    Standardization of serum cholesterol assays by use of serum calibrators and direct addition of Liebermann-Burchard reagent

    Serum cholesterol concentrations of subjects in epidemiological studies were measured after direct addition of Liebermann-Burchard reagent; results were calibrated with human serum pools assayed according to Abell et al. (J. Biol. Chem. 195:357-366, 1952). Accuracy and precision were monitored for six years by analysis of internal-control pools and blind external-control pools. For various internal-control pools, the imprecision (CV) of the long-term averages of run means ranged from 0.5 to 0.9%. The within-run CV for internal control and patients' sera was about 1%. For blind control sera with different concentrations (provided by the Centers for Disease Control, Atlanta, GA, over the same period), the average difference per three-month period between the values found and the target values was usually between -0.5% and 0.7% for medium-concentration pools and between -2% and 2% for low- and high-concentration pools (extreme values: -2.4% and 2.5%). The CV per three-month period ranged from 0.6 to 2.7%. Sera from subjects on diets of high or low linoleic acid content were analyzed to study the effect of the fatty acid portion of serum cholesterol esters; the differences between values obtained with the comparison method and the direct method was insignificant on both diets. We conclude that the use of serum calibrators eliminates the bias inherent in the direct method

    Teaching Intercultural Competence in Translator Training

    In this position paper we define an interculturally competent translator as one that demonstrates a high level of intercultural knowledge, skills, attitude and flexibility throughout his or her professional engagements. We argue that to attain this goal in translator training intercultural competence needs to be introduced into the curriculum explicitly and in a conceptually clear manner. In this article we provide an overview of earlier attempts at discussing the role of intercultural communication in translator training curricula and we discuss the various pedagogical and practical challenges involved. We also look at some future challenges, identifying increasing societal diversity as both a source of added urgency into intercultural training and a challenge for traditional biculturally based notions of translators’ intercultural competence and we argue for the central role of empathy. Finally, and importantly, we introduce the contributions to the special issue

    Transcreation and the professions

    In Cultus 6 David Crystal said "I don't expect my translator to be a mind-reader [...] knowing about the presuppositions and intentions underlying the utterances made by the participants". Issue 7 will focus on this very point. Who should translate, and who is translating the presuppositions and intentions underlying the utterances made by the participants in the real world? A term for this process is "transcreation", which has been used in the Arts (in particular poetry and drama) to talk about transposition into a different language or into a different medium (a poem is transcreated into an art form or onto the stage). Transcreation has also been closely linked with localisation, but differs in that a transcreator is expected to take much more account of the original language/culture/context than a localiser who will be 100% client or end-user oriented. As interculturalists should we agree or disagree with Crystal? Why should (or should not) the translator or interpreter be a transcreator? We would like contributions that focus on these issues, and in particular on these other (non) professionals such as crowd-sourced volunteer translators, fansubbers, international journalists, subject specialists with some foreign language understanding, cultural mediators, community interpreters and child interpreters. How important are they, and what sort of job are they doing? Also what, if any difference is there between these transcreators and translators/interpreters; and what should the profession be doing about accounting for the unsaid, for the presuppositions and intentions underlying the utterances

    A frequent PLCγ1 mutation in adult T-cell leukemia/lymphoma determines functional properties of the malignant cells

    BACKGROUND: Development of adult T-cell leukemia/lymphoma (ATL) involves human T-cell leukemia virus type 1 (HTLV-1) infection and accumulation of somatic mutations. The most frequently mutated gene in ATL (36 % of cases) is phospholipase C gamma1 (PLCG1). PLCG1 is also frequently mutated in other T-cell lymphomas. However, the functional consequences of the PLCG1 mutations in cancer cells have not been characterized. METHODS: We compared the activity of the wild-type PLCγ1 with that of a mutant carrying a hot-spot mutation of PLCγ1 (S345F) observed in ATL, both in cells and in cell-free assays. To analyse the impact of the mutation on cellular properties, we quantified cellular proliferation, aggregation, chemotaxis and apoptosis by live cell-imaging in an S345F+ ATL-derived cell line (KK1) and a KK1 cell line in which we reverted the mutation to the wild-type sequence using CRISPR/Cas9 and homology-directed repair. FINDINGS: The PLCγ1 S345F mutation results in an increase of basal PLC activity in vitro and in different cell types. This higher basal activity is further enhanced by upstream signalling. Reversion of the S345F mutation in the KK1 cell line resulted in reduction of the PLC activity, lower rates of proliferation and aggregation, and a marked reduction in chemotaxis towards CCL22. The PLCγ1-pathway inhibitors ibrutinib and ritonavir reduced both the PLC activity and the tested functions of KK1 cells. INTERPRETATION: Consistent with observations from clinical studies, our data provide direct evidence that activated variants of the PLCγ1 enzyme contribute to the properties of the malignant T-cell clone in ATL.     Training the Trainers in Embedding Assessment Literacy into Module Design: A Case Study of a Collaborative Transcreation Project

