    Induction of CCL8/MCP-2 by mycobacteria through the activation of TLR2/PI3K/Akt signaling pathway.

    Pleural tuberculosis (TB), together with lymphatic TB, constitutes more than half of all extrapulmonary cases. Pleural effusions (PEs) in TB are representative of lymphocytic PEs which are dominated by T cells. However, the mechanism underlying T lymphocytes homing and accumulation in PEs is still incompletely understood. Here we performed a comparative analysis of cytokine abundance in PEs from TB patients and non-TB patients by protein array analysis and observed that MCP-2/CCL8 is highly expressed in the TB-PEs as compared to peripheral blood. Meanwhile, we observed that CCR5, the primary receptor used by MCP-2/CCL8, is mostly expressed on pleural CD4(+) T lymphocytes. Furthermore, we found that infection with either Mycobacterium bovis Bacillus Calmette-Guérin (BCG) or Mycobacterium tuberculosis H37Rv induced production of MCP-2/CCL8 at both transcriptional and protein level in Raw264.7 and THP-1 macrophage cells, mouse peritoneal macrophages as well as human PBMC monocyte-derived macrophages (MDMs). The induction of MCP-2/CCL8 by mycobacteria is dependent on the activation of TLR2/PI3K/Akt and p38 signaling pathway. We conclude that accumulation of MCP-2/CCL8 in TB-PEs may function as a biomarker for TB diagnosis

    The Low Temperature Induced Physiological Responses of Avena nuda

    The paperaim of the was to study the effect of low temperature stress on Avena nuda L. seedlings. Cold stress leads to many changes of physiological indices, such as membrane permeability, free proline content, malondialdehyde (MDA) content, and chlorophyll content. Cold stress also leads to changes of some protected enzymes such as peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT). We have measured and compared these indices of seedling leaves under low temperature and normal temperature. The proline and MDA contents were increased compared with control; the chlorophyll content gradually decreased with the prolongation of low temperature stress. The activities of SOD, POD, and CAT were increased under low temperature. The study was designated to explore the physiological mechanism of cold tolerance in naked oats for the first time and also provided theoretical basis for cultivation and antibiotic breeding in Avena nuda L

    A mutual information theory-based approach for assessing uncertainties in deterministic multi-category precipitation forecasts

    The very nature of weather forecasts and verifications and the way they are used make it impossible for one single or absolute standard of evaluation. However, little research has been conducted on verifying deterministic multi-category forecasts, which is based on the attribute of uncertainty. The authors propose a new approach using two mutual information theory-based scores for assessing the comprehensive uncertainty of all categories and the uncertainty for a certain category in deterministic multi-category precipitation forecasts, respectively. Specifically, the comprehensive uncertainty is defined as the average reduction in uncertainty about the observations resulting from the use of a predictive model to provide all categories forecasts; the uncertainty of a certain category is defined as the reduction in uncertainty about the observations resulting from the use of a predictive model to provide a certain category forecast. By applying the proposed approach and traditional verification methods, the four precipitation forecasting products from the China Meteorological Administration (CMA), European Centre for Medium-Range Weather Forecasts (ECMWF), National Centers for Environmental Prediction (NCEP) and United Kingdom Meteorological Office (UKMO) were verified in the Dahuofang Reservoir Drainage Basin, China. The results indicate that: (1) the proposed approach can better capture the changing patterns of uncertainties with lead times and distinguish the forecasting performance among different forecast products; (2) the proposed approach is resistant to the extreme bias; (3) the proposed approach needs a careful choice of bin width; and (4) the bias analysis is necessary before verifying the uncertainties in precipitation forecasts

    In this article, we are committed to studying the three-dimensional incompressible Navier-Stokes equations, where the viscosity depends on density according to a power law. We investigate the Cauchy problem by constructing an approximation system and bootstrap argument. Finally, we establish the existence of a global strong solution under the conditions of small initial data and the compatibility condition. Meanwhile, the algebraic decay-in-time rates for the solution are also obtained. It is worth pointing out that the degradation of viscosity is allowed

    Research on Deformation Prediction of VMD-GRU Deep Foundation Pit Based on PSO Optimization Parameters

