Cooperative DNA-binding by Bicoid provides a mechanism for threshold-dependent gene activation in the Drosophila embryo
The Bicoid morphogen directs pattern formation along the anterior-posterior (A-P) axis of the Drosophila embryo. Bicoid is distributed in a concentration gradient that decreases exponentially from the anterior pole, however, it transcribes target genes such as hunchback in a step-function-like pattern; the expression domain is uniform and has a sharply defined posterior boundary. A 'gradient-affinity' model proposed to explain Bicoid action states that (i) cooperative gene activation by Bicoid generates the sharp on/off switch for target gene transcription and (ii) target genes with different affinities for Bicoid are expressed at different positions along the A-P axis. Using an in vivo yeast assay and in vitro methods, we show that Bicoid binds DNA with pairwise cooperativity; Bicoid bound to a strong site helps Bicoid bind to a weak site. These results support the first aspect of the model, providing a mechanism by which Bicoid generates sharp boundaries of gene expression. However, contrary to the second aspect of the model, we find no significant difference between the affinity of Bicoid for the anterior gene hunchback and the posterior gene knirps, We propose, instead, that the arrangement of Bicoids bound to the target gene presents a unique signature to the transcription machinery that, in combination with overall affinity, regulates the extent of gene transcription along the A-P axis
Stimulation of the pituitary-adrenal axis with arginine vasopressin in patients with depression
The ACTH response to arginine vasopressin was the same in patients with depression while cortisol response was significantly greater in patients with depression when compared to the control population. Measurement of p + d -> 3He + eta in S(11) Resonance
We have measured the reaction p + d -> 3He + eta at a proton beam energy of
980 MeV, which is 88.5 MeV above threshold using the new ``germanium wall''
detector system. A missing--mass resolution of the detector system of 2.6% was
achieved. The angular distribution of the meson is forward peaked. We found a
total cross section of (573 +- 83(stat.) +- 69(syst.))nb. The excitation
function for the present reaction is described by a Breit Wigner form with

Multiple steroid hormone levels in depressed patients and normal controls before and after exogenous ACTH
Forty depressed patients and 36 age- and sex-matched controls were given 250 [mu]g ACTH1-24 by bolus. Plasma steroid hormone levels were measured prior to and 60 min after ACTH administration. The depressed patients had significantly greater cortisol (F), 11-deoxycortisol (S), androstenedione (AD), and 17[alpha]-hydroxyprogesterone (17[alpha]-OHP) responses (delta; pp=0.091) than the controls. There was no significant difference in the corticosterone (B) response between the two groups.With the exception of 11[beta]-OHAD, all the steroid hormones were significantly negatively correlated with age in the controls, but only S and AD marginally demonstrated this relationship in the depressed patients. F, S, AD, 17[alpha]-OHP, and B, but not 11[beta]-OHAD, were significantly positively correlated with each other in the controls, but only F was significantly correlated with AD in the depressed patients. These data suggest that the hypercortisolemia found in some depressed patients involves increased precursor and metabolite levels both at baseline and in response to exogenous ACTH, compared to controls. Furthermore, variability in these precursors is greater in depressed patients, and their relationship to age is lost. Detailed comparison of the pp -> \pi^+pn and pp -> \pi^+d reactions at 951 MeV
The positively charged pions produced in proton-proton collisions at a beam
momentum of 1640 MeV/c were measured in the forward direction with a high
resolution magnetic spectrograph. The missing mass distribution shows the bound
state (deuteron) clearly separated from the continuum. Despite the very
good resolution, there is no evidence for any significant production of the
system in the spin-singlet state. However, the cross section ratio is about twice as large as
that predicted from -wave final-state-interaction theory and it is suggested
that this is due to -state effects in the system.Comment: 8 pages, 3 figure
