    Soil quality indicators in a rhodic kandiudult under different uses in northern Parana, Brazil.

    Sustainable use of soil, maintaining or improving its quality, is one of the goals of diversification in farmlands. From this point of view, bioindicators associated with C, N and P cycling can be used in assessments of land-use effects on soil quality. The aim of this study was to investigate chemical, microbiological and biochemical properties of soil associated with C, N and P under different land uses in a farm property with diversified activity in northern Parana, Brazil. Seven areas under different land uses were assessed: fragment of native Atlantic Forest; growing of peach-palm (Bactrys gasipaes); sugarcane ratoon (Saccharum officinarum) recently harvested, under renewal; growing of coffee (Coffea arabica) intercropped with tree species; recent reforestation (1 year) with native tree species, previously under annual crops; annual crops under no-tillage, rye (Cecale cereale); secondary forest, regenerated after abandonment (for 20 years) of an avocado (Persea americana) orchard. The soil under coffee, recent reforestation and secondary forest showed higher concentrations of organic carbon, but microbial biomass and enzyme activities were higher in soils under native forest and secondary forest, which also showed the lowest metabolic coefficient, followed by the peach-palm area. The lowest content of water-dispersible clay was found in the soil under native forest, differing from soils under sugarcane and secondary forest. Soil cover and soil use affected total organic C contents and soil enzyme and microbial activities, such that more intensive agricultural uses had deeper impacts on the indicators assessed. Calculation of the mean soil quality index showed that the secondary forest was closest to the fragment of native forest, followed by the peach-palm area, coffee-growing area, annual crop area, the area of recent reforestation and the sugarcane ratoon area

    Purification and preliminary characterization of a xylanase from Thermomyces lanuginosus strain SS-8

    Thermomyces lanuginosus SS-8 was isolated from soil samples that had been collected from near self-heating plant material and its extracellular cellulase-free xylanase purified approximately 160-fold using ion exchange chromatography and continuous elution electrophoresis. This xylanase was thermoactive (optimum temperature 60 °C) at pH 6.0 and had a molecular weight of 23.79 kDa as indicated by SDS-PAGE electrophoresis. The xylanase rapidly hydrolyzed xylan directly to xylose without the production of intermediary xylo-oligosaccharides within 15 min of incubation under optimum conditions. This trait of rapidly degrading xylan to xylose as a sole end-product could have biotechnological potential in degradation of agro-wastes for bioethanol manufacturing industry

    A novel xylan degrading β-D-xylosidase: purification and biochemical characterization

    Aspergillus ochraceus, a thermotolerant fungus isolated in Brazil from decomposing materials, produced an extracellular b-xylosidase that was purified using DEAE-cellulose ion exchange chromatography, Sephadex G-100 and Biogel P-60 gel filtration. b-xylosidase is a glycoprotein (39 % carbohydrate content) and has a molecular mass of 137 kDa by SDS-PAGE, with optimal temperature and pH at 70 C and 3.0–5.5, respectively.b-xylosidase was stable in acidic pH (3.0–6.0) and 70 C for 1 h. The enzyme was activated by 5 mM MnCl2 (28 %)and MgCl2 (20 %) salts. The b-xylosidase produced by A. ochraceus preferentially hydrolyzed p-nitrophenyl-b- D-xylopyranoside, exhibiting apparent Km and Vmax values of 0.66 mM and 39 U (mg protein)-1 respectively, and to a lesser extent p-nitrophenyl-b-D-glucopyranoside. The enzyme was able to hydrolyze xylan from different sources,suggesting a novel b-D-xylosidase that degrades xylan. HPLC analysis revealed xylans of different compositions which allowed explaining the differences in specificity observed by b-xylosidase.     Strong protective effect of the APOL1 p.N264K variant against G2-associated focal segmental glomerulosclerosis and kidney disease

    African Americans have a significantly higher risk of developing chronic kidney disease, especially focal segmental glomerulosclerosis -, than European Americans. Two coding variants (G1 and G2) in the APOL1 gene play a major role in this disparity. While 13% of African Americans carry the high-risk recessive genotypes, only a fraction of these individuals develops FSGS or kidney failure, indicating the involvement of additional disease modifiers. Here, we show that the presence of the APOL1 p.N264K missense variant, when co-inherited with the G2 APOL1 risk allele, substantially reduces the penetrance of the G1G2 and G2G2 high-risk genotypes by rendering these genotypes low-risk. These results align with prior functional evidence showing that the p.N264K variant reduces the toxicity of the APOL1 high-risk alleles. These findings have important implications for our understanding of the mechanisms of APOL1-associated nephropathy, as well as for the clinical management of individuals with high-risk genotypes that include the G2 allele

