A Reliable Ex Vivo Invasion Assay Of Human Reticulocytes By Plasmodium Vivax
Currently, there are no reliable RBC invasion assays to guide the discovery of vaccines against Plasmodium vivax, the most prevalent malaria parasite in Asia and South America. Here we describe a protocol for an ex vivo P vivax invasion assay that can be easily deployed in laboratories located in endemic countries. The assay is based on mixing enriched cord blood reticulocytes with matured, trypsin-treated P vivax schizonts concentrated from clinical isolates. The reliability of this assay was demonstrated using a large panel of P vivax isolates freshly collected from patients in Thailand. © 2011 by The American Society of Hematology.11813e74e81Guerra, C.A., Howes, R.E., Patil, A.P., The international limits and population at risk of Plasmodium vivax transmission in 2009 (2010) PLoS Negl Trop Dis., 4 (8), pp. e774Baird, J.K., Resistance to therapies for infection by Plasmodium vivax (2009) Clin Microbiol Rev., 22 (3), pp. 508-534Baird, J.K., Neglect of Plasmodium vivax malaria (2007) Trends in Parasitology, 23 (11), pp. 533-539. , DOI 10.1016/j.pt.2007.08.011, PII S1471492207002425Price, R.N., Tjitra, E., Guerra, C.A., Yeung, S., White, N.J., Anstey, N.M., Vivax malaria: Neglected and not benign (2007) Am J Trop Med Hyg., 77 (6 SUPPL.), pp. 79-87Beeson, J.G., Crabb, B.S., Towards a vaccine against Plasmodium vivax malaria (2007) PLoS Med., 4 (12), pp. e350Chitnis, C.E., Sharma, A., Targeting the Plasmodium vivax Duffy-binding protein (2008) Trends Parasitol., 24 (1), pp. 29-34Miller, L.H., Mason, S.J., Clyde, D.F., McGinniss, M.H., The resistance factor to Plasmodium vivax in blacks.
