    Evaluation of the possible role of HPV16, CMV and EBV in Cervical Carcinoma progression using In Situ Hybridization technique

    Background:   Cervical carcinoma was considered as a major problem and life threaten of women; therefore it is worthy to study the association of Epstein–Barr virus and cytomegalovirus co-infection with human papillomavirus type 16 in uterine cervical carcinoma progression. Cervical carcinoma is known closely associated with human papillomavirus. Objective: To identify whether Epstein–Barr virus and cytomegalovirus play a co-factors role in the cervical carcinogenesis besides human papillomavirus type 16 infection.            Patients and Methods: Current study included paraffin embedded sections from 50 cases of cervical cancer in Baghdad during the period from 2012 till 2014. They were examined for the presence of human papillomavirus type 16, cytomegalovirus and Epstein-Barr virus DNA using in situ hybridization technique. Results: This study showed that (33, 66%), (22, 44%) and (38, 76%) out of 50 cervical cancer specimens were positive to human papillomavirus type 16, cytomegalovirus and Epstein–Barr virus DNAs signals respectively by using in situ hybridization technique. Conclusion: The co-operation effects of CMV and EBV onto HPV16 might progress the oncogenesis of cervical carcinoma in female patients

    A Cross Sectional Study of Knowledge and attitudes levels about Polio Vaccination among selected sample of Technical Institute \ AL-Suwaira staff members

    Background: The previous laboratory investigations confirmed local polio case was informed on 28th  January 2000 and another two wild polio cases were reported from Resafa \ Baghdad province in 2014. The date of onset of these cases were February and April 2014 and both cases were connected to 2013 of Syria outbreak. Objective:To evaluate the knowledge and attitudes levels towards polio vaccination among staff of Technical Institute - AL-Suwaira. Patients and Methods: A study of cross-sectional design consisted of 170 a selected samples of Technical Institute \ AL-Suwaira staff members. Collected during January 2018.The knowledge and attitudes levels estimated by using typical design of self-administered questionnaire. Descriptive statistics were used to explain the results. Results: Over all, 170 participants reacted to the questionnaire by a response rate of 93.5%. All respondents established a good knowledge and positive attitudes levels to poliovirus vaccination. Mean knowledge score (built on11 knowledge questions) and mean attitude score (built on 10 attitudes statements) of participants about polio vaccine were 14.45±2.38 and 26.5±4.5 respectively. The mainstream of respondents agreed that all children should be taken the polio vaccine (88.2%), and the sterility concerns that associated with polio vaccines (14.1%). Television (n=126, 39%) and Internet (n = 66, 21%) were the main social media sources that used by staff members to educate and modify themselves about polio vaccination.  Knowledge defects were recognized in the management issues and how to dealing with vaccines (45.9%), the incubation period of poliovirus infection (38.8%) and the consequences of the poliovirus infection (56.5%) on the victims. Conclusion: Staff members of Technical Institute \ AL-Suwaira showed a good knowledge and positive attitudes levels about polio vaccination. Even though the statistics are indicative of knowledge gaps of participants, further studies are needed to assess these levels for better education program against polio

    Seroprevalence of Toxoplasma gondii in human and chickens (Gallus domesticus) with histological effects on the internal chicken organs in AL-Door district, Salahdin province

    The present study included seroprevalence detection of Toxoplasma gondii in human and chickens (Gallus domesticus) in AL-Door district, Salahdin province, and the histological effects of T. gondii on some of internal organs (brain, kidneys, liver and muscles) from chickens. Positive seroprevalence result for human toxoplasmosis was recorded 22.3% with highest prevalence for positive IgG that reported 14.1%. Based on gender, significant increasing was recorded for toxoplasmosis in males 27.3% when compared with female 12.7%, while according to age, significant variances were reported among the age groups, the highest seroprevalence was 27.8% in age group 31-40 years. For seroprevalence of chicken toxoplasmosis, the percentage of positive results was 66.7%. Different histological effects were recorded in the current study such as congested blood vessels in the brain cortex tissue with vacuolation in the outer regions of brain, atrophied glomeruli in the renal cortex, masses of hepatocytes with presence of Kupffer cells, also cellular aggregation of fibroblast and WBCs in muscles

