Magneto-optical properties of MnBiAl thin films
Mn-Bi-Al thin films. were produced by sequential evaporation of the constituents, followed by an anneal at 300 °C. The temperature and composition dependencies of the Kerr rotation angle, absolute reflectivity, and magnetic anisotropy were measured. The results show that, up to 30 at. % Al concentration, the thin films retain the pure MnBi hexagonal structure. Further, for suitable Al content, the films have the same large Kerr rotation as MnBi. Pure MnBi films exhibit perpendicular anisotropy at room temperature and in-plane anisotropy for temperatures below 142 K. In contrast,. the Al-doped films prepared here have perpendicular anisotropy down to at least 85 K. The increased coercivities of the Al-doped films are attributed to the occupation of grain-boundary and interstitial sites of the NiAs-type hexagonal structure by the Al-atoms
Reconstruction of the eruptive history of Usu volcano, Hokkaido, Japan, inferred from petrological correlation between tephras and dome lavas
Usu volcano has erupted nine times since 1663. Most eruptive events started with an explosive eruption, which was followed by the formation of lava domes. However, the ages of several summit lava domes and craters remain uncertain. The petrological features of tephra deposits erupted from 1663 to 1853 are known to change systematically. In this study, we correlated lavas with tephras under the assumption that lava and tephra samples from the same event would have similar petrological features. Although the initial explosive eruption in 1663 was not accompanied by lava effusion, lava dome or cryptodome formation was associated with subsequent explosive eruptions. We inferred the location of the vent associated with each event from the location of the associated lava dome and the pyroclastic flow deposit distribution and found that the position of the active vent within the summit caldera differed for each eruption from the late 17th through the 19th century. Moreover, we identified a previously unrecognized lava dome produced by a late 17th century eruption; this dome was largely destroyed by an explosive eruption in 1822 and was replaced by a new lava dome during a later stage of the 1822 event at nearly the same place as the destroyed dome. This new interpretation of the sequence of events is consistent with historical sketches and documents. Our results show that petrological correlation, together with geological evidence, is useful not only for reconstructing volcanic eruption sequences but also for gaining insight into future potential disasters
Gefitinib induction followed by chemoradiotherapy in EGFR-mutant, locally advanced non-small-cell lung cancer: LOGIK0902/OLCSG0905 phase II study
Background: The role of epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitor (TKI) induction coupled with standard concurrent chemoradiotherapy (CRT) is unclear in unresectable, stage III, EGFR-mutant non-small-cell lung cancer (NSCLC). Therefore, a phase II trial was conducted to evaluate the efficacy and safety of gefitinib induction followed by CRT in this disease setting.
Patients and methods: Patients with unresectable, EGFR-mutant, stage III NSCLC were administered gefitinib monotherapy (250 mg/day) for 8 weeks. Subsequently, patients without disease progression during induction therapy were administered cisplatin and docetaxel (40 mg/m(2) each) on days 1, 8, 29, and 36 with concurrent radiotherapy at a total dose of 60 Gy. The primary endpoint was the 2-year overall survival (OS) rate, which was hypothesized to reach 85%, with a threshold of the lower limit of 60%.
Results: Twenty patients (median age: 66 years; male/female: 9/11; histology: 20 adenocarcinoma; stage IIIA/IIIB: 9/11; and exon 19/21: 10/10) were enrolled. The 2-year OS rate was 90% (90% confidence interval: 71.4% to 96.8%), indicating that this trial met the primary objective. The overall response rate and 1- and 2-year progression-free survival rates were 85.0%, 58.1%, and 36.9%, respectively. Grade >= 3 adverse events (>10%) included hepatic toxicity during the induction phase and neutropenia and febrile neutropenia in the CRT phase. Radiation pneumonitis grade >= 3 or treatment-related death did not occur.
Conclusions: This is the first prospective study to demonstrate the favorable efficacy and safety of EGFR-TKI induction followed by standard CRT in EGFR-mutant, stage III NSCLC. Further confirmatory studies are needed
Antioxidant, analgesic and anti-inflammatory activities of Leucas cephalotes (Roxb.ex Roth) Spreng
The whole plant of the methanolic extract from Leucas cephalotes was screened for invitro antioxidant (using the DPPH method), invivo analgesic (using hot plate test in mice) and anti-inflammatory (using rat paw edema test) activities. The methanolic extract of Leucas cephalotes (MELC) scavenged the DPPH radicals in a dose-dependent manner. The IC50 value to scavenge DPPH radicals was found to be 421.3µg/ml. A significant (pO extrato metanólico total de Leucas cephalotes foi submetido à triagem para as atividades antioxidante in vitro (utilizando o método DPPH), analgésica (utilizando teste da placa quente, em camundongos) e antiinflamatória (utilizando teste de edema da pata de rato), nas doses de 200 e 400 mg/kg. O extrato metanólico de Leucas cephalotes (MELC) inativou radicais difenil picril hidrazila (DPPH) de forma dose-dependente. O IC50 para essa atividade foi de 421,3 µg/mL. Observou-se atividade analgésica significativa (p<0,0005) a 60 minutos, com 200 mg/kg, e, com 400 mg/kg, observou-se atividade máxima. A resposta antiinflamatória máxima foi produzida, respectivamente, em 3 h e 2 h, com doses de 200 e 400 mg/kg. Estes resultados sugerem que o extrato metanólico de Leucas cephalotes apresenta efeitos analgésico e antiinflamatório significativos, comparáveis aos fármacos padrão
Modeling rare gene variation to gain insight into the oldest biomarker in autism: construction of the serotonin transporter Gly56Ala knock-in mouse
Alterations in peripheral and central indices of serotonin (5-hydroxytryptamine, 5-HT) production, storage and signaling have long been associated with autism. The 5-HT transporter gene (HTT, SERT, SLC6A4) has received considerable attention as a potential risk locus for autism-spectrum disorders, as well as disorders with overlapping symptoms, including obsessive-compulsive disorder (OCD). Here, we review our efforts to characterize rare, nonsynonymous polymorphisms in SERT derived from multiplex pedigrees carrying diagnoses of autism and OCD and present the initial stages of our effort to model one of these variants, Gly56Ala, in vivo. We generated a targeting vector to produce the Gly56Ala substitution in the Slc6a4 locus by homologous recombination. Following removal of a neomycin resistance selection cassette, animals exhibiting germline transmission of the Ala56 variant were bred to establish a breeding colony on a 129S6 background, suitable for initial evaluation of biochemical, physiological and behavioral alterations relative to SERT Gly56 (wildtype) animals. SERT Ala56 mice were achieved and exhibit a normal pattern of transmission. The initial growth and gross morphology of these animals is comparable to wildtype littermate controls. The SERT Ala56 variant can be propagated in 129S6 mice without apparent disruption of fertility and growth. We discuss both the opportunities and challenges that await the physiological/behavioral analysis of Gly56Ala transgenic mice, with particular reference to modeling autism-associated traits
