    Acetylsalicylic acid reduces the severity of dextran sodium sulfate-induced colitis and increases the formation of anti-inflammatory lipid mediators

    The role of non-steroidal anti-inflammatory drugs in inflammatory bowel disease is controversial, as they have been implicated in disease aggravation. Different from other cyclooxygenase inhibitors, acetylsalicylic acid (ASA) enhances the formation of anti-inflammatory and proresolution lipoxins derived from arachidonic acid as well as resolvins from omega-3 polyunsaturated fatty acids such as docosahexaenoic acid (DHA). In this study, we examined the effect of ASA on murine dextran sodium sulfate colitis. A mouse magnetic resonance imaging (MRI) protocol and post mortem assessment were used to assess disease severity, and lipid metabolites were measured using liquid chromatography-coupled tandem mass spectrometry. Decreased colitis activity was demonstrated by phenotype and MRI assessment in mice treated with ASA, and confirmed in postmortem analysis. Analysis of lipid mediators showed sustained formation of lipoxin A4 and an increase of DHA-derived 17-hydroxydocosahexaenoic acid (17-HDHA) after treatment with ASA. Furthermore, in vitro experiments in RAW264.7 murine macrophages demonstrated significantly increased phagocytosis activity after incubation with 17-HDHA, supporting its proresolution effect. These results show a protective effect of ASA in a murine colitis model and could give a rationale for a careful reassessment of ASA therapy in patients with inflammatory bowel disease and particularly ulcerative colitis, possibly combined with DHA supplementation

    High Quality, Low Cost Egg Incubator for BIC Church in Choma, Zambia

    The Egg Incubator team is partnering with the Brethren in Christ Church located in Choma, Zambia to design a high-quality, low-cost chicken egg incubator to supply the pastors and church members with a means of food and income. The design will need to take into account the accessibility and cost of the tools and materials. The current prototype features separate heating and humidity systems, a control system to maintain a set temperature and humidity, and tilting egg racks. The heating system consists of two stovetop coils to produce heat and a fan to transfer it to the air. The humidifier utilizes an atomizer in a pan of water to create a mist that mixes with the hot air to create humidity. The control system uses a proportional integral derivative controller (PID) to keep the temperature at 37 ± 1 °C and the humidity at 60–70%. The egg racks are tilted by a motor that runs every 6 hours to prevent the embryos from sticking to the shell. With a fully functioning prototype, the team has begun to incubate 60 real fertilized eggs. During the 21-day incubation process, a final prototype iteration is being designed and will be built on-site in Zambia in May 2022.     Evaluation of a method for enhancing interaural level differences at low frequencies.

    A method (called binaural enhancement) for enhancing interaural level differences at low frequencies, based on estimates of interaural time differences, was developed and evaluated. Five conditions were compared, all using simulated hearing-aid processing: (1) Linear amplification with frequency-response shaping; (2) binaural enhancement combined with linear amplification and frequency-response shaping; (3) slow-acting four-channel amplitude compression with independent compression at the two ears (AGC4CH); (4) binaural enhancement combined with four-channel compression (BE-AGC4CH); and (5) four-channel compression but with the compression gains synchronized across ears. Ten hearing-impaired listeners were tested, and gains and compression ratios for each listener were set to match targets prescribed by the CAM2 fitting method. Stimuli were presented via headphones, using virtualization methods to simulate listening in a moderately reverberant room. The intelligibility of speech at ±60° azimuth in the presence of competing speech on the opposite side of the head at ±60° azimuth was not affected by the binaural enhancement processing. Sound localization was significantly better for condition BE-AGC4CH than for condition AGC4CH for a sentence, but not for broadband noise, lowpass noise, or lowpass amplitude-modulated noise. The results suggest that the binaural enhancement processing can improve localization for sounds with distinct envelope fluctuations

    Neurexins and Neuroligins: Recent Insights from Invertebrates

    During brain development, each neuron must find and synapse with the correct pre- and postsynaptic partners. The complexity of these connections and the relatively large distances some neurons must send their axons to find the correct partners makes studying brain development one of the most challenging, and yet fascinating disciplines in biology. Furthermore, once the initial connections have been made, the neurons constantly remodel their dendritic and axonal arbours in response to changing demands. Neurexin and neuroligin are two cell adhesion molecules identified as important regulators of this process. The importance of these genes in the development and modulation of synaptic connectivity is emphasised by the observation that mutations in these genes in humans have been associated with cognitive disorders such as Autism spectrum disorders, Tourette syndrome and Schizophrenia. The present review will discuss recent advances in our understanding of the role of these genes in synaptic development and modulation, and in particular, we will focus on recent work in invertebrate models, and how these results relate to studies in mammals

    Spatial Audiometry: Detection of Spondaic Words in Noise.

    It is important to select appropriate stimuli and test conditions for developing standardized spatial audiometric tests. In three experiments, binaural detection thresholds (BDTs) for a target signal, located at either 0, 45, 90, 135, 180, 225, 270, or 315 degrees azimuth, were measured in the presence of a masker positioned at one of these eight locations. Target signals included spondaic words from the CID W-1 list. The masker was speech spectrum noise (SSN) or multitalker noise (MTN) presented at a constant level (65 dBA). Bekesy tracking was used to measure BDTs in listeners with normal hearing. Results indicate that BDTs are significantly influenced by the (a) angular separation between the target and noise source and (b) choice of spondaic words used as target stimuli. BDTs for various spondaic words differed as much as 13 dB for a given angular separation. BDTs measured in SSN and MTN for otherwise identical test conditions differed less than 3 dB. A single spondaic word appears to be appropriate for spatial audiometric tests of detection. Nonsignificant differences between masked BDTs obtained for SSN and MTN noises indicate that for spatial detection, the masking effects of these noises are comparable. These results indicate that the development of a clinical test of spatial detection should include the use of SSN and a single spondaic word, with detection being measured for a set of four or five signal and noise source configurations
