    Spatial configuration, spatial cognition and spatial behaviour: the role of architectural intelligibility in shaping spatial experience

    This thesis investigates the role of spatial configuration in shaping resident's experience of their neighbourhood. Studies to date have found that spatial configuration affects spatial behaviour and movement patterns (e.g., Hillier et al, 1993), however there has been little investigation of the cognitive processes that might underlie this relationship. Other research into cognition of the urban environment suggests that local spatial factors may play a role in cognitive processes (e.g., Hart & Moor, 1973), however these studies have not addressed global spatial configuration in quantitative terms. No studies to date have sought to integrate cognitive, behavioural and configurational factors within a single framework. Using Hillier's (1996) definition of intelligibility as the relationship between local and global configurational factors, this thesis investigates the relationship between resident's cognition, observed patterns of movement and the spatial configuration of an area. Two adjacent areas in Hampstead Garden Suburb in North London were investigated in detail. One area is relatively intelligible, the other less so. Structured interview surveys were carried out with local residents to elicit aspects of their cognition of the local area and detailed observations were made of movement patterns in the two neighbourhoods. Analysis of the spatial characteristics of the two areas using 'space syntax' methods provided a common basis for analyses of these data. The findings confirm that spatial configuration, spatial cognition and space use patterns are all related to one another. The main finding is that the degree of intelligibility of the area is the most significant intervening variable in relations between the three variables. The more intelligible area showed more powerful correlations between spatial configuration and patterns of movement, as well as giving rise to perceptions of greater legibility and increased neighbourhood size by local residents. Strong correlations were also identified between residents' cognitive maps and observed patterns of movement in the area. The correlations were again found to be stronger in the intelligible area than the unintelligible area. These findings suggest that spatial configuration may play an important role in determining people's daily spatial experience by increasing or reducing their sense of autonomy. By reducing the ability to predict either one's precise location within his/her global context, or the likely behaviour of others in space, unintelligible urban configurations may result in perceptions of a lesser sense of personal control over one's own actions in the environment. The thesis concludes that 'architectural intelligibility' may be a basic aspect in achieving human spatial needs

    The antifibrotic potential of a sustained release formulation of a PDGFβ-receptor targeted rho kinase inhibitor

    Rho kinase activity in hepatic stellate cells (HSCs) is associated with activation, transformation and contraction of these cells, leading to extracellular matrix production and portal hypertension in liver cirrhosis. Inhibition of rho kinase activity can reduce these activities, but may also lead to side effects, for instance systemic hypotension. This can be circumvented by liver-specific delivery of a rho kinase inhibitor to effector cells. Therefore, we targeted the rho kinase inhibitor Y27632 to the key pathogenic cells in liver fibrosis, i.e. myofibroblasts including activated HSCs that highly express the PDGF beta-receptor, using the drug carrier pPB-MSA. This carrier consists of mouse serum albumin (MSA) covalently coupled to several PDGF beta-recognizing moieties (pPB). We aimed to create a prolonged release system of such a targeted construct, by encapsulating pPB-MSA-Y27632 in biodegradable polymeric microspheres, thereby reducing short-lasting peak concentrations and the need for frequent administrations. Firstly, we confirmed the vasodilating potency of PDGF beta-receptor targeted Y27632 in vitro in a contraction assay using HSCs seeded on a collagen gel. We subsequently demonstrated the in vivo antifibrotic efficacy of pPB-MSA-Y27632-loaded microspheres in the Mdr2 - / - mouse model of progressive biliary liver fibrosis. A single subcutaneous microsphere administration followed by organ harvest one week later clearly attenuated liver fibrosis progression and significantly suppressed the expression of fibrosis related genes, such as several collagens, profibrotic cytokines and matrix metalloproteinases. In conclusion, we demonstrate that polymeric microspheres are suitable as drug delivery system for the sustained systemic delivery of targeted protein constructs with antifibrotic potential, such as pPB-MSA-Y27632. This formulation appears suitable for the sustained treatment of liver fibrosis and possibly other chronic diseases

    Standalone vertex finding in the ATLAS muon spectrometer

    A dedicated reconstruction algorithm to find decay vertices in the ATLAS muon spectrometer is presented. The algorithm searches the region just upstream of or inside the muon spectrometer volume for multi-particle vertices that originate from the decay of particles with long decay paths. The performance of the algorithm is evaluated using both a sample of simulated Higgs boson events, in which the Higgs boson decays to long-lived neutral particles that in turn decay to bbar b final states, and pp collision data at √s = 7 TeV collected with the ATLAS detector at the LHC during 2011

    Measurements of Higgs boson production and couplings in diboson final states with the ATLAS detector at the LHC