    As a key guarantee and cornerstone of building quality, the importance of deformation prediction for deep foundation pits cannot be ignored. However, the deformation data of deep foundation pits have the characteristics of nonlinearity and instability, which will increase the difficulty of deformation prediction. In response to this characteristic and the difficulty of traditional deformation prediction methods to excavate the correlation between data of different time spans, the advantages of variational mode decomposition (VMD) in processing non-stationary series and a gated cycle unit (GRU) in processing complex time series data are considered. A predictive model combining particle swarm optimization (PSO), variational mode decomposition, and a gated cyclic unit is proposed. Firstly, the VMD optimized by the PSO algorithm was used to decompose the original data and obtain the Internet Message Format (IMF). Secondly, the GRU model optimized by PSO was used to predict each IMF. Finally, the predicted value of each component was summed with equal weight to obtain the final predicted value. The case study results show that the average absolute errors of the PSO-GRU prediction model on the original sequence, EMD decomposition, and VMD decomposition data are 0.502 mm, 0.462 mm, and 0.127 mm, respectively. Compared with the prediction mean square errors of the LSTM, GRU, and PSO-LSTM prediction models, the PSO-GRU on the PTB0 data of VMD decomposition decreased by 62.76%, 75.99%, and 53.14%, respectively. The PTB04 data decreased by 70%, 85.17%, and 69.36%, respectively. In addition, compared to the PSO-LSTM model, it decreased by 8.57% in terms of the model time. When the prediction step size increased from three stages to five stages, the mean errors of the four prediction models on the original data, EMD decomposed data, and VMD decomposed data increased by 28.17%, 3.44%, and 14.24%, respectively. The data decomposed by VMD are more conducive to model prediction and can effectively improve the accuracy of model prediction. An increase in the prediction step size will reduce the accuracy of the deformation prediction. The PSO-VMD-GRU model constructed has the advantages of reliable accuracy and a wide application range, and can effectively guide the construction of foundation pit engineering

    Cytoprotective mechanism of ferulic acid against high glucose-induced oxidative stress in cardiomyocytes and hepatocytes

    Background: Ferulic acid (FA), a phenolic acid, is a potential therapy for diabetes mellitus. FA has been shown to protect against hepatic and myocardial injury and oxidative stress in obese rats with late-stage diabetes, but the mechanism of the antioxidative activity of FA is still unclear. Objective: The aim of this study was to elucidate whether FA can prevent damage to cardiomyocytes and hepatocytes caused by high glucose (HG)-induced oxidative stress and whether the protection effects of FA on these cells are related to the Keap1-Nrf2-ARE signaling pathways. Design: Cells were divided into four groups: a control group (cultured with normal medium), an HG group (medium containing 80 mmol/L glucose), an FA+HG group (medium containing 80 mmol/L glucose and 1, 5, or 10 µg/mL FA), and a dimethylbiguanide (DMBG)+HG group (medium containing 80 mmol/L glucose and 50 µg/mL DMBG). Results: FA treatment significantly increased cell viability and significantly decreased cell apoptosis compared with the HG-treated group. Moreover, FA down-regulated the expression of Keap1 protein and up-regulated the expression of Nrf2 protein and gene transcription of HO-1 and glutathione S-transferase (GST) in a dose-dependent manner. Conclusion: FA alleviated the HG-induced oxidative stress and decreased cell apoptosis in hepatocytes and cardiomyocytes. These effects were associated with the Keap1-Nrf2-ARE signaling pathway

    miR398 Attenuates Heat-Induced Leaf Cell Death via Its Target CSD1 in Chinese Cabbage

    Previous research has shown that miR398 contributed to plant thermotolerance by silencing its target gene COPPER/ZINC SUPEROXIDE DISMUTASE1 (CSD1) in Arabidopsis thaliana. However, the phylogenesis of miR398 and CSD1 in Brassica crop and their role in regulating leaf cell death under heat stress remains unexplored. Here, we characterized the homologous genes of miR398a and CSD1 in Brassica rapa ssp. pekinensis (Chinese cabbage) and found miR398a abundance was accumulated under heat stress (38 °C and 46 °C for 1 h) in Chinese cabbage, while the expression level of its targets BraCSD1-1 and BraCSD2-1 were downregulated. To further explore their role in heat response, we constructed the transgenic plants overexpressing artificial miR398a (aBra-miR398a), Bra-miR398a target mimic (Bra-MIM398a), and BraCSD1-1 in Chinese cabbage for genetic study. Under high temperatures, p35S::aBra-miR398a lines reduced the areas of leaf cell death and delayed the leaf cell death. By contrast, p35S::Bra-MIM398a and p35S::BraCSD1-1 plants enlarged the areas of leaf cell death and displayed the earliness of leaf cell death. Finally, we found that the expression level of stress-responsive genes BraLEA76, BraCaM1, BraPLC, BraDREB2A, and BraP5CS increased in transgenic plants overexpressing aBra-miR398a, which may contribute to their resistance to heat-induced leaf cell death. Taken together, these results revealed the function of Bra-miR398a in attenuating leaf cell death to ensure plant thermotolerance, indicating that the miR398-CSD1 module could be potential candidates for heat-resistant crop breeding

