    Sibling Relationships with an Autistic Sibling

    This study was conducted to determine how typically developing siblings feel about their sibling with Autism or another related disorder. The methods used were a combination of quantitative data and qualitative interviews. The quantitative data consisted of basic demographic data about the family such as the parent’s marital status, household members, and the diagnosis of the Autistic child. The qualitative data consisted of interviews with one or two typical siblings from that family. This study interviewed eight typical siblings from six different families with an Autistic child: two of the siblings were brothers and six were sisters of a child diagnosed with Autism, Asperger’s Disorder or PDD-NOS. Two of the children with Autism were twins with a typical twin. Plus three out of the six Autistic children were girls, which is not typical of Autism. In this study, the typical siblings expressed moments of frustration and fun with their Autistic sibling. Overall, the siblings were mostly empathetic, supportive and loving towards their Autistic sibling

    Mutagenesis of the dimer interface residues of tethered and untethered HIV-1 protease result in differential activity and suggest multiple mechanisms of compensation

    As is the case for all retroviruses, the protease of HIV-1 is only functional as a homodimer; dimerization of two protease monomers results in the formation of the enzyme active site. This dimer structure is supported primarily by interactions between the first four amino-terminal and the last four carboxy-terminal amino acids. These eight amino acids form a beta-sheet in which hydrophobic residues are oriented towards the core of the molecule and polar residues are directed towards the solvent. Although the structure of the dimer interface has been determined, the forces that support dimerization have not been fully characterized. Here, we describe a tethered construct in which two protease monomers are joined by a 5 amino acid linker. We evaluate the relative role of each dimer interface residue in functional homo- and heterodimers. Our studies indicate that the hydrophobic residues of the dimer interface are particularly important in maintaining enzyme activity and that enzyme activity is more sensitive to substitutions of the C-terminal amino acids. Further, we demonstrate that the presence of the tether is able to compensate for mutations within the dimer interface that inactivate the enzyme

    The Lark on the Strand: A Study of a Traditional Irish Flute Player and His Music

    This thesis provides an in-depth study of a traditional Irish flute player, Jack Coen. Jack, raised in the village of Woodford in County Galway, immigrated to America in 1949 at the age of twenty-four. With his large repertoire of Irish traditional dance tunes, Jack has played music at parties, dances, with the New York Ceilidhe Band, and at festivals such as the 1976 Festival of American Folklife in Washington, D.C. Jack teaches the flute and the tin whistle and is recognized both as a teacher and as a player. By examining Jack Coen’s music we learn about the style, technique, and repertoire of a traditional Irish flute player. And by focusing on the performer, we observe the relationship between the stylistic features of Jacks’ music and his sense of musical aesthetics; that is, his perception of how the traditional Irish flute should sound and the way in which he aims to achieve a musical quality on the flute. From the performer we also understand more about the traditional process of transmission by discovering how the music has been learned, the different situations of performance, and the manner in which the music is taught. This understanding provides us with the knowledge of how the music has been carried on from generation to generation and by whom. In the case of Jack Coen, and the many Irish immigrant musicians who came to America before him, we see how an Irish musical tradition has functioned when transplanted to another culture

    Cancer Incidence in World Trade Center Rescue and Recovery Workers, 2001–2008

    Background: World Trade Center (WTC) rescue and recovery workers were exposed to a complex mix of pollutants and carcinogens. Objective: The purpose of this investigation was to evaluate cancer incidence in responders during the first 7 years after 11 September 2001. Methods: Cancers among 20,984 consented participants in the WTC Health Program were identified through linkage to state tumor registries in New York, New Jersey, Connecticut, and Pennsylvania. Standardized incidence ratios (SIRs) were calculated to compare cancers diagnosed in responders to predicted numbers for the general population. Multivariate regression models were used to estimate associations with degree of exposure. Results: A total of 575 cancers were diagnosed in 552 individuals. Increases above registry-based expectations were noted for all cancer sites combined (SIR = 1.15; 95% CI: 1.06, 1.25), thyroid cancer (SIR = 2.39; 95% CI: 1.70, 3.27), prostate cancer (SIR = 1.21; 95% CI: 1.01, 1.44), combined hematopoietic and lymphoid cancers (SIR = 1.36; 95% CI: 1.07, 1.71), and soft tissue cancers (SIR = 2.26; 95% CI: 1.13, 4.05). When restricted to 302 cancers diagnosed ≥ 6 months after enrollment, the SIR for all cancers decreased to 1.06 (95% CI: 0.94, 1.18), but thyroid and prostate cancer diagnoses remained greater than expected. All cancers combined were increased in very highly exposed responders and among those exposed to significant amounts of dust, compared with responders who reported lower levels of exposure. Conclusion: Estimates should be interpreted with caution given the short follow-up and long latency period for most cancers, the intensive medical surveillance of this cohort, and the small numbers of cancers at specific sites. However, our findings highlight the need for continued follow-up and surveillance of WTC responders

    In Situ Imaging of Bacterial Outer Membrane Projections and Associated Protein Complexes Using Electron Cryo-Tomography

    The ability to produce outer membrane projections in the form of tubular membrane extensions (MEs) and membrane vesicles (MVs) is a widespread phenomenon among diderm bacteria. Despite this, our knowledge of the ultrastructure of these extensions and their associated protein complexes remains limited. Here, we surveyed the ultrastructure and formation of MEs and MVs, and their associated protein complexes, in tens of thousands of electron cryo-tomograms of ~90 bacterial species that we have collected for various projects over the past 15 years (Jensen lab database), in addition to data generated in the Briegel lab. We identified outer MEs and MVs in 13 diderm bacterial species and classified several major ultrastructures: (1) tubes with a uniform diameter (with or without an internal scaffold), (2) tubes with irregular diameter, (3) tubes with a vesicular dilation at their tip, (4) pearling tubes, (5) connected chains of vesicles (with or without neck-like connectors), (6) budding vesicles and nanopods. We also identified several protein complexes associated with these MEs and MVs which were distributed either randomly or exclusively at the tip. These complexes include a secretin-like structure and a novel crown-shaped structure observed primarily in vesicles from lysed cells. In total, this work helps to characterize the diversity of bacterial membrane projections and lays the groundwork for future research in this field

    Bodyweight Perceptions among Texas Women: The Effects of Religion, Race/Ethnicity, and Citizenship Status

    Despite previous work exploring linkages between religious participation and health, little research has looked at the role of religion in affecting bodyweight perceptions. Using the theoretical model developed by Levin et al. (Sociol Q 36(1):157–173, 1995) on the multidimensionality of religious participation, we develop several hypotheses and test them by using data from the 2004 Survey of Texas Adults. We estimate multinomial logistic regression models to determine the relative risk of women perceiving themselves as overweight. Results indicate that religious attendance lowers risk of women perceiving themselves as very overweight. Citizenship status was an important factor for Latinas, with noncitizens being less likely to see themselves as overweight. We also test interaction effects between religion and race. Religious attendance and prayer have a moderating effect among Latina non-citizens so that among these women, attendance and prayer intensify perceptions of feeling less overweight when compared to their white counterparts. Among African American women, the effect of increased church attendance leads to perceptions of being overweight. Prayer is also a correlate of overweight perceptions but only among African American women. We close with a discussion that highlights key implications from our findings, note study limitations, and several promising avenues for future research
