    Children\u27s Friends in Ability vs. Randomly Grouped Classrooms

    For over forty years ability grouping has been of professional concern to educators in the United States (Reisner, 1936). There have been fundamental changes in educational theory and practice during that time, one of which is the recognition that educational practices must adjust to individual differences. The interpretation of equal opportunity in education has gradually changed from meaning the same methods, standards, and course content for all children, to meaning the full opportunity for each child to develop his own potential in a school program suited to his individual capacities and needs. The interpersonal relationships and social development of school children have received an increasing amount of attention as we have come to realize the pervasive effect they have on educational objectives and as optimal social adjustment has itself become one of these objectives. As Brumbaugh (1960, p. 99) has pointed out: Mental health and social adjustment are words to conjure with when there is discussion about separate grouping. A half century ago, the fear was that stigma would attach to a child in a special class for those with below average intelligence. It is now replaced by anxiety lest those at the other end of the scale would have feelings of superiority and become egotistical little snobs. There are enough studies of children in such classes to indicate that this does not happen but there is also some evidence that there are concomitant effects which are used to oppose ability grouping on a social segregation argument. Taba et al. (1952) as an example of this point of view write: Of special interest for intergroup education is the fact that the static single bases for grouping have almost always fixed homogeneity simultaneously along lines of socioeconomic status, race, and religious background. For example, any type of ability grouping also inadervertently introduces segregation by economic class, race, and neighborhood. Because of their cultural handicaps, children from deviant backgrounds tend to be at the bottom of the heap, as far as school achievement is concerned. Hence, in ability grouping, they are thrown together and separated from other children.... This segregation, of course, prevents learning common culture by association with other children. The stigma attached to the lower ability groups further destroys motivation and self-respect, Thus, a basis is built for both physical and psychological isolation. (pp. 138-1939

    Seasonal abundance of thrips (Thysanoptera) in capsicum and chilli crops in south-east Queensland, Australia

    Thrips can be important pests of capsicum and chilli crops, causing damage through their feeding and by vectoring viral diseases. As different species vary in their ability to transmit viruses and in their susceptibility to insecticides, it is important to know which species are present in a crop. The seasonal occurrence of thrips in capsicum and chilli crops in the Bundaberg district of south-east Queensland was investigated from July 2002 to June 2003. Fifty flowers were collected weekly from crops on seven farms and the adult thrips extracted and identified. Thrips palmi Karny and Frankliniella occidentalis (Pergande) were collected in the greatest numbers, with T. palmi predominant in autumn crops (March to July) and F. occidentalis predominant in spring crops (August to November). Pseudanaphothrips achaetus (Bagnall) was common, while Thrips tabaci Lindeman, Thrips imaginis Bagnall and Frankliniella schultzei (Trybom) were collected in low numbers

    Joint Modeling and Registration of Cell Populations in Cohorts of High-Dimensional Flow Cytometric Data

    In systems biomedicine, an experimenter encounters different potential sources of variation in data such as individual samples, multiple experimental conditions, and multi-variable network-level responses. In multiparametric cytometry, which is often used for analyzing patient samples, such issues are critical. While computational methods can identify cell populations in individual samples, without the ability to automatically match them across samples, it is difficult to compare and characterize the populations in typical experiments, such as those responding to various stimulations or distinctive of particular patients or time-points, especially when there are many samples. Joint Clustering and Matching (JCM) is a multi-level framework for simultaneous modeling and registration of populations across a cohort. JCM models every population with a robust multivariate probability distribution. Simultaneously, JCM fits a random-effects model to construct an overall batch template -- used for registering populations across samples, and classifying new samples. By tackling systems-level variation, JCM supports practical biomedical applications involving large cohorts

