Steel beam–column joint with discontinuous vertical reinforcing bars
The authors have previously proposed steel beam–column connections for precast concrete frames. The steel–concrete composite frames combined the advantages of the fast assembly of steel and the low cost of concrete structures. However, when not enough space is available at column–beam joints, steel sections from beams cannot be connected with column brackets. To address this issue, this paper explores the strategy of disconnecting some vertical reinforcing bars at the joints by connecting vertical steel reinforcements to steel plates placed above and below column steels, to provide a load transfer path. Loads from re-bars are transferred to steel plates, column steels, and back to steel plates and re-bars below the column steels. This strategy provided space for beam–column joints of composite frames. Extensive experiments were performed to verify load transfer from re-bars to steel plates above joints and from the steel plates to re-bars below the joint. The flexural load-bearing capacity of a column with a total of 24 vertical re-bars was compared to that of columns with discontinuous re-bars at the joints; the number of discontinuous re-bars at the joint used in the column specimens tested was 0 (0.0%), 4 (16.7%), 12 (50.0%), and 20 (83.3%)
SnapFusion: Text-to-Image Diffusion Model on Mobile Devices within Two Seconds
Text-to-image diffusion models can create stunning images from natural
language descriptions that rival the work of professional artists and
photographers. However, these models are large, with complex network
architectures and tens of denoising iterations, making them computationally
expensive and slow to run. As a result, high-end GPUs and cloud-based inference
are required to run diffusion models at scale. This is costly and has privacy
implications, especially when user data is sent to a third party. To overcome
these challenges, we present a generic approach that, for the first time,
unlocks running text-to-image diffusion models on mobile devices in less than
seconds. We achieve so by introducing efficient network architecture and
improving step distillation. Specifically, we propose an efficient UNet by
identifying the redundancy of the original model and reducing the computation
of the image decoder via data distillation. Further, we enhance the step
distillation by exploring training strategies and introducing regularization
from classifier-free guidance. Our extensive experiments on MS-COCO show that
our model with denoising steps achieves better FID and CLIP scores than
Stable Diffusion v with steps. Our work democratizes content creation

Electrical current suppression in Pd-doped vanadium pentoxide nanowires caused by reduction in PdO due to hydrogen exposure
Pd nanoparticle-doped vanadium pentoxide nanowires (Pd-VONs) were synthesized. Electrical current suppression was observed when the Pd-VON was exposed to hydrogen gas, which cannot be explained by the work function changes mentioned in previous report such as Pd-doped carbon nanotubes and SnO 2 nanowires. Using the x-ray photoelectron spectroscopy, we found that the reduction in PdO due to hydrogen exposure plays an important role in the current suppression of the Pd-VON.open4
Human herpesvirus 6A induces apoptosis of primary human fetal astrocytes via both caspase-dependent and -independent pathways
<p>Abstract</p> <p>Background</p> <p>Human herpesvirus 6 (HHV-6) is a T-lymphtropic and neurotropic virus that can infect various types of cells. Sequential studies reported that apoptosis of glia and neurons induced by HHV-6 might act a potential trigger for some central nervous system (CNS) diseases. HHV-6 is involved in the pathogenesis of encephalitis, multiple sclerosis (MS) and fatigue syndrome. However, the mechanisms responsible for the apoptosis of infected CNS cells induced by HHV-6 are poorly understood. In this study, we investigated the cell death processes of primary human fetal astrocytes (PHFAs) during productive HHV-6A infection and the underlying mechanisms.</p> <p>Results</p> <p>HHV-6A can cause productive infection in primary human fetal astrocytes. Annexin V-PI staining and electron microscopic analysis indicated that HHV-6A was an inducer of apoptosis. The cell death was associated with activation of caspase-3 and cleavage of poly (ADP-ribose) polymerase (PARP), which is known to be an important substrate for activated caspase-3. Caspase-8 and -9 were also significantly activated in HHV-6A-infected cells. Moreover, HHV-6A infection led to Bax up-regulation and Bcl-2 down-regulation. HHV-6A infection increased the release of Smac/Diablo, AIF and cytochrome c from mitochondria to cytosol, which induced apoptosis via the caspase-dependent and -independent pathways. In addition, we also found that anti-apoptotic factors such as IAPs and NF-κB decreased in HHV-6A infected PHFAs.</p> <p>Conclusion</p> <p>This is the first demonstration of caspase-dependent and -independent apoptosis in HHV-6A-infected glial cells. These findings would be helpful in understanding the mechanisms of CNS diseases caused by HHV-6.</p
