Effect of male age in rainbow trout (Oncorhynchus mykiss) on duration of sperm motility, spermatocrite and eyed egg rate production
Effects of three levels of Rainbow Trout male age (2+, 3+ and 4+ years old), on duration of sperm motility, spermatocrite and eyed egg rate in rainbow trout were studied in Kelardasht Fish Breeding Center. The sperm from 2+ year old males activated by water showed the shortest motility (24.86 sec) (P= 0.007), while those of the 3+ and 4+ years old revealed the longest sperm motility. No significant difference was found between sperm motility of the 3+ and 4+ males. The average spermatocrite in different age groups of male fish was significantly different. This was showed to be 32.66% in 2+ years and 2126 % in 3+ year's male age groups respectively. Eyed egg rate was highest in 2+ years age (86/82%), and was significantly different with the 3+ age group (P=0.02). The eyed egg rate of the 4+ age group was not significantly different with those of the 2+ and 3+ age groups
Purification and partial characterization of serum immunoglobulin from Persian sturgeon (Acipenser persicus)
In this study, immunoglobulins from serum of Persian sturgeon (Acipenser persicus) were purified and partially characterized. Immunoglobulins were purified from the pooled sera by a combination of salt precipitation, Ion-exchange chromatography and gel filtration methods. DEAE sepharosefast flow and sepharose CL-6B columns were used for Ion exchange-chromatography, and gel filtration, respectively. The purity, molecular weight and molecular distribution of the immunoglobulin preparations was determined by gel electrophoresis (SDS-P AGE) in reducing and non-reducing situations. In gel filtrated immunoglobulins two distinct peaks, high molecular weight (HMW Ig) and low molecular weight (LMW Ig) were obtained. Both HMW Ig and LMW Ig had identical heavy and light chains of 72-75 KDa and 27-29 KDa, respectively, in reducing SDS-P AGE. HMW Ig contained a group of bands, including two major bands in non-reducing SDS-PAGE, In contrast LMW Ig contain more than half of the total immunoglobulin, was 190 KDa. In ion exchange chromatography, immunoglobulins were eluted in three peaks. The first was exclusively monomer and others were mixture of monomer and polymers. This is the first report on persian sturgeon immunoglobulins. Results of this investigation showed that persian sturgeon immunoglobulins was not homogenous in respect of molecular distribution, PI and the type of light chain. The presence of more than one genes for light and\or heavy chains or post transcriptional and\or post modifications may be responsible for these variations
Induction of tetraploidy in rainbow trout (Oncorhynchus mykiss) by heat shock
This study has been conducted for determination of optimum time and duration of thermal shock on tetraploidy induction in Rainbow trout. This study was carried out by induction of 28°C thermal shock in different times after fertilization (49.5, 54.0, 58.5, 63.0, 67.5, 72.0, 76.5 and 81,0°C/hour and different duration (8, 10 and 12 min). Nuclear erythrocytes, cellular area and volume measurement were determined among blood smears of fingerling and statistic analysis showed that, induction of tetraploidy was conducted between 0-75% on various treatment, but highest tetraploidy yield (8.4%) was achieved by thermal shock on 28°C within 12 minutes, 74 degree-hour after fertilization. Moreover, tetra ploidy diagnostic was also approved by determination of nucleolus number in fishes. In this respect tetraploid and diploid fishes have 3-4 and 1-2 nucleolus, respectively
The possibility of sex identification in beluga (Huso huso) by using PCR-RAPD technique
The goal of this study was to find a sex-specific DNA sequences in the male and female genomes of beluga (Huso huso), by random amplified polymorphic DNA (RAPD) assays performed on pooled DNA samples. Of the 310 different primers was tested, 7 primers produced no band. The most likely possibility is that PCR conditions have not been suitable for these 7 primers. It is also possible that these RAPD primers did not find suitable binding position to generate fragments. A single band was produced by primer 295 (OPT -17) which was specific to the male DNA. Further analysis of individual male and female beluga demonstrated that this primer yielded a product that were in fact variable and reproducible, but were the result of variation in some individual males and females and not male-specific. Therefore, of the 4146 RAPDs scored, none were tightly linked to a sex-determining gene. The results support the idea that sex chromosomes are either not present or they weakly differentiated in the genome of beluga
Comparison of digestive enzyme activity in the stomach, pyloric caeca and intestine in diploid and triploid female of rainbow trout (Oncorhynchus mykiss)
The effects of chromosome manipulation on the digestive enzyme activity in the rainbow trout were studied. The enzymes included Pepsin, Trypsin, Chymotrypsin, cileimylase, Lipase and Alkaline Phosphatase which were assessed in diploid and triploid female of rainbow trout. Pepsin activity in the stomach of the assessed fish showed no significant difference between the diploid and triploid fish (P>0.05). The measurement of Trypsin and Chymotrypsin activity in the intestine and pyloric caeca revealed no significant difference in the treated and untreated fish (P>0.05). The activity of a-Amylase, Lipase and Alkaline Phosphatase showed no significant difference in the intestine and pyloric caeca of the diploid and triploid fish (P>0.05).The results indicated that chromosome manipulation in rainbow trout had no effects on digestive enzyme activity
Karyotypic characterization of the grass carp, Ctenopharyngodon idella by Leukocyte culture
The karyotype of grass carp, Ctenopharyngodon idella, was studied for determination the number and kind of chromosomes of this fish by leukocyte culture method.
The examination of 47 metaphase plates prepared from 3 specimen indicated that the Chromosome numbers of this species was found 2n=48 and the arm numbers Was determined as NF=84. The karyotype prepared this fish consists of 10 pairs of metacentric, 8 pairs of submetacentric and 6 pairs of subtelocentric chromosomes (10m+8sm+6st). largest chromosome in this species is pair of the submetacentric chromosomes.Publishe
Inheritance of RAPD marker in female grass carp (Ctenopharyngodon idella), male bighead carp (Hypophthalmichthys nobilis) and their F1 hybrids
Genetic variation and inheritance of randomly amplified polymorphic DNA (RAPD) markers in female grass carp (Ctenopharyngodon idella) and male bighead carp (Hypophthalmichthys nobilis) and their F1 offspring hybrid have been studied. For this purpose, genomic DNA was extracted from muscle tissue according to phenol-chloroform method, and six decamer primers were used for amplifying polymorphic DNA. Results from 77 produced bands showed that all RAPD bands pattern of parents were present in F1 offspring hybrid, which indicated the high influence and dominance of the mentioned markers. The low difference in polymorphisms in used markers between F1 offspring hybrid and parents shows that parents are heterozygous in some loci, which can cause low difference in hybrids of two distinct genera. Although our results revealed that RAPD markers had a suitable efficiency in distinguishing parents from F1 hybrids, genetic diversity of triploid and diploid F1 hybrid were not detected by introduced primers.Publishe
