    Conditioning of Leverage Scores and Computation by QR Decomposition

    The leverage scores of a full-column rank matrix A are the squared row norms of any orthonormal basis for range(A). We show that corresponding leverage scores of two matrices A and A + \Delta A are close in the relative sense, if they have large magnitude and if all principal angles between the column spaces of A and A + \Delta A are small. We also show three classes of bounds that are based on perturbation results of QR decompositions. They demonstrate that relative differences between individual leverage scores strongly depend on the particular type of perturbation \Delta A. The bounds imply that the relative accuracy of an individual leverage score depends on: its magnitude and the two-norm condition of A, if \Delta A is a general perturbation; the two-norm condition number of A, if \Delta A is a perturbation with the same norm-wise row-scaling as A; (to first order) neither condition number nor leverage score magnitude, if \Delta A is a component-wise row-scaled perturbation.     Experimental Investigation of Unsteady Thrust Augmentation Using a Speaker-Driven Jet

    An experimental investigation is described in which a simple speaker-driven jet was used as a pulsed thrust source (driver) for an ejector configuration. The objectives of the investigation were twofold. The first was to expand the experimental body of evidence showing that an unsteady thrust source, combined with a properly sized ejector generally yields higher thrust augmentation values than a similarly sized, steady driver of equivalent thrust. The second objective was to identify characteristics of the unsteady driver that may be useful for sizing ejectors, and for predicting the thrust augmentation levels that may be achieved. The speaker-driven jet provided a convenient source for the investigation because it is entirely unsteady (i.e., it has no mean velocity component) and because relevant parameters such as frequency, time-averaged thrust, and diameter are easily variable. The experimental setup will be described, as will the two main measurements techniques employed. These are thrust and digital particle imaging velocimetry of the driver. It will be shown that thrust augmentation values as high as 1.8 were obtained, that the diameter of the best ejector scaled with the dimensions of the emitted vortex, and that the so-called formation time serves as a useful dimensionless parameter by which to characterize the jet and predict performance

    Excavation on the Moon: Regolith Collection for Oxygen Production and Outpost Site Preparation

    The development of a robust regolith moving system for lunar and planetary processing and construction is critical to the NASA mission to the Moon and Mars. Oxygen production may require up to 200 metric tons of regolith collection per year; outpost site development may require several times this amount. This paper describes progress in the small vehicle implement development and small excavation system development. Cratos was developed as a platform for the ISRU project to evaluate the performance characteristics of a low center of gravity, small (0.75m x 0.75m x 0.3m), low-power, tracked vehicle performing excavation, load, haul, and dump operations required for lunar ISRU. It was tested on loose sand in a facility capable of producing level and inclined surfaces, and demonstrated the capability to pick up, carry, and dump sand, allowing it to accomplish the delivery of material to a site.     An Optimized Cell-Based Assay To Assess Influenza Virus Replication by Measuring Neuraminidase Activity and Its Applications for Virological Surveillance

    Year-round virological characterization of circulating epidemic influenza viruses is conducted worldwide to detect the emergence of viruses that may escape pre-existing immunity or acquire resistance to antivirals. High throughput phenotypic assays are needed to complement the sequence-based analysis of circulating viruses and improve pandemic preparedness. The recent entry of a polymerase inhibitor, baloxavir, into the global market further highlighted this need. Here, we optimized a cell-based assay that considerably streamlines antiviral and antigenic testing by replacing lengthy immunostaining and imaging procedures used in current assay with measuring the enzymatic activity of nascent neuraminidase (NA) molecules expressed on the surface of virus-infected cells. For convenience, this new assay was named IRINA (Influenza Replication Inhibition Neuraminidase-based Assay). IRINA was successfully validated to assess inhibitory activity of baloxavir on virus replication by testing a large set (\u3e150) of influenza A and B viruses, including drug resistant strains and viruses collected during 2017–2022. To test its versatility, IRINA was utilized to evaluate neutralization activity of a broadly reactive human anti-HA monoclonal antibody, FI6, and post-infection ferret antisera, as well as the inhibition of NA enzyme activity by NA inhibitors. Performance of IRINA was tested in parallel using respective conventional assays. IRINA offers an attractive alternative to current phenotypic assays, while maintaining reproducibility and high throughput capacity. Additionally, the improved turnaround time may prove to be advantageous when conducting time sensitive studies, such as investigating a new virus outbreak. This assay can meet the needs of surveillance laboratories by providing a streamlined and cost-effective approach for virus characterization

