimental estimates. Effective thermal
conductivity of the jammed system is simulated and found to follow the behavior
predicted by granular effective medium theory. Finally, a theory is presented
that links the properties of bi-porous cohesive powders to the present systems
based on recent experimental observations of jammed packings of fine powder.
This theory produces quantitative experimental agreement with metal hydride
