etwa hundert Wissenschaftlerinnen und Wissenschaftlern
aus dem In- und Ausland die
Möglichkeit zur Vorstellung und Diskussion
unterschiedlicher Perspektiven und Herangehensweisen
an die jeweiligen Forschungsfelder
im Zusammenhang mit Transnationalisierungsprozessen
aus der Organisations-,

Effekte von Prostaglandin E2 und EP-Rezeptoragonisten auf die Ausreifung, Antigenaufnahme und Antigenpräsentation von humanen Monozyten-abgeleiteten dendritischen Zellen
Im Mittelpunkt der Arbeit stehen die Auswirkungen des Prostanoids Prostaglandin E2 (PGE2) auf die Fähigkeit von Monozyten-abgeleiteten dendritischen Zellen (MoDC) zur Antigenpräsentation auf MHC-I- und MHC II Molekülen. Die Kreuzpräsentation von exogenem Antigen auf MHC I Molekülen ist eine entscheidende Fähigkeit der dendritischen Zellen (DC), die es ihnen ermöglicht zytotoxische T-Zellen zu aktivieren. PGE2 wird in Aktivierungsprotokollen als „Goldstandard“ in Kombination mit Zytokinen, wie TNF-α, IL-1β und IL 6, als Reifestimulus für DC in klinischen DC-basierten Tumorvakzinierungsstudien verwendet. Bisher wurde über den Effekt von PGE2 auf die Antigenpräsentation von Tumorantigen in Proteinform nicht berichtet.
In dieser Arbeit wurde die Auswirkung von PGE2 und spezifischen EP Rezeptoragonisten auf verschiedene Funktionen von MoDC, die für die Antigenpräsentation auf MHC-I- und MHC-II-Molekülen relevant sind, untersucht. Zuerst wurde die Auswirkung von PGE2 und den EP Rezeptoragonisten auf die Ausreifung der MoDC untersucht. Hierbei wurde ersichtlich, dass MoDC nach Inkubation mit PGE2, vermittelt über EP2/4 Rezeptoren, die Aktivierungsmarker HLA A, HLA-B und HLA-C3, HLA-DR, CD83 und CD86 hochregulieren.
Als nächstes wurde der Einfluss von PGE2 auf die Antigenaufnahmefähigkeit der MoDC untersucht. Hierbei wurde die Phagozytosefähigkeit anhand von Zelllysat und apoptotischen Tumorzellen und die Makropinozytosefähigkeit anhand von Dextran Partikeln analysiert. Es konnte gezeigt werden, dass weder PGE2 noch spezifische EP Rezeptoragonisten die Antigenaufnahme signifikant beeinflussten.
Durch Präsentation von Antigen auf MHC-II-Molekülen sind DC in der Lage, tumorspezifische CD4+ T-Zellen zu aktivieren. Die MHC-II Präsentationsfähigkeit der MoDC unter dem Einfluss von PGE2 wurde mit NY-ESO-1157-170 Peptid, NY-ESO-1 Protein sowie mit NY ESO 1-transfiziertem CHO Zelllysat als Antigenquellen untersucht. PGE2 wurde zu den MoDC entweder 24 h vor (Präinkubation) oder zur gleichen Zeit wie das Antigen (Simultaninkubation) hinzugefügt. Die Versuche ergaben, dass die MHC-II-Antigenpräsentation der MoDC durch PGE2 nicht signifikant beeinflusst wird.
Die Auswirkung von PGE2 auf die Kreuzpräsentationsfähigkeit der MoDC wurde mit einer Formulierung aus NY-ESO-1 Protein und dem ISCOMATRIX® adjuvant (NY ESO-1/IMX) und einem Immunkomplex bestehend aus NY-ESO-1 Protein und dem Antikörper anti NY ESO-1 mAk (Klon ES121; NY-ESO-1/IC) untersucht. Hierbei konnte gezeigt werden, dass sowohl die Kreuzpräsentation von NY-ESO-1/IMX als auch von NY ESO 1/IC durch PGE2 dosisabhängig signifikant gehemmt wurde. Dieser Effekt wird, ähnlich wie die Zell-Reifung, über die EP2/4-Rezeptoren vermittelt.