    Understanding the treatment benefit of hyperimmune anti-influenza intravenous immunoglobulin (Flu-IVIG) for severe human influenza

    Background: Antibody-based therapies for respiratory viruses are of increasing importance. The INSIGHT 006 trial administered anti-influenza hyperimmune intravenous immunoglobulin (Flu-IVIG) to patients hospitalized with influenza. Flu-IVIG treatment improved outcomes in patients with influenza B but showed no benefit for influenza A. Methods: To probe potential mechanisms of Flu-IVIG utility, sera collected from patients hospitalized with influenza A or B viruses (IAV or IBV) were analyzed for antibody isotype/subclass and Fcγ receptor (FcγR) binding by ELISA, bead-based multiplex, and NK cell activation assays. Results: Influenza-specific FcγR-binding antibodies were elevated in Flu-IVIG–infused IBV- and IAV-infected patients. In IBV-infected participants (n = 62), increased IgG3 and FcγR binding were associated with more favorable outcomes. Flu-IVIG therapy also improved the odds of a more favorable outcome in patients with low levels of anti-IBV Fc-functional antibody. Higher FcγR-binding antibody was associated with less favorable outcomes in IAV-infected patients (n = 50), and Flu-IVIG worsened the odds of a favorable outcome in participants with low levels of anti-IAV Fc-functional antibody. Conclusion: These detailed serological analyses provide insights into antibody features and mechanisms required for a successful humoral response against influenza, suggesting that IBV-specific, but not IAV-specific, antibodies with Fc-mediated functions may assist in improving influenza outcome. This work will inform development of improved influenza immunotherapies

    Cratos: A Simple Low Power Excavation and Hauling System for Lunar Oxygen Production and General Excavation Tasks

    The development of a robust excavating and hauling system for lunar and planetary excavation is critical to the NASA mission to the Moon and Mars. Cratos was developed as a low center of gravity, small (.75m x .75m x 0.3m), low power tracked test vehicle. The vehicle was modified to excavate and haul because it demonstrated good performance capabilities in a laboratory and field testing. Tested on loose sand in the SLOPE facility, the vehicle was able to pick up, carry, and dump sand, allowing it to accomplish the standard requirements delivery of material to a lunar oxygen production site. Cratos can pick up and deliver raw material to a production plant, as well as deliver spent tailings to a disposal site. The vehicle can complete many other In-Situ Resource Utilization (ISRU) excavation chores and in conjunction with another vehicle or with additional attachments may be able to accomplish all needed ISRU tasks

    Influenza Activity and Composition of the 2022-23 Influenza Vaccine - United States, 2021-22 Season

    Before the emergence of SARS-CoV-2, the virus that causes COVID-19, influenza activity in the United States typically began to increase in the fall and peaked in February. During the 2021-22 season, influenza activity began to increase in November and remained elevated until mid-June, featuring two distinct waves, with A(H3N2) viruses predominating for the entire season. This report summarizes influenza activity during October 3, 2021-June 11, 2022, in the United States and describes the composition of the Northern Hemisphere 2022-23 influenza vaccine. Although influenza activity is decreasing and circulation during summer is typically low, remaining vigilant for influenza infections, performing testing for seasonal influenza viruses, and monitoring for novel influenza A virus infections are important. An outbreak of highly pathogenic avian influenza A(H5N1) is ongoing; health care providers and persons with exposure to sick or infected birds should remain vigilant for onset of symptoms consistent with influenza. Receiving a seasonal influenza vaccine each year remains the best way to protect against seasonal influenza and its potentially severe consequences
