PCR-RFLP and sequencing analysis of ribosomal DNA of Bursaphelenchus nematodes related to pine wilt disease
La réaction en chaîne des polymérases/polymorphisme des fragments de restriction (PCR-RFLP) a été utilisée pour séparer des isolats du nématode #Bursaphelenchus. Les isolats de #B. xylophilus examinés provenaient du Japon, des USA, de Chine et du Canada, et ceux de #B. mucronatus du Japon, de Chine et de la France. L'ADN ribosomal contenant le gène 5.8S, les segments de transciption interne 1 et 2, et les segments partiels des gènes 18S et 28S ont été amplifiés par PCR. La digestion des produits amplifiés provenant de chaque isolat à l'aide de douze endonucléases de restriction et l'examen des données en RFLP qui en découlent révèlent, par une analyse en grappe, une séparation significative entre #B. xylophilus et #B. mucronatus. Parmi les isolats de #B. xylophilus examinés, les isolats pathogènes du Japon, ceux de Chine et des USA étaient tous identiques, tandis que les isloats non pathogènes du Japon étaient légèrement distincts et que ceux du Canada formaient une grappe séparée. Parmi les isolats de #B. mucronatus, deux isolats provenant du Japon étaient très semblables ; de même un autre isolat du Japon et un isolat de Chine étaient identiques. Les données provenant des séquences d'ADN montrent 98 différences (substitution nucléotidiques ou séparations) dans les 884 paires de bases examinées chez les isolats de #B. xylophilus et #B. mucronatus. Les données provenant des séquences d'ADN chez #Aphelenchus avenae et #Aphelenchoides fragariae diffèrent non seulement de celles des #Bursaphelenchus mais aussi entre elles. A novel set of features for continuous hand gesture recognition
Applications requiring the natural use of the human hand as a human–computer interface motivate research on continuous hand gesture recognition. Gesture recognition depends on gesture segmentation to locate the starting and end points of meaningful gestures while ignoring unintentional movements. Unfortunately, gesture segmentation remains a formidable challenge because of unconstrained spatiotemporal variations in gestures and the coarticulation and movement epenthesis of successive gestures. Furthermore, errors in hand image segmentation cause the estimated hand motion trajectory to deviate from the actual one. This research moves toward addressing these problems. Our approach entails using gesture spotting to distinguish meaningful gestures from unintentional movements. To avoid the effects of variations in a gesture’s motion chain code (MCC), we propose instead to use a novel set of features: the (a) orientation and (b) length of an ellipse least-squares fitted to motion-trajectory points and (c) the position of the hand. The features are designed to support classification using conditional random fields. To evaluate the performance of the system, 10 participants signed 10 gestures several times each, providing a total of 75 instances per gesture. To train the system, 50 instances of each gesture served as training data and 25 as testing data. For isolated gestures, the recognition rate using the MCC as a feature vector was only 69.6 % but rose to 96.0 % using the proposed features, a 26.1 % improvement. For continuous gestures, the recognition rate for the proposed features was 88.9 %. These results show the efficacy of the proposed method
Urinary sodium-to-potassium ratio and intake of sodium and potassium among men and women from multiethnic general populations: the INTERSALT Study
The Na/K ratio may be more strongly related to blood pressure and cardiovascular disease than sodium or potassium. The casual urine Na/K ratio can provide prompt on-site feedback, and with repeated measurements, may provide useful individual estimates of the 24-h ratio. The World Health Organization has published guidelines for sodium and potassium intake, but no generally accepted guideline prevails for the Na/K ratio. We used standardized data on 24 h and casual urinary electrolyte excretion obtained from the INTERSALT Study for 10,065 individuals aged 20-59 years from 32 countries (52 populations). Associations between the casual urinary Na/K ratio and the 24-h sodium and potassium excretion of individuals were assessed by correlation and stratification analyses. The mean 24-h sodium and potassium excretions were 156.0 mmol/24 h and 55.2 mmol/24 h, respectively; the mean 24-h urinary Na/K molar ratio was 3.24. Pearson's correlation coefficients (r) for the casual urinary Na/K ratio with 24-h sodium and potassium excretions were 0.42 and -0.34, respectively, and these were 0.57 and -0.48 for the 24-h ratio. The urinary Na/K ratio predicted a 24-h urine Na excretion of <85 mmol/day (the WHO recommended guidelines) with a sensitivity of 99.7% and 94.0%, specificity of 39.5% and 48.0%, and positive predictive value of 96.3% and 61.1% at the cutoff point of 1 in 24 h and casual urine Na/K ratios, respectively. A urinary Na/K molar ratio <1 may be a useful indicator for adherence to the WHO recommended levels of sodium and, to a lesser extent, the potassium intake across different populations; however, cutoff points for Na/K ratio may be tuned for localization