    Targeted metatranscriptomics of compost derived consortia reveals a GH11 exerting an unusual exo-1,4-β-xylanase activity

    Background: Using globally abundant crop residues as a carbon source for energy generation and renewable chemicals production stands out as a promising solution to reduce current dependency on fossil fuels. In nature, such as in compost habitats, microbial communities efficiently degrade the available plant biomass using a diverse set of synergistic enzymes. However, deconstruction of lignocellulose remains a challenge for industry due to recalcitrant nature of the substrate and the inefficiency of the enzyme systems available, making the economic production of lignocellulosic biofuels difficult. Metatranscriptomic studies of microbial communities can unveil the metabolic functions employed by lignocellulolytic consortia and identify new biocatalysts that could improve industrial lignocellulose conversion. Results: In this study, a microbial community from compost was grown in minimal medium with sugarcane bagasse sugarcane bagasse as the sole carbon source. Solid-state nuclear magnetic resonance was used to monitor lignocellulose degradation; analysis of metatranscriptomic data led to the selection and functional characterization of several target genes, revealing the first glycoside hydrolase from Carbohydrate Active Enzyme family 11 with exo-1,4-β-xylanase activity. The xylanase crystal structure was resolved at 1.76 Å revealing the structural basis of exo-xylanase activity. Supplementation of a commercial cellulolytic enzyme cocktail with the xylanase showed improvement in Avicel hydrolysis in the presence of inhibitory xylooligomers. Conclusions: This study demonstrated that composting microbiomes continue to be an excellent source of biotechnologically important enzymes by unveiling the diversity of enzymes involved in in situ lignocellulose degradation

    Etude des relations statistiques entre deux signaux temporels par analyse interspectrale

    La recherche des liaisons statistiques entre deux signaux temporels est un problème très important en physique de la mesure. Les liaisons entre deux signaux peuvent être de nature très diverse. Nous nous limiterons ici à la caractérisation de liaisons linéaires et stationnaires entre deux signaux temporels appréhendés à travers une réalisation particulière de durée finie de chacun d'entre eux. Nous montrons tout d'abord que, dans la situation décrite ci-dessus et dans le cas de signaux localement blancs, la méthode optimale de recherche et de caractérisation des dépendances linéaires est une analyse interspeotrale s'appuyant sur la mesure des densités spectrales de puissance et de la densité spectrale de puissance croisée. Nous menons ensuite l'étude complète de la méthode de mesure de la matrice interspeotrale dans le cas d'un dispositif réalisant la transformée de FOURIER des fonctions d'autoet d'intercorrélation et nous discutons les propriétés au second ordre de l'estimation de la matrice interspeotrale ainsi obtenue. Nous étudions également les problèmes posés par la quantification des signaux dans le corrélateur. Enfin, nous présentons les résultats obtenus dans un cas pratique d'étude de signaux magnétiques naturels de la gamme de Hertz captés à 1 000 km de distance

    Influência da sanificação da água e das práticas de ordenha na qualidade do leite

    RESUMO Objetivou-se avaliar a influência da cloração da água utilizada em salas de ordenha, assim como do manejo e da infraestrutura da ordenha, sobre a qualidade microbiológica da água e do leite. Foi instalado um equipamento para cloração de água, por duas semanas, na caixa de água de 20 propriedades leiteiras. Foram coletadas amostras de água e leite ao primeiro dia (sem cloro: controle), no sétimo e 14° dias (com cloro) e no 21º dia após a desinstalação dos cloradores (sem cloro: controle). Foram realizadas análises microbiológicas da água e do leite (contagem de células somáticas do leite, bactérias psicotróficas, mesófilas e coliformes totais), análises físico-químicas da água (pH, dureza e matéria orgânica), e aplicou-se um questionário estruturado aos produtores visando conhecer as técnicas de manejo de ordenha adotadas na propriedade. O uso de cloração na água melhorou (P0,05). Práticas adequadas de manejo e higiene de ordenha e adequada estrutura para a ordenha estão relacionadas a baixas contagens de microrganismos no leite. Conclui-se que a cloração melhora a qualidade microbiológica da água, sem afetar a qualidade microbiológica do leite, a qual é melhorada pela adoção de boas práticas de ordenha e adequada infraestrutura