    Scoping review on social care economic evaluation methods

    Background: In the UK and internationally there is widespread acceptance of the value of economic evaluations to inform decisions about health care  interventions. The general methods of economic evaluation of health care interventions are now well established. By contrast, approaches to social care economic evaluation are substantially less well developed. There is considerable uncertainty and disagreement about which methods to apply, and diversity in methodological practices. This makes it hard for decision makers to interpret the findings of different studies and make comparisons of value for money between different interventions evaluated using different methods. Despite previous attempts to co-ordinate methods in this area by providing guidelines (NICE, 2013, NICE, 2014), there remains considerable methodological uncertainty. NICE commissioned this scoping review to support developing a long-term strategy for how to consider social care economics in guidelines. Aims: The project aims to inform NICE on the methods available and the methods in development for use in undertaking economic evaluation of social care interventions. A further aim of the scoping review is to assess how well these methods address current methodological priorities for NICE in social care economic evaluation, and to identify gaps that the work will not address. On this basis, another aim is to provide recommendations to NICE on work required to address identified gaps in the future. Methods: A systematic review of the published literature and a survey of experts were undertaken to identify key methods used to undertake recent economic evaluations of social care interventions. Each study was assessed in terms of the key requirements for economic evaluation. Data were extracted on: the perspective of the analysis, the interventions compared, the evidence used on costs and effects, opportunity cost, uncertainty, and equity. Expert advisors commented on the findings of the review and this informed the results that were drawn from the studies. Recommendations were made to improve the conduct and reporting of studies, and areas of further research were identified. Results: Thirty social care economic evaluations were identified for review. Findings were reported on key requirements for economic evaluation comprising: the perspective of relevance to the decision maker, an evaluation comparing all relevant alternative interventions, use of all available evidence on costs and effects of relevance to the decision, analysis of whether the benefits of an intervention were greater than the forgone benefits of displaced interventions, assessment of the uncertainty associated with the decision, and exploration of the equity implications of the decision. Conclusions: Methods guidance for the economic evaluation of social care interventions needs to reflect what is feasible given the available evidence and what is appropriate for social care. A more developed evidence base is required in order to undertake economic evaluation of social care interventions. This should include undertaking primary studies where the evidence is not sufficient. Studies based on decision models and secondary evidence should be used where there is sufficient evidence available to do so. Investment in applied economic evaluations of social care interventions will support more informed recommendations and also develop research capacity in social care. Further methodological research is required to improve the way economic evaluations are undertaken in this field. This includes: agreement on the objectives of social care and the appropriate outcome measures development of cost-effectiveness threshold in social care given the agreed outcome measures how to account for costs and benefits falling on different sectors accounting for informal care equity-informative economic evaluations of social care interventions better scoping of economic evaluations application of evidence synthesis, decision modelling and expert elicitation application of value of information methods. NICE should consider these priorities in their discussions with the MRC Methodology Research Programme, to establish whether it can commission research on some or all of these areas

    Quality of hospital care for sick newborns and severely malnourished children in Kenya: A two-year descriptive study in 8 hospitals

    BACKGROUND: Given the high mortality associated with neonatal illnesses and severe malnutrition and the development of packages of interventions that provide similar challenges for service delivery mechanisms we set out to explore how well such services are provided in Kenya. METHODS: As a sub-component of a larger study we evaluated care during surveys conducted in 8 rural district hospitals using convenience samples of case records. After baseline hospitals received either a full multifaceted intervention (intervention hospitals) or a partial intervention (control hospitals) aimed largely at improving inpatient paediatric care for malaria, pneumonia and diarrhea/dehydration. Additional data were collected to: i) examine the availability of routine information at baseline and their value for morbidity, mortality and quality of care reporting, and ii) compare the care received against national guidelines disseminated to all hospitals. RESULTS: Clinical documentation for neonatal and malnutrition admissions was often very poor at baseline with case records often entirely missing. Introducing a standard newborn admission record (NAR) form was associated with an increase in median assessment (IQR) score to 25/28 (22-27) from 2/28 (1-4) at baseline. Inadequate and incorrect prescribing of penicillin and gentamicin were common at baseline. For newborns considerable improvements in prescribing in the post baseline period were seen for penicillin but potentially serious errors persisted when prescribing gentamicin, particularly to low-birth weight newborns in the first week of life. Prescribing essential feeds appeared almost universally inadequate at baseline and showed limited improvement after guideline dissemination. CONCLUSION: Routine records are inadequate to assess newborn care and thus for monitoring newborn survival interventions. Quality of documented inpatient care for neonates and severely malnourished children is poor with limited improvement after the dissemination of clinical practice guidelines. Further research evaluating approaches to improving care for these vulnerable groups is urgently needed. We also suggest pre-service training curricula should be better aligned to help improve newborn survival particularly