    Translator trainers are being asked to respond in their course and module design to a variety of changing requirements, including institutional, professional and pedagogical ones. This paper proposes a way that translator trainers can respond to two sets of these new requirements. The first are those for a widening conception of translation brought about by the rapid globalisation of markets and the needs for intercultural mediators (Katan, 2016; Massey et al., 2017; Massey & Wieder, 2019). Indeed, Katan finishes the paper with the startling statement ‘There is really no question about it: if T/Is are going to survive they must make the transcreational turn’ (ibid.: 378). The second set of requirements comes from the process of articulating what attributes a graduate should possess and how these attributes are developed (Hughes, 2010). As the Higher Education Academy (HEA, 2012; see also Elkington, 2016) state, one way to ensure that the graduate skills are developed is through new assessment practices based on assessment for learning and assessment literacy. New assessment practices mean that assessment ‘does not just contribute to learning at university, but develops learning and evaluative skills essential for employment and lifelong learning’ (ibid.: 10). This paper offers translation trainers an approach to module design which can address both these sets of demands. The module is designed with a collaborative transcreation project at its core and has incorporated assessment literacy into the design. The study is supported with quantitative and qualitative data gained from a survey of participating students. By introducing the case study of our module design and linking the design to the underlying theories which informed it, the paper provides trainers with a set of concepts which could be applied to their own curricula needs in order to future proof their students in the changing employment market

    Screening for protein-protein interactions using Förster resonance energy transfer (FRET) and fluorescence lifetime imaging microscopy (FLIM)

    We present a high content multiwell plate cell-based assay approach to quantify protein interactions directly in cells using Förster resonance energy transfer (FRET) read out by automated fluorescence lifetime imaging (FLIM). Automated FLIM is implemented using wide-field time-gated detection, typically requiring only 10 s per field of view (FOV). Averaging over biological, thermal and shot noise with 100's to 1000's of FOV enables unbiased quantitative analysis with high statistical power. Plotting average donor lifetime vs. acceptor/donor intensity ratio clearly identifies protein interactions and fitting to double exponential donor decay models provides estimates of interacting population fractions that, with calibrated donor and acceptor fluorescence intensities, can yield dissociation constants. We demonstrate the application to identify binding partners of MST1 kinase and estimate interaction strength among the members of the RASSF protein family, which have important roles in apoptosis via the Hippo signalling pathway. KD values broadly agree with published biochemical measurements

    Importance of Vacancies and Doping in the Hole-Transporting Nickel Oxide Interface with Halide Perovskites

    International audienceNickel oxide (NiO) is a commonly used contact material for a variety of thin-film optoelectronic technologies based on organic or hybrid materials. In such setups, interfaces play a crucial role as they can reduce, if not kill, the device performances by bringing additional traps or energy barriers, hindering the extraction of charge carriers from the active layer. Here, we computationally examine a prototype halide perovskite architecture, NiO/MAPbI (MA = CHNH), that has shown excellent photovoltaic performance and, in particular, a large open-circuit voltage. We show that efficient hole collection is achieved only when considering the role of vacancies induced by standard material deposition techniques. Specifically, Ni vacancies lead to nearly perfect valence band energy level alignment between the active layer and the contact material. Finally, we show how Li doping greatly improves the performances of the device and further propose alternative dopants. Our results suggest the high tunability of NiO interfaces for the design of optimized optoelectronic devices far beyond that of halide perovskites
