    The experience of loneliness among international students participating in the BBC Loneliness Experiment: Thematic analysis of qualitative survey data

    Loneliness is associated with poor mental and physical health in the general population. It is thought to be prevalent among international students, but few studies have investigated this. To capture what loneliness means in the everyday lives of international students we analysed data from 521 international students aged 16–40 years who participated online in the 2018 British Broadcasting Corporation (BBC) Loneliness Experiment. Using the approach of thematic analysis of free‐text responses to a question probing the perceived meaning of loneliness, we identified six main themes: (1) Negative psychological and social aspects of loneliness; (2) The distressing experience of being alone; (3) Disrupted ability to make meaningful connections; (4) Sense of entrapment in a state of loneliness; (5) Awareness of others’ stigmatizing perceptions of oneself as lonely; (6) Perceptions of positive aspects of loneliness. These findings identified a range of negative social and psychological dimensions of loneliness in international students in relation to their well-being, emotional health, and social functioning, hampering the benefits to be gained from their overseas study. Higher education teaching staff, pastoral staff, and mental health professionals need to be more aware of the prevalence and impacts of loneliness and should tackle it in culturally appropriate ways

    The long-lived queen : reproduction and longevity in female eusocial Damaraland mole-rats (Fukomys damarensis)

    The inverse relationship between reproduction and lifespan is one of the main concepts of life history theory. This association has been observed in most taxa, although exceptions have been found in which a breeding female outlives her non-reproductive cohorts. This relationship is well known in social insects, and it has recently come to light that reproductive females of certain social mole-rat species also exhibit extended lifespans relative to non-breeders. We analysed mark–recapture data over 13 years in 12 colonies of Damaraland mole-rats (Fukomys damarensis) to assess if colony queens lived longer than non-reproductive adult females. Queens were recaptured up to 8.5 years after initial capture (X = 6.2 years); significantly longer than non-reproductive females (X = 1.3 years), suggesting that a colony queen lives longer than her non-reproductive subordinates.     Rare Genetic Variation in 135 Families With Family History Suggestive of X-Linked Intellectual Disability.

    Families with multiple male children with intellectual disability (ID) are usually suspected of having disease due to a X-linked mode of inheritance and genetic studies focus on analysis of segregating variants in X-linked genes. However, the genetic cause of ID remains elusive in approximately 50% of affected individuals. Here, we report the analysis of next-generation sequencing data in 274 affected individuals from 135 families with a family history suggestive of X-linked ID. Genetic diagnoses were obtained for 19% (25/135) of the families, and 24% (33/135) had a variant of uncertain significance. In 12% of cases (16/135), the variants were not shared within the family, suggesting genetic heterogeneity and phenocopies are frequent. Of all the families with reportable variants (43%, 58/135), we observed that 55% (32/58) were in X-linked genes, but 38% (22/58) were in autosomal genes, while the remaining 7% (4/58) had multiple variants in genes with different modes on inheritance. This study highlights that in families with multiple affected males, X linkage should not be assumed, and both individuals should be considered, as different genetic etiologies are common in apparent familial cases

    Optimization and Lead Selection of Benzothiazole Amide Analogs Toward a Novel Antimycobacterial Agent

    Mycobacteria remain an important problem worldwide, especially drug resistant human pathogens. Novel therapeutics are urgently needed to tackle both drug-resistant tuberculosis (TB) and difficult-to-treat infections with nontuberculous mycobacteria (NTM). Benzothiazole adamantyl amide had previously emerged as a high throughput screening hit against M. tuberculosis (Mtb) and was subsequently found to be active against NTM as well. For lead optimization, we applied an iterative process of design, synthesis and screening of several 100 analogs to improve antibacterial potency as well as physicochemical and pharmacological properties to ultimately achieve efficacy. Replacement of the adamantyl group with cyclohexyl derivatives, including bicyclic moieties, resulted in advanced lead compounds that showed excellent potency and a mycobacteria-specific spectrum of activity. MIC values ranged from 0.03 to 0.12 ÎŒg/mL against M. abscessus (Mabs) and other rapid- growing NTM, 1–2 ÎŒg/mL against M. avium complex (MAC), and 0.12–0.5 ÎŒg/mL against Mtb. No pre-existing resistance was found in a collection of n = 54 clinical isolates of rapid-growing NTM. Unlike many antibacterial agents commonly used to treat mycobacterial infections, benzothiazole amides demonstrated bactericidal effects against both Mtb and Mabs. Metabolic labeling provided evidence that the compounds affect the transfer of mycolic acids to their cell envelope acceptors in mycobacteria. Mapping of resistance mutations pointed to the trehalose monomycolate transporter (MmpL3) as the most likely target. In vivo efficacy and tolerability of a benzothiazole amide was demonstrated in a mouse model of chronic NTM lung infection with Mabs. Once daily dosing over 4 weeks by intrapulmonary microspray administration as 5% corn oil/saline emulsion achieved statistically significant CFU reductions compared to vehicle control and non-inferiority compared to azithromycin. The benzothiazole amides hold promise for development of a novel therapeutic agent with broad antimycobacterial activity, though further work is needed to develop drug formulations for direct intrapulmonary delivery via aerosol