Systemic Delivery of Oncolytic Adenoviruses Targeting Transforming Growth Factor-β Inhibits Established Bone Metastasis in a Prostate Cancer Mouse Model
Abstract We have examined whether Ad.sT?RFc and TAd.sT?RFc, two oncolytic viruses expressing soluble transforming growth factor-? receptor II fused with human Fc (sTGF?RIIFc), can be developed to treat bone metastasis of prostate cancer. Incubation of PC-3 and DU-145 prostate tumor cells with Ad.sT?RFc and TAd.sT?RFc produced sTGF?RIIFc and viral replication; sTGF?RIIFc caused inhibition of TGF-?-mediated SMAD2 and SMAD3 phosphorylation. Ad(E1-).sT?RFc, an E1? adenovirus, produced sTGF?RIIFc but failed to replicate in tumor cells. To examine the antitumor response of adenoviral vectors, PC-3-luc cells were injected into the left heart ventricle of nude mice. On day 9, mice were subjected to whole-body bioluminescence imaging (BLI). Mice bearing hind-limb tumors were administered viral vectors via the tail vein on days 10, 13, and 17 (2.5?1010 viral particles per injection per mouse, each injection in a 0.1-ml volume), and subjected to BLI and X-ray radiography weekly until day 53. Ad.sT?RFc, TAd.sT?RFc, and Ad(E1-).sT?RFc caused significant inhibition of tumor growth; however, Ad.sT?RFc was the most effective among all the vectors. Only Ad.sT?RFc and TAd.sT?RFc inhibited tumor-induced hypercalcemia. Histomorphometric and synchrotron micro-computed tomographic analysis of isolated bones indicated that Ad.sT?RFc induced significant reduction in tumor burden, osteoclast number, and trabecular and cortical bone destruction. The CDEX-1 1 kg Point-Contact Germanium Detector for Low Mass Dark Matter Searches
The CDEX Collaboration has been established for direct detection of light
dark matter particles, using ultra-low energy threshold p-type point-contact
germanium detectors, in China JinPing underground Laboratory (CJPL). The first
1 kg point-contact germanium detector with a sub-keV energy threshold has been
tested in a passive shielding system located in CJPL. The outputs from both the
point-contact p+ electrode and the outside n+ electrode make it possible to
scan the lower energy range of less than 1 keV and at the same time to detect
the higher energy range up to 3 MeV. The outputs from both p+ and n+ electrode
may also provide a more powerful method for signal discrimination for dark
matter experiment. Some key parameters, including energy resolution, dead time,
decay times of internal X-rays, and system stability, have been tested and
measured. The results show that the 1 kg point-contact germanium detector,
together with its shielding system and electronics, can run smoothly with good
performances. This detector system will be deployed for dark matter search

Pilot Scale Production of Highly Efficacious and Stable Enterovirus 71 Vaccine Candidates
BACKGROUND: Enterovirus 71 (EV71) has caused several epidemics of hand, foot and mouth diseases (HFMD) in Asia and now is being recognized as an important neurotropic virus. Effective medications and prophylactic vaccine against EV71 infection are urgently needed. Based on the success of inactivated poliovirus vaccine, a prototype chemically inactivated EV71 vaccine candidate has been developed and currently in human phase 1 clinical trial. PRINCIPAL FINDING: In this report, we present the development of a serum-free cell-based EV71 vaccine. The optimization at each step of the manufacturing process was investigated, characterized and quantified. In the up-stream process development, different commercially available cell culture media either containing serum or serum-free was screened for cell growth and virus yield using the roller-bottle technology. VP-SFM serum-free medium was selected based on the Vero cell growth profile and EV71 virus production. After the up-stream processes (virus harvest, diafiltration and concentration), a combination of gel-filtration liquid chromatography and/or sucrose-gradient ultracentrifugation down-stream purification processes were investigated at a pilot scale of 40 liters each. Although the combination of chromatography and sucrose-gradient ultracentrifugation produced extremely pure EV71 infectious virus particles, the overall yield of vaccine was 7-10% as determined by a VP2-based quantitative ELISA. Using chromatography as the downstream purification, the virus yield was 30-43%. To retain the integrity of virus neutralization epitopes and the stability of the vaccine product, the best virus inactivation was found to be 0.025% formalin-treatment at 37 °C for 3 to 6 days. Furthermore, the formalin-inactivated virion vaccine candidate was found to be stable for >18 months at 4 °C and a microgram of viral proteins formulated with alum adjuvant could induce strong virus-neutralizing antibody responses in mice, rats, rabbits, and non-human primates. CONCLUSION: These results provide valuable information supporting the current cell-based serum-free EV71 vaccine candidate going into human Phase I clinical trials
