    Skeletal muscle is a highly active tissue comprising up to 40% of total body weight. This highly dynamic tissue relies on mitochondria for calcium homeostasis, program cell death, and ATP production through mitochondrial respiration. The composition of mitochondrial phospholipids influences the functional efficiency of mitochondrial proteins, and healthy membranes include unsaturated acyl chains to promote respiration. Increasing membrane saturation has been implicated in muscle wasting, a condition caused by apoptosis and identified by the release of specific proteins from the mitochondria. It is unclear how membrane composition promotes the release of these pro-apoptotic proteins, specifically cytochrome c, to induce apoptosis. Thus, in this work synthetic membranes mimicking the composition of mitochondrial membranes from healthy and dystrophic mouse muscle were used. Cytochrome c conformation and function was measured with and without these membranes. There were no differences in protein conformation or function between the healthy and dystrophic membrane mimetics. However, cytochrome c affinity to these membranes was increased with greater unsaturation. This suggests that decreasing membrane saturation, as implicated in muscle wasting diseases, promotes the release of cytochrome c and apoptotic-mediated cell death. As such, interventions to improve or maintain membrane unsaturation may prove to be an alternative therapy for muscle wasting diseases

    THE DEVELOPMENT OF GUIDELINES FOR DESIGNING DIGITAL MEDIA TO ENGAGE VISITORS WITH NON-VISIBLE OUTDOOR HERITAGE

    This PhD investigates the role of digital media in optimising visitor engagement with non-visible outdoor heritage. Motivated by concerns that digital media products developed for the heritage sector might not be reaching their potential to enrich the visit experience and concerned about a lack of clarity as to what constitutes visitor engagement; this thesis proposes guidance for the production of interpretive digital media and a framework for visitor engagement. Cultural heritage sites featured in this study are characteristically outdoor locations; frequently non-stewarded with very little tangible evidence of the historical or cultural relevance of the site. The unique potential of digital media products to address the specific challenges of engaging visitors with invisible heritage in these locations is discussed within this thesis. The practice of interpreting heritage is investigated to identify the processes, stages, experiences and behavioural states associated with a high level of engagement. Visitor engagement is defined in this study as being a transformational experience in which the visitor’s emotional and/or cognitive relationship with the heritage is altered. This is achieved when the visitor sufficiently experiences appropriate states of engagement across all stages of the visitor engagement framework. This study proposes guidance to advise and support heritage professionals and their associated designers in the design, development and implementation of interpretive digital media products. Within this guide sits the engagement framework which proposes a framework for engagement, defining the stages (process) and the states (experiences and behaviours) of visitor engagement with cultural heritage. In using this resource the cultural heritage practitioner can be confident of their capacity to run and deliver interpretive digital media projects regardless of their expertise in design or technology. This thesis proposes that well designed interpretive digital media can optimise the engagement of visitors in ways which cannot be achieved by any other single method of interpretation. This PhD contributes a design guide and an engagement framework to the existing field of knowledge regarding interpretive digital design

    Mark and his Gentile Audience: A Traditio-Historical and Socio-Cultural Investigation of Mk 4.35-9.29 and its Interface with Gentile Polytheism in the Roman Near East

    This thesis takes a novel, inter-disciplinary approach to an examination of the Markan evangelist’s portrayal of Jesus’ interface with Gentiles in a central section of his Gospel (Mk 4.35-9.29). As a framework to this section, Mark created a connected account of Jesus’ itinerary that included trips to perform miracles in the Gentile territories of Gerasa, Tyre, Bethsaida, the wider Decapolis and Caesarea Philippi. This thesis examines the role of these pericopae in the narrative as a whole and challenges the view that Mark’s geographical references were largely symbolic, rural and for the most part aimed at Jewish followers. The study scrutinizes Mark’s choice of geographical locations, systematically examines recent research on the religious milieu in these specific locations and brings this research into connection with the Gentile mission portrayed by Mark. The polytheistic and social environment in which Mark’s first century audience functioned has received little attention in recent scholarship and represents a lacuna in New Testament historical-critical research which this study addresses. A detailed exegesis of this section of the narrative concludes that Mark (a) deliberately redacts his text to place miracles in geographical regions where Gentiles predominate; (b) emphasizes obduracy and faithlessness on the part of Jewish officialdom and the Jewish disciples, in contrast to an implied understanding on the part of the Gentiles; (c) orchestrates a prolonged and sustained Jesus mission to the Gentiles as a precursor to his own community’s mission, to respond to their need for support and reassurance and (d) formulates his narrative to engage with his intended first century audience's Graeco-Roman religious and social worldview, inviting them to make comparison between the activities of Jesus and other contemporary miracle-performing men and polytheistic gods

