    Multi-drug therapy for epilepsy influenced bispectral index after a bolus propofol administration without affecting propofol's pharmacokinetics: a prospective cohort study

    Some previous studies have indicated that valproate (VPA) might change the pharmacokinetics and enhance the effects of propofol. We evaluated whether clinical VPA therapy affected the propofol blood level, the protein-unbound free propofol level, and/or the anesthetic effects of propofol in the clinical setting. The subjects were divided into the control group (not medicated with antiepileptics), the mono-VPA group (medicated with VPA alone), and the poly-VPA group (medicated with VPA, other antiepileptics, and/or psychoactive drugs). General anesthesia was induced via the administration of a single bolus of propofol and a remifentanil infusion, and when the bispectral index (BIS) exceeded 60 sevoflurane was started. There were no significant differences in the total blood propofol level at 5, 10, 15, and 20 min or the protein-unbound free propofol level at 5 min after the intravenous administration of propofol between the 3 groups. However, the minimum BIS was significantly lower and the time until the BIS exceeded 60 was significantly longer in the poly-VPA group. In the multivariate regression analysis, belonging to the poly-VPA group was found to be independently associated with the minimum BIS value and the time until the BIS exceeded 60. Clinical VPA therapy did not influence the pharmacokinetics of propofol. However, multi-drug therapy involving VPA might enhance the anesthetic effects of propofol

    Novel Soft Meals Developed by 3D Printing

    Recently, 3D printing is being applied to various fields. 3D printing of foods has been developed; however, there are many challenges. To overcome the challenges, we have started a new research group named “Yonezawa Itadakimasu Research Group,” to focus on the development of 3D printing applications for manufacturing food. We have developed Novel jelly foods that are shaped by 3D printed molds. Fused deposition modeling (FDM) 3D printer for food manufacturing makes the 3D printed molds. First step of making 3D printing mold is to print a cast. Then, food grade silicone is poured into the cast to make a mold. This type of 3D printed mold can be used widely, such as making sweets, restaurant menus, and care foods by changing the design depending on the use of application. Secondly, we started to develop 3D food printers. This type of challenge to develop future foods by 3D printing technology may have a major impact on the care food because the looks of foods are important and will be improved by 3D printing

    本稿では、バランス・スポーツ“スラックライン”を初年次教育の題材として用いた事例的研究について報告する。スラックラインとは、綱渡りを競技として発展させたものであり、現在、スポーツやレジャー、身体教育や健康促進、リハビリテーションの一環としてなど、様々な分野・用途で応用されている。本研究では、誰でも気軽に楽しめ、省スペースで実施でき、コミュニケーションのツールともなるスラックラインを、初年次教育における仮説検証学習の題材として応用し、学生らが自らの身体を使って体験的に学ぶことで、学習が促進されるかどうかについて検討した。授業後のアンケートの結果、スラックラインを取り入れた授業によって、自身や他者についての理解や気づきが促され、コミュニケーションが活性化し、楽しい学習の場が提供できたことが示唆された。これらの結果は、スラックラインが初年次教育の題材として有効である可能性を部分的に示している。論

    Helicobacter pylori infection and gastroduodenal diseases in Vietnam: a cross-sectional, hospital-based study

    <p>Abstract</p> <p>Background</p> <p>The rate of <it>H. pylori </it>infection in Vietnam is reportedly high, but the spectrum of <it>H. pylori</it>-associated gastroduodenal diseases has not been systematically investigated. Moreover, despite the similarities of ethnicity and diet, the age-standardized incidence rate of gastric cancer in the northern city of Hanoi is higher than that in the southern city of Ho Chi Minh, but the reason for this phenomenon is unknown. The virulence of Vietnamese <it>H. pylori </it>has also not been investigated in detail.</p> <p>Methods</p> <p>Individuals undergoing esophagogastroduodenoscopy were randomly recruited. <it>H. pylori </it>infection status was determined based on the combined results of culture, histology, immunohistochemistry, rapid urine test and serum ELISA. Peptic ulcer (PU) and gastroesophageal reflux disease was diagnosed by endoscopy, and chronic gastritis was determined histologically. <it>H. pylori </it>virulence factors were investigated by PCR and sequencing.</p> <p>Results</p> <p>Among the examined patients, 65.6% were infected with <it>H. pylori</it>. The prevalence of infection was significantly higher in those over 40 years of age than in those aged ≤40. Chronic gastritis was present in all <it>H. pylori</it>-infected individuals, 83.1% of whom had active gastritis, and 85.3% and 14.7% had atrophy and intestinal metaplasia, respectively. PU was present in 21% of infected patients, whereas its incidence was very low in non-infected individuals. The prevalence of PU was significantly higher in Hanoi than in Ho Chi Minh. The prevalence of <it>vacA m1</it>, which has been identified as an independent risk factor for PU in Vietnam, was significantly higher among <it>H. pylori </it>isolates from Hanoi than among those from Ho Chi Minh.</p> <p>Conclusions</p> <p><it>H. pylori </it>infection is common in Vietnam and is strongly associated with PU, active gastritis, atrophy and intestinal metaplasia. <it>vacA m1 </it>is associated with an increased risk for PU and might contribute to the difference in the prevalence of PU and gastric cancer between Hanoi and Ho Chi Minh.</p

    The whole blood transcriptional regulation landscape in 465 COVID-19 infected samples from Japan COVID-19 Task Force

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

    O presente trabalho é uma pesquisa sobre a timidez a partir de uma perspectiva estética, o Ma, a fim de desvendar entendimentos mais sensíveis sobre o assunto. Na escola, a timidez é normalmente tratada de forma extremamente negativa, relacionada a patologização e adestramento dos sujeitos que se entendem como tímidos. Por isso, a visão de educação sensível, que preza por um saber mais pleno e íntegro, representa uma importante alternativa a esta categorização. Assim, é no ramo da estética, de estudo da beleza, do sublime, da arte e sensibilidade geral do ser humano, que tal exploração se desenrola. Considerando o processo criativo pessoal reflito sobre a importância de compreender as emoções na formação humana tendo como princípio o respeito à singularidade e à diversidade. A abordagem do trabalho parte dos fundamentos da Arte-educação, em especial da Estética Oriental do “entre”, a ênfase não é na Psicologia, mas considera-se as contribuições de Henry Wallon e sua teoria do desenvolvimento humano.The present work is a research on shyness from an aesthetic perspective, the Ma, in order to unveil more sensitive understandings on the subject. At school, shyness is normally treated in an extremely negative way, related to the pathologization and training of subjects who perceive themselves as shy. Therefore, the vision of sensitive education, which values fuller and more complete knowledge, represents an important alternative to this categorization. Thus, it is in the field of aesthetics, the study of beauty, the sublime, art and the general sensibility of human beings, that such an exploration takes place. Considering the personal creative process, I reflect on the importance of understanding emotions in human development, having as a principle the respect for uniqueness and diversity. The approach of the work is based on the foundations of Art Education, in particular the Oriental Aesthetics of the “between”, the emphasis is not on Psychology, but on the contributions of Henry Wallon and his theory of human development.Não recebi financiament