    Subsequently, the early reactivation or old human cytomegalovirus (HCMV) infection may be interfere with IgE and interleukin-33 (IL-33) levels disturbances throughout the life intervals and act as synergistic factors to play a significant role in the pathogenesis of various diseases; like asthma or coronary heart diseases (CHD). The main subject of the current study was to determine the role of CMV infection onto IgE levels and IL-33 disturbances in the pathogenesis of asthma and atherosclerosis as a case-control study. A total of 175 participants were with a mean age of 38.17 ± 7.51 years [age range: 26–52] years, which were distributed as patients with atherosclerosis (n=50) and asthmatic patients (n=50) as the cases of our study and 75 healthy subjects as a control group. Both groups of asthmatic and atherosclerosis patients show a significant association with the seropositivity of CMV-IgG which was detected in 33 (18.9%) and 30 (17.1%). While, CMV-PP65 Ag which was detected in 26 (14.9%) and 28 (16.0%) respectively (P < 0.0001). High level of abnormal IL-33 was detected in 36 (20.6%) with 45 (25.7%) of abnormal IgE was determined in patients with asthma followed by 26 (14.9%) and 44 (25.1%) for abnormal levels of IL-33 and IgE respectively in patients with atherosclerosis. Furthermore, there was a positive significant association between the seropositivity of CMV-IgG and CMV-PP65 Ag with abnormal IgE and IL-33 levels in the pathogenesis of asthma and atherosclerosis (P < 0.0001), and impact of our study support the hypothesis that previous cytomegalovirus infection or early reactivation with unregulated IL-33 expression or high IgE level play a significant role in the pathogenesis of coronary heart diseases and asthma

    IMPROVING THE ANTIBACTERIAL ACTIVITY BY THE COMBINATION OF ZIRCONIUM OXIDE NANOPARTICLES (ZrO2) AND CEFTAZIDIME AGAINST KLEBSIELLA PNEUMONIAE

    Introduction: Klebsilla pneumoniae is one of must opportunistic pathogens that causes nosocomial infection, UTI, respiratory tract infections and blood infections. ZrO2 nanoparticles have antimicrobial activity against some pathogenic bacteria and fungi. Ceftazidime is one of third generation cephalosporins groups of antibiotecs, characterized by its broad spectrum on bacteria in general and particularly on Enterobacteriaceae family like Klebsiella spp. Method: Diverse clinical samples of Klebsilla pneumoniae were isolated from several hospitals in Baghdad – Iraq and ZrO2 nanoparticles was investigated against it. Ceftazidime was also investigated against K. pneumoniae. Both of ZrO2 nanoparticles and ceftazidime were mixed together and investigated against K. pneumoniae. Results: The result showed that ZrO2 nanoparticles were effectivity on inhibiting opportunistic pathogens. By using zirconium oxide nanoparticles on Klebsiella pneumonia isolates in 24h. of incubation time, inhibition zones were (38,34,10,10,8,0) mm respectively on agar plates. By using ceftazidime alone against the same bacteria inhibition zones were (40,32,10,9,8,0) mm. respectively. Conclusion:The present study results that the antibacterial activity of ceftazidime against bacteria was increased when combination between ZrO2 nanoparticles and the antibiotic had done, because, inhibition zones in case of mixing both of ZrO2 nanoparticles and ceftazidime were (43,40,12,12,10,0) mm respectively. So that we can conclude that the combination of zirconium oxide nanoparticles (ZrO2) and ceftazidime was a useful method for the treatment of Klebsilla pneumonia that cause nosocomial infection, UTI, respiratory tract infections and blood infections

    PURIFICATION OF INULINASE FROM KLEBSIELLA PNEUMONIAE AND STUDY THE ANTIBACTERIAL EFFECT OF COMBINATION OF INULINASE AND CEFTAZIDIME

    Introduction: Klebsiella pneumoniae are Gram-negative which cause many diseases such as urinary tract infections, respiratory tract infections and septicemia. Inulinase is an enzyme used in food manufacture and pharmaceuticals. Inulinase is used in decreasing lipid ratio and, cholesterol in blood and considered as a prebiotic factor inside intestine. Many microorganisms can produce inulinase, such as yeast, fungi and bacteria; among such bacteria: Bacillus spp., Arthrobacter spp., and Pseudomonas spp. but there are no studies about inulinase production by K. pneumoniae have been reported. So the current study aims at investing the ability of producing and purification inulinase by K. pneumoniae. Method: K. pneumoniae were isolated from many hospitals and screened for the production of inulinase. Isolation percentage was 32%. A combination between the enzyme and the ceftazidime were assayed for detecting the antibacterial activity agonist Gram positive and Gram negative bacteria were done.  Results:  It is found that K. pneumoniae K4 isolate is the best producer of this enzyme. Inulinase, purified with ammonium sulfate at 70% saturation with specific activity 7.01 U/mg protein. As well, it's found that inulinase had increased the activity of ceftazidime against bacteria when combination between this enzyme and the antibiotic had done. Conclusion: This study proves for the first time that K. pneumoniae can produce inulinase which can be used in tremendous applications and also proves the broad spectrum bioactivity of inulinase against microbial pathogens.