    Enhanced Fusion Pore Expansion Mediated by the Trans-Acting Endodomain of the Reovirus FAST Proteins

    The reovirus fusion-associated small transmembrane (FAST) proteins are virus-encoded membrane fusion proteins that function as dedicated cell–cell fusogens. The topology of these small, single-pass membrane proteins orients the majority of the protein on the distal side of the membrane (i.e., inside the cell). We now show that ectopic expression of the endodomains of the p10, p14, and p15 FAST proteins enhances syncytiogenesis induced by the full-length FAST proteins, both homotypically and heterotypically. Results further indicate that the 68-residue cytoplasmic endodomain of the p14 FAST protein (1) is endogenously generated from full-length p14 protein expressed in virus-infected or transfected cells; (2) enhances syncytiogenesis subsequent to stable pore formation; (3) increases the syncytiogenic activity of heterologous fusion proteins, including the differentiation-dependent fusion of murine myoblasts; (4) exerts its enhancing activity from the cytosol, independent of direct interactions with either the fusogen or the membranes being fused; and (5) contains several regions with protein–protein interaction motifs that influence enhancing activity. We propose that the unique evolution of the FAST proteins as virus-encoded cellular fusogens has allowed them to generate a trans-acting, soluble endodomain peptide to harness a cellular pathway or process involved in the poorly understood process that facilitates the transition from microfusion pores to macrofusion and syncytiogenesis

    Analysis of the function of IL-10 in chickens using specific neutralising antibodies and a sensitive capture ELISA

    AbstractIn mammals, the inducible cytokine interleukin 10 is a feedback negative regulator of inflammation. To determine the extent to which this function is conserved in birds, recombinant chicken IL-10 was expressed as a secreted human Ig Fc fusion protein (chIL-10-Fc) and used to immunise mice. Five monoclonal antibodies (mAb) which specifically recognise chicken IL-10 were generated and characterised. Two capture ELISA assays were developed which detected native chIL-10 secreted from chicken bone marrow-derived macrophages (chBMMs) stimulated with lipopolysaccharide (LPS). Three of the mAbs detected intracellular IL-10. This was detected in only a subset of the same LPS-stimulated chBMMs. The ELISA assay also detected massive increases in circulating IL-10 in chickens challenged with the coccidial parasite, Eimeria tenella. The same mAbs neutralised the bioactivity of recombinant chIL-10. The role of IL-10 in feedback control was tested in vitro. The neutralising antibodies prevented IL-10-induced inhibition of IFN-γ synthesis by mitogen-activated lymphocytes and increased nitric oxide production in LPS-stimulated chBMMs. The results confirm that IL-10 is an inducible feedback regulator of immune response in chickens, and could be the target for improved vaccine efficacy or breeding strategies

    Comparative genetic analysis: the utility of mouse genetic systems for studying human monogenic disease

    One of the long-term goals of mutagenesis programs in the mouse has been to generate mutant lines to facilitate the functional study of every mammalian gene. With a combination of complementary genetic approaches and advances in technology, this aim is slowly becoming a reality. One of the most important features of this strategy is the ability to identify and compare a number of mutations in the same gene, an allelic series. With the advent of gene-driven screening of mutant archives, the search for a specific series of interest is now a practical option. This review focuses on the analysis of multiple mutations from chemical mutagenesis projects in a wide variety of genes and the valuable functional information that has been obtained from these studies. Although gene knockouts and transgenics will continue to be an important resource to ascertain gene function, with a significant proportion of human diseases caused by point mutations, identifying an allelic series is becoming an equally efficient route to generating clinically relevant and functionally important mouse models