Zusammenfassend erlauben die Ergebnisse den Schluss, dass PGE2 über einen EP2/4-Rezeptor-vermittelten Mechanismus die Kreuzpräsentationsfähigkeit der MoDC inhibiert, ohne jedoch die Antigenaufnahme oder die MHC-II Präsentation signifikant zu beeinflussen. Eine klinische Relevanz der Ergebnisse besteht bei der Verwendung von DC für therapeutische Tumorvakzinierungen zur Induktion einer gegen Malignome gerichteten Immunantwort. Die Benutzung von PGE2 als Reifestimulus für MoDC, die mit Protein-Antigenen gepulst werden, birgt das Risiko, die Induktion von tumorantigenspezifischen CD8+ T-Zellen negativ zu beeinflussen

Micropropagation supports reintroduction of an apulian artichoke landrace in sustainable cropping systems
Artichoke (Cynara cardunculus L. var. scolymus (L.) Fiori) is a perennial plant of the Aster-aceae’s family native to the Mediterranean basin. Italy has rich artichoke biodiversity, but many landraces are subjected to genetic erosion caused by increasing use of commercial varieties that are more homogenous in production. An Apulian landrace ‘Troianella’ was established in vitro to valorize and provide high-quality material for propagation in nurseries and, subsequently, for cultivation in production fields. The shoot proliferation was tested on four different growth media, adding cytokinin (-6-benzylamminopurine (BAP-0.05 mg L−1 ). Among these, the best results were achieved on MS519-A and BM media in which MS macronutrients were supplemented with additional doses of CaCl2 and MgSO4 (plus 120 mg L−1 and 190 mg L−1, respectively). In vitro root induction was obtained with 10 mg L−1 of Indole-3-acetic acid (IAA) and 30 g L−1 of sucrose. Plants derived from tissue culture were acclimatized in greenhouse using mycorrhizal symbiosis to increase survival during the acclimatization phase and to improve their performance after transplanting in field. Three arbuscular mycorrhizal (AM) fungi (Septoglomus viscosum, Funelliformis mosseae, and Symbivit, a commercial mix) were added to a sterile substrate and compared to a sterile control without any AM fungal inocula. After 3 months, the best growth and plant appearance were on substrates with S. viscosum fungus or the commercial mycorrhizal fungi mix. The results supported a development of an efficient micropropagation protocol and a production of high quality plant material for sustainable farming of the endangered ’Troianella’ landrace
Genomic Resources for Asparagales
Enormous genomic resources have been developed for plants in the monocot order Poales; however, it is not known how useful these resources will be for other economically important monocots. Asparagales are a monophyletic order sister to class Commelinanae that carries Poales, and is the second most economically important monocot order. Development of genomic resources for and their application to Asparagales are challenging because of huge nuclear genomes and the relatively long generation times required to develop segregating families. We synthesized a normalized eDNA library of onion (Allium cepa) and produced II ,008 unique expressed sequence tags (ESTs) for comparative genomic analyses of Asparagales and Poales. Alignments of onion ESTs, Poales ESTs, and genomic sequences from rice were used to design oligonucleotide primers amplifying genomic regions from asparagus, garlic, and onion. Sequence analyses of these genomic regions revealed microsatellites, insertions/deletions, and single nucleotide polymorphisms for comparative mapping of rice and Asparagales vegetables. Initial mapping revealed no obvious synteny at the recombinationallevel between onion and rice, indicating that genomic resources developed for Poales may not be applicable to the monocots as a whole. Genomic analyses of Asparagales would greatly benefit from EST sequencing and deep-coverage, large-insert genomic libraries of representative small-genome model species within the higher and lower Asparagales, such as asparagus and orchid, respectively
Criopreservação de embriões zigóticos de coqueiro Anão Verde do Brasil de Jiqui
The objective of this work was to evaluate rewarming procedures and recovery media for Brazilian Green Dwarf coconut (Cocos nucifera) embryos cryopreserved by vitrification. The rewarming procedures evaluated were: T1, water bath at 38±2°C; and T2, unloading solution consisting of Y3 medium + 1.2 mol L-1 sucrose. The recovery media assessed were: M1, Y3 medium + 45 g L-1 sucrose + 1 mg L-1 gibberellic acid + 1 g L-1 activated charcoal + 2.2 g L-1 Gelrite; and M2, Y3 medium + 60 g L-1 sucrose + 2.2 g L-1 Gelrite. Rewarming procedures showed no significant effect, and the M1 media induced 72.5% regeneration.O objetivo deste trabalho foi avaliar processos de descongelamento e meios de regeneração para embriões zigóticos de coqueiro Anão Verde do Brasil de Jiqui (Cocos nucifera) criopreservados por vitrificação. Avaliaram-se os processos de descongelamento: T1, banho-maria a 38±2°C; e T2, solução de descarregamento composta de meio Y3 + 1,2 mol L-1 de sacarose. Os meios de regeneração analisados foram: M1, meio Y3 + 45 g L-1 de sacarose + 1 mg L-1 de ácido giberélico + 1 g L-1 de carvão ativado + 2,2 g L-1 de Gelrite; e M2, meio Y3 + 60 g L-1 de sacarose + 2,2 g L-1 de Gelrite. O descongelamento não apresentou efeito significativo, e o meio M1 induziu 72,5% de regeneração
Sterilization procedures and Plant Preservative Mixture on in vitro establishment of Miscanthus sinensis Andersson.
Information about the establishment phase of Miscanthus sinensis Andersson as sterile procedure to avoid the contamination were few reported. The aim of this study was to evaluate different immersion sterilization procedures with and without the biocide, Plant Preservative Mixture (PPM?) on in vitro contamination of two Miscanthus genotypes. The plant material were canes from adult plants of two accessions PI 668371 and PI 668375 cultivated in the germplasm bank of USDA-ARS, Griffin, Georgia. Apical meristems were submitted to immersion sterilization procedures (70% isopropyl alcohol and 1.5, 5.0 or 8.25% NaOCl for different time periods, followed by rinsing three times with autoclaved distilled water. After that, the explants were inoculated on MS basal medium in the presence or absence of 1 mL L-1 PPM?. The experimental design was fully randomized in a 2 x 5 x 2 factorial scheme (genotypes x sterilization process x PPM?) with five replicates per treatment and five apical meristems per experimental unit. Aseptic treatments showed no differences for percentage of bacterial and fungal contaminations and percentage of explant survival. In the presence of PPM ? there was less bacterial contamination in the accessions (28%) than in the absence (100%). For the percentage of fungal contamination, PPM ? had no significant effect on PI 668371 accession. However, on PI 668375 accession there was a lower percentage of fungal contamination (16%). The biocide PPM ? may be an efficient agent to prevent bacterial contamination on in vitro cultures of Miscanthus spp and fungal contamination in PI 668375 accession