    Perceptions of women senior leaders in the UK Higher Education during the COVID-19 pandemic

    Studies suggest the COVID-19 pandemic disproportionately impacted women. Related research and literature suggest leadership approaches commonly associated with women are more effective during crises, a notion that we sought to explore in this study. This investigation explored how senior women leaders in the UK Higher Education exhibited leadership during the pandemic, whether their approaches had changed during this time and the impact leading during this period had on them. The research used semi-structured interviews with six women senior leaders to uncover insights into their lived experiences. Findings show while their fundamental leadership styles did not change, all participants exhibited situational approaches with higher levels of distributed leadership than pre-pandemic. In several instances, their perception was this was based on gendered differences in their leadership approaches, probably developed because of their socialisation as women. The pressures on participants during the pandemic had a significant personal toll, with this period being articulated as the most difficult in their careers. Peer-to-peer networks functioned as critical support mechanisms for these women leaders. The greatest emergent stressor, however, was government policy changes unrelated to COVID-19 which participants considered would have greater long-term impact and served to destabilise higher education at an already difficult time

    Self-identified strategies to manage intake of tempting foods: cross-sectional and prospective associations with BMI and snack intake

    Objectives:Individuals often use self-directed strategies to manage intake of tempting foods, but what these strategies are and whether they are effective is not well understood. This study assessed the frequency of use and subjective effectiveness of self-directed strategies in relation to BMI and snack intake.Design:A cross-sectional and prospective study with three timepoints (T1: baseline, T2: 3 months, T3: 3 years). At T1, demographics, frequency of use and subjective effectiveness of 41 identified strategies were assessed. At T2 and T3, current weight was reported, and at T2 frequency of snack intake was also recorded.Setting:Online study in the UK.Participants:Data from N=368 participants (Mage=34.41 years; MBMI=25.06 kg/m2) was used for analysis at T1, N=170 (46.20% of the total sample) at T2 and N=51 (13.59%) at T3.Results:Two strategy factors were identified via principal axis factoring: 1) Diet, exercise, reduction of temptations, and cognitive strategies, and 2) Planning, preparation and eating style. For strategy 1, frequency of use, but not subjective effectiveness, was positively related to BMI at T1. Subjective effectiveness predicted an increase in BMI from T1 and T2 to T3. No relationship to snack intake was found. For strategy 2, frequency of use was negatively related to BMI at T1. Neither frequency of use nor subjective effectiveness were related to changes in BMI over time, but subjective effectiveness was negatively correlated with unhealthy snack intake.Conclusion:Self-directed strategies to reduce the intake of tempting foods are not consistently related to BMI or snack intake

    Curricular initiatives that enhance student knowledge and perceptions of sexual and gender minority groups: a critical interpretive synthesis

    Background: There is no accepted best practice for optimizing tertiary student knowledge, perceptions, and skills to care for sexual and gender diverse groups. The objective of this research was to synthesize the relevant literature regarding effective curricular initiatives designed to enhance tertiary level student knowledge, perceptions, and skills to care for sexual and gender diverse populations.Methods: A modified Critical Interpretive Synthesis using a systematic search strategy was conducted in 2015. This method was chosen to synthesize the relevant qualitative and quantitative literature as it allows for the depth and breadth of information to be captured and new constructs to be illuminated. Databases searched include AMED, CINAHL EBM Reviews, ERIC, Ovid MEDLINE, Ovid Nursing Database, PsychInfo, and Google Scholar. Results: Thirty-one articles were included in this review. Curricular initiatives ranging from discrete to multimodal approaches have been implemented. Successful initiatives included discrete sessions with time for processing, and multi-modal strategies. Multi-modal approaches that encouraged awareness of one’s lens and privilege in conjunction with facilitated communication seemed the most effective.Conclusions: The literature is limited to the evaluation of explicit curricula. The wider cultural competence literature offers further insight by highlighting the importance of broad and embedded forces including social influences, the institutional climate, and the implicit, or hidden, curriculum. A combined interpretation of the complementary cultural competence and sexual and gender diversity literature provides a novel understanding of the optimal content and context for the delivery of a successful curricular initiative