    Cartilage Turnover Reflected by Metabolic Processing of Type II Collagen: A Novel Marker of Anabolic Function in Chondrocytes

    The aim of this study was to enable measurement of cartilage formation by a novel biomarker of type II collagen formation. The competitive enzyme-linked immunosorbent assay (ELISA) Pro-C2 was developed and characterized for assessment of the beta splice variant of type II procollagen (PIIBNP). This is expected to originate primarily from remodeling of hyaline cartilage. A mouse monoclonal antibody (Mab) was raised in mouse, targeting specifically PIIBNP (QDVRQPG) and used in development of the assay. The specificity, sensitivity, 4-parameter fit and stability of the assay were tested. Levels of PIIBNP were quantified in human serum (0.6–2.2 nM), human amniotic fluid (163–188 nM) and sera from different animal species, e.g., fetal bovine serum (851–901 nM) with general good linearity (100% (SD 7.6) recovery) and good intra- and inter-assay variation (CV% < 10). Dose (0.1 to 100 ng/mL) and time (7, 14 and 21 days) dependent release of PIIBNP were evaluated in the conditioned medium from bovine cartilage explants (BEX) and human cartilage explants (HEX) upon stimulation with insulin-like growth factor (IGF-1), transforming growth factor (TGF)-β1 and fibroblastic growth factor-2 (FGF-2). TGF-β1 and IGF-1 in concentrations of 10–100 ng/mL significantly (p < 0.05) induced release of PIIBNP in BEX compared to conditions without treatment (WO). In HEX, IGF-1 100 ng/mL was able to induce a significant increase of PIIBNP after one week compared to WO. FGF-2 did not induce a PIIBNP release in our models. To our knowledge this is the first assay, which is able to specifically evaluate PIIBNP excretion. The Pro-C2 assay seems to provide a promising and novel marker of type II collagen formation

    Poor performance of malaria rapid diagnostic tests for the detection of Plasmodium malariae among returned international travellers in China

    Abstract Background Malaria is a worldwide infectious disease. For countries that have achieved malaria elimination, the prevention of re-establishment due to infections in returned travellers has become important. The accurate and timely diagnosis of malaria is the key in preventing re-establishment, and malaria rapid diagnostic tests (RDTs) are frequently used due to their convenience. However, the RDT performance in Plasmodium malariae (P. malariae) infection diagnosis remains unknown. Methods This study analysed epidemiological features and diagnosis patterns of imported P. malariae cases from 2013 to 2020 in Jiangsu Province and evaluated the sensitivity of four parasite enzyme lactate dehydrogenase (pLDH)-targeting RDTs (Wondfo, SD BIONLINE, CareStart and BioPerfectus) and one aldolase-targeting RDT(BinaxNOW) for P. malariae detection. Furthermore, influential factors were investigated, including parasitaemia load, pLDH concentration and target gene polymorphisms. Results The median duration from symptom onset to diagnosis among patients with P. malariae infection was 3 days, which was longer than that with Plasmodium falciparum (P. falciparum) infection. The RDTs had a low detection rate (39/69, 56.5%) among P. malariae cases. All tested RDT brands had poor performance in P. malariae detection. All the brands except the worst-performing SD BIOLINE, achieved 75% sensitivity only when the parasite density was higher than 5000 parasites/μL. Both pLDH and aldolase showed relatively conserved and low gene polymorphism rates. Conclusions The diagnosis of imported P. malariae cases was delayed. The RDTs had poor performance in P. malariae diagnosis and may threaten the prevention of malaria re-establishment from returned travellers. The improved RDTs or nucleic acid tests for P. malariae cases are urgently needed for the detection of imported cases in the future