    Measurements are presented of production properties and couplings of the recently discovered Higgs boson using the decays into boson pairs, H →γ γ, H → Z Z∗ →4l and H →W W∗ →lνlν. The results are based on the complete pp collision data sample recorded by the ATLAS experiment at the CERN Large Hadron Collider at centre-of-mass energies of √s = 7 TeV and √s = 8 TeV, corresponding to an integrated luminosity of about 25 fb−1. Evidence for Higgs boson production through vector-boson fusion is reported. Results of combined fits probing Higgs boson couplings to fermions and bosons, as well as anomalous contributions to loop-induced production and decay modes, are presented. All measurements are consistent with expectations for the Standard Model Higgs boson

    Measurement of the top quark-pair production cross section with ATLAS in pp collisions at \sqrt{s}=7\TeV

    A measurement of the production cross-section for top quark pairs(\ttbar) in pppp collisions at \sqrt{s}=7 \TeV is presented using data recorded with the ATLAS detector at the Large Hadron Collider. Events are selected in two different topologies: single lepton (electron ee or muon μ\mu) with large missing transverse energy and at least four jets, and dilepton (eeee, μμ\mu\mu or eμe\mu) with large missing transverse energy and at least two jets. In a data sample of 2.9 pb-1, 37 candidate events are observed in the single-lepton topology and 9 events in the dilepton topology. The corresponding expected backgrounds from non-\ttbar Standard Model processes are estimated using data-driven methods and determined to be 12.2±3.912.2 \pm 3.9 events and 2.5±0.62.5 \pm 0.6 events, respectively. The kinematic properties of the selected events are consistent with SM \ttbar production. The inclusive top quark pair production cross-section is measured to be \sigmattbar=145 \pm 31 ^{+42}_{-27} pb where the first uncertainty is statistical and the second systematic.     Measurement of the top quark pair cross section with ATLAS in pp collisions at √s=7 TeV using final states with an electron or a muon and a hadronically decaying τ lepton

    A measurement of the cross section of top quark pair production in proton-proton collisions recorded with the ATLAS detector at the Large Hadron Collider at a centre-of-mass energy of 7 TeV is reported. The data sample used corresponds to an integrated luminosity of 2.05 fb -1. Events with an isolated electron or muon and a τ lepton decaying hadronically are used. In addition, a large missing transverse momentum and two or more energetic jets are required. At least one of the jets must be identified as originating from a b quark. The measured cross section, σtt-=186±13(stat.)±20(syst.)±7(lumi.) pb, is in good agreement with the Standard Model prediction

    Hunt for new phenomena using large jet multiplicities and missing transverse momentum with ATLAS in 4.7 fb−1 of √s=7 TeV proton-proton collisions

    Results are presented of a search for new particles decaying to large numbers of jets in association with missing transverse momentum, using 4.7 fb−1 of pp collision data at s√=7TeV collected by the ATLAS experiment at the Large Hadron Collider in 2011. The event selection requires missing transverse momentum, no isolated electrons or muons, and from ≥6 to ≥9 jets. No evidence is found for physics beyond the Standard Model. The results are interpreted in the context of a MSUGRA/CMSSM supersymmetric model, where, for large universal scalar mass m 0, gluino masses smaller than 840 GeV are excluded at the 95% confidence level, extending previously published limits. Within a simplified model containing only a gluino octet and a neutralino, gluino masses smaller than 870 GeV are similarly excluded for neutralino masses below 100 GeV

    Network and Psychological Effects in Urban Movement

    Correlations are regularly found in space syntax studies between graph-based configurational measures of street networks, represented as lines, and observed movement patterns. This suggests that topological and geometric complexity are critically involved in how people navigate urban grids. This has caused difficulties with orthodox urban modelling, since it has always been assumed that insofar as spatial factors play a role in navigation, it will be on the basis of metric distance. In spite of much experimental evidence from cognitive science that geometric and topological factors are involved in navigation, and that metric distance is unlikely to be the best criterion for navigational choices, the matter has not been convincingly resolved since no method has existed for extracting cognitive information from aggregate flows. Within the space syntax literature it has also remained unclear how far the correlations that are found with syntactic variables at the level of aggregate flows are due to cognitive factors operating at the level of individual movers, or they are simply mathematically probable network effects, that is emergent statistical effects from the structure of line networks, independent of the psychology of navigational choices. Here we suggest how both problems can be resolved, by showing three things: first, how cognitive inferences can be made from aggregate urban flow data and distinguished from network effects; second by showing that urban movement, both vehicular and pedestrian, are shaped far more by the geometrical and topological properties of the grid than by its metric properties; and third by demonstrating that the influence of these factors on movement is a cognitive, not network, effect