Recovering 3D Shape with Absolute Size from Endoscope Images Using RBF Neural Network
Medical diagnosis judges the status of polyp from the size and the 3D shape of the polyp from its medical endoscope image. However the medical doctor judges the status empirically from the endoscope image and more accurate 3D shape recovery from its 2D image has been demanded to support this judgment. As a method to recover 3D shape with high speed, VBW (Vogel-BreuĂź-Weickert) model is proposed to recover 3D shape under the condition of point light source illumination and perspective projection. However, VBW model recovers the relative shape but there is a problem that the shape cannot be recovered with the exact size. Here, shape modification is introduced to recover the exact shape with modification from that with VBW model. RBF-NN is introduced for the mapping between input and output. Input is given as the output of gradient parameters of VBW model for the generated
sphere. Output is given as the true gradient parameters of true values of the generated sphere. Learning mapping with
NN can modify the gradient and the depth can be recovered according to the modified gradient parameters. Performance of
the proposed approach is confirmed via computer simulation and real experiment
Development of a simultaneous analytical method for five conjugated cholesterol metabolites in urine and investigation of their performance as diagnostic markers for Niemann-Pick disease type C
Niemann-Pick disease type C (NPC) is an autosomal recessive disorder characterized by progressive nervous degeneration. Because of the diversity of clinical symptoms and onset age, the diagnosis of this disease is difficult. Therefore, biomarker tests have attracted significant attention for earlier diagnostics. In this study, we developed a simultaneous analysis method for five urinary conjugated cholesterol metabolites, which are potential diagnostic biomarkers for a rapid, convenient, and noninvasive chemical diagnosis, using liquid chromatography/tandem mass spectrometry (LC/MS/MS). By the method, their urinary concentrations were quantified and the NPC diagnostic performances were evaluated. The developed LC/MS/MS method showed high accuracy and and satisfied all analytical method validation criteria. Analyzing the urine of healthy controls and patients with NPC, three of five urinary conjugated cholesterol metabolites concentrations corrected by urinary creatinine were significantly higher in the patients with NPC. As a result of receiver operating characteristics analysis, the urinary metabolites might have excellent diagnostic marker performance. 3β-sulfooxy-7β-hydroxy-5-cholenoic acid showed particularly excellent diagnostic performance with both 100% clinical sensitivity and specificity, suggesting that it is a useful NPC diagnostic marker. The urinary conjugated cholesterol metabolites exhibited high NPC diagnostic marker performance and could be used for NPC diagnosis
Structural Determination of Lysosphingomyelin-509 and Discovery of Novel Class Lipids from Patients with Niemann–Pick Disease Type C
Niemann–Pick disease type C (NPC) is an autosomal recessive disorder caused by the mutation of cholesterol-transporting proteins. In addition, early treatment is important for good prognosis of this disease because of the progressive neurodegeneration. However, the diagnosis of this disease is difficult due to a variety of clinical spectrum. Lysosphingomyelin-509, which is one of the most useful biomarkers for NPC, was applied for the rapid and easy detection of NPC. The fact that its chemical structure was unknown until recently implicates the unrevealed pathophysiology and molecular mechanisms of NPC. In this study, we aimed to elucidate the structure of lysosphingomyelin-509 by various mass spectrometric techniques. As our identification strategy, we adopted analytical and organic chemistry approaches to the serum of patients with NPC. Chemical derivatization and hydrogen abstraction dissociation–tandem mass spectrometry were used for the determination of function groups and partial structure, respectively. As a result, we revealed the exact structure of lysosphingomyelin-509 as N-acylated and O-phosphocholine adducted serine. Additionally, we found that a group of metabolites with N-acyl groups were increased considerably in the serum/plasma of patients with NPC as compared to that of other groups using targeted lipidomics analysis.