    Erosion characteristics and floc strenght of Athabasca river cohesive sediments: towards managing sediment-related issues

    Purpose: Most of Canada’s tar sands exploitations are located in the Athabasca river basin. Deposited cohesive sediments in Athabasca river and tributaries are a potential source of PAHs in the basin. Erosional behavior of cohesive sediments depends not only of fluid turbulence but on sediments structure and particularly the influence of organic content. This research tries to describe this behavior in Athabasca river sediments. Methods: An experimental study of cohesive sediments dynamics in one of the tributaries, the Muskeg river, was developed in a rotating annular flume. Variation of the shear stress allowed the determination of erosional strength for beds with different consolidation periods. Particle size measurements were made with a laser diffraction device operated in a continuous flow through mode. Optical analyses of flocs (ESEM and TEM) were performed with samples taken at the end of the experiments. Results: An inverse relationship between suspended sediment concentration (SS) and the consolidation period was found. The differences are related in this research to the increasing organic content of the sediments with consolidation period. The particle size measurements during the experiments showed differences on floc strength that are also related to changing organic content during different consolidation periods. ESEM and TEM observations confirm the structural differences for beds with different consolidation periods. The effects of SFGL on floc structure and in biostabilization of the bed are discussed. Conclusions: It is recommended in this paper that consolidation period should be taken into account for the modeling of erosion of cohesive sediments in the Athabasca river. Relating to transport models of pollutants (PAHs) it is highly recommended to consider flocs organic content, particularly algae, in the resuspension module.Environment Canada, CONACY

    Appropriateness of acute admissions and last in-patient day for patients with long term neurological conditions

    <p>Abstract</p> <p>Background</p> <p>To examine the appropriateness of admissions and in-patient stay for patients with long term neurological conditions (LTNCs). To identify variables predictive of appropriateness and explore management alternatives.</p> <p>Methods</p> <p>Adults admitted as acute patients to Derby Hospitals NHS Foundation Trust (England). Data were collected prospectively and examined by a multi-disciplinary expert panel to determine the appropriateness of admission and length of stay (LoS). Management alternatives were discussed.</p> <p>Results</p> <p>A total of 119 participants were recruited. 32 admissions were inappropriate and 83 were for an inappropriate duration. Whether a participant lived in their own home was predictive of an inappropriate admission. The number of LTNCs, number of presenting complaints and whether the participant lived alone in their own home were predictive of an inappropriate LoS. For admissions judged to be inappropriate, the panel suggested management alternatives.</p> <p>Conclusion</p> <p>Patients with LTNCs are being admitted to hospital when other services, e.g. ambulatory care, are available which could meet their needs. Inefficiencies in hospital procedures, such as discharge planning and patient transfers, continue to exist. Recognition of the need to plan for discharge at admission and to ensure in-patient services are provided in a timely manner may contribute towards improved efficiency.</p

    Extended Longevity of Reproductives Appears to be Common in Fukomys Mole-Rats (Rodentia, Bathyergidae)

    African mole-rats (Bathyergidae, Rodentia) contain several social, cooperatively breeding species with low extrinsic mortality and unusually high longevity. All social bathyergids live in multigenerational families where reproduction is skewed towards a few breeding individuals. Most of their offspring remain as reproductively inactive “helpers” in their natal families, often for several years. This “reproductive subdivision” of mole-rat societies might be of interest for ageing research, as in at least one social bathyergid (Ansell's mole-rats Fukomys anselli), breeders have been shown to age significantly slower than non-breeders. These animals thus provide excellent conditions for studying the epigenetics of senescence by comparing divergent longevities within the same genotypes without the inescapable short-comings of inter-species comparisons. It has been claimed that many if not all social mole-rat species may have evolved similar ageing patterns, too. However, this remains unclear on account of the scarcity of reliable datasets on the subject. We therefore analyzed a 20-year breeding record of Giant mole-rats Fukomys mechowii, another social bathyergid species. We found that breeders indeed lived significantly longer than helpers (ca. 1.5–2.2fold depending on the sex), irrespective of social rank or other potentially confounding factors. Considering the phylogenetic positions of F. mechowii and F. anselli and unpublished data on a third Fukomys-species (F. damarensis) showing essentially the same pattern, it seems probable that the reversal of the classic trade-off between somatic maintenance and sexual reproduction is characteristic of the whole genus and hence of the vast majority of social mole-rats
