    Effects of Dietary Protein and Fat Content on Intrahepatocellular and Intramyocellular Lipids during a 6-Day Hypercaloric, High Sucrose Diet: A Randomized Controlled Trial in Normal Weight Healthy Subjects.

    Sucrose overfeeding increases intrahepatocellular (IHCL) and intramyocellular (IMCL) lipid concentrations in healthy subjects. We hypothesized that these effects would be modulated by diet protein/fat content. Twelve healthy men and women were studied on two occasions in a randomized, cross-over trial. On each occasion, they received a 3-day 12% protein weight maintenance diet (WM) followed by a 6-day hypercaloric high sucrose diet (150% energy requirements). On one occasion the hypercaloric diet contained 5% protein and 25% fat (low protein-high fat, LP-HF), on the other occasion it contained 20% protein and 10% fat (high protein-low fat, HP-LF). IHCL and IMCL concentrations (magnetic resonance spectroscopy) and energy expenditure (indirect calorimetry) were measured after WM, and again after HP-LF/LP-HF. IHCL increased from 25.0 ± 3.6 after WM to 147.1 ± 26.9 mmol/kg wet weight (ww) after LP-HF and from 30.3 ± 7.7 to 57.8 ± 14.8 after HP-LF (two-way ANOVA with interaction: p < 0.001 overfeeding x protein/fat content). IMCL increased from 7.1 ± 0.6 to 8.8 ± 0.7 mmol/kg ww after LP-HF and from 6.2 ± 0.6 to 6.9 ± 0.6 after HP-LF, (p < 0.002). These results indicate that liver and muscle fat deposition is enhanced when sucrose overfeeding is associated with a low protein, high fat diet compared to a high protein, low fat diet

    SARS Antibody Testing in Children: Development of Oral Fluid Assays for IgG Measurements.

    Seroepidemiological studies to monitor antibody kinetics are important for assessing the extent and spread of SARS-CoV-2 in a population. Noninvasive sampling methods are advantageous for reducing the need for venipuncture, which may be a barrier to investigations, particularly in pediatric populations. Oral fluids are obtained by gingiva-crevicular sampling from children and adults and are very well accepted. Enzyme immunoassays (EIAs) based on these samples have acceptable sensitivity and specificity compared to conventional serum-based antibody EIAs and are suitable for population-based surveillance. We describe the development and evaluation of SARS-CoV-2 IgG EIAs using SARS-CoV-2 viral nucleoprotein (NP) and spike (S) proteins in IgG isotype capture format and an indirect receptor-binding-domain (RBD) IgG EIA, intended for use in children as a primary endpoint. All three assays were assessed using a panel of 1,999 paired serum and oral fluids from children and adults participating in school SARS-CoV-2 surveillance studies during and after the first and second pandemic wave in the United Kingdom. The anti-NP IgG capture assay was the best candidate, with an overall sensitivity of 75% (95% confidence interval [CI]: 71 to 79%) and specificity of 99% (95% CI: 78 to 99%) compared with paired serum antibodies. Sensitivity observed in children (80%, 95% CI: 71 to 88%) was higher than that in adults (67%, CI: 60% to 74%). Oral fluid assays (OF) using spike protein and RBD antigens were also 99% specific and achieved reasonable but lower sensitivity in the target population (78%, 95% CI [68% to 86%] and 53%, 95% CI [43% to 64%], respectively). IMPORTANCE We report on the first large-scale assessment of the suitability of oral fluids for detection of SARS-CoV-2 antibody obtained from healthy children attending school. The sample type (gingiva-crevicular fluid, which is a transudate of blood but is not saliva) can be self collected. Although detection of antibodies in oral fluids is less sensitive than that in blood, our study suggests an optimal format for operational use. The laboratory methods we have developed can reliably measure antibodies in children, who are able to take their own samples. Our findings are of immediate practical relevance for use in large-scale seroprevalence studies designed to measure exposure to infection, as they typically require venipuncture. Overall, our data indicate that OF assays based on the detection of SARS-CoV-2 antibodies are a tool suitable for population-based seroepidemiology studies in children and highly acceptable in children and adults, as venipuncture is no longer necessary

    Defining Terms Used for Animals Working in Support Roles for People with Support Needs

The same term may be used to describe different roles, or two terms may mean the same thing. This confusion is evident among researchers, practitioners, and end users. Because certain animal roles are provided with legal protections and/or government-funding support in some jurisdictions, it is necessary to clearly define the existing terms to avoid confusion. The aim of this paper is to provide operationalized definitions for nine terms, which would be useful in many world regions: "assistance animal", "companion animal", "educational/school support animal", "emotional support animal", "facility animal", "service animal", "skilled companion animal", "therapy animal", and "visiting/visitation animal". At the International Society for Anthrozoology (ISAZ) conferences in 2018 and 2020, over 100 delegates participated in workshops to define these terms, many of whom co-authored this paper. Through an iterative process, we have defined the nine terms and explained how they differ from each other. We recommend phasing out two terms (i.e., "skilled companion animal" and "service animal") due to overlap with other terms that could potentially exacerbate confusion. The implications for several regions of the world are discussed

    A review of the management and treatment of brine solutions

    There has been a considerable expansion in the use of desalination plants for supplying potable water to many communities in the world. However, the production of brine, which is known to be a concentrated solution, is an integral part of the operation of the desalination plant, which faces critical environmental challenges because of its high salinity. Therefore, a cost-effective and environmentally benign brine management system is required prior to its proper disposal. Currently, several disposal options including surface water discharge, deep well injection, and evaporation ponds have been used. However, these processes are unsustainable and their use is limited by a high capital cost and limited applications. Various conventional treatments including physicochemical, oxidation and biological processes with varying extents of organics removal have been reported. Nowadays, membrane-based technologies appear to be cost-effective methods for managing brine concentrate as they can recover valuable resources and produce clean water with high recovery. This paper critically reviews current technologies for concentrate management, including disposal practices and treatment processes. The characteristics of the brine in terms of water quality and its impact on open water bodies are discussed. This paper examines emerging membrane processes such as forward osmosis, membrane distillation, and electrodialysis that show promise for minimizing brine volume, in recovering valuable resources and improvement of water recovery. This paper also discusses the fact that integrated membrane processes are better for brine management for resource recovery together with water purification in wastewater treatment facilities and can achieve a zero liquid discharge

    [Extract] The world population started to increase at a faster rate from the early 1800s which inevitably led to the generation of the wastes in the forms of the solids, liquids and gases. These wastes contributed significantly to the ill health of the population since then and forced scientists, microbiologists and others to find ways to treat them. Fig. 1 shows the population increase in every fifty years since 1800 along with scientific findings that are related to the improvement of human health. The figure also shows the industrial development that occurred alongside which obviously deteriorated the environment further. The naturally occurring aerobic and anaerobic processes in carbon, nitrogen, phosphorus, and sulphur cycles helped the scientists to further their knowledge on treating all the three forms (solids, liquid and gaseous) of the wastes to reduce their impact on the environment as well as the health of the humans and other living organisms
