    Marker assisted introgression of opaque2 gene into herbicide resistant elite maize inbred lines

    Marker assisted selection in combination with conventional breeding can greatly accelerate the introgression of modified opaque2 genotype into herbicide resistant maize. By combining these two approaches, time and costs are greatly minimized. The application of opaque2 allele specific SSR markers was done on materials already undergoing selection in a breeding program for converting herbicide resistant maize lines into quality protein maize (QPM) which is the equivalent of modified opaque2 phenotype. The breeder had selected QPM lines using the light table in the previous cycle and we used leaf samples to extract DNA for analysis of the presence of the opaque2 gene using SSR markers. Two co-dominant SSR markers phi057 and umc1066 and a dominant marker phi112 were used. Umc1216, a modifier marker was also tested in combination with the opaque2 markers with theobjective of using the marker to select for modifiers for the opaque2 phenotype. The modified FTA paper technology protocol was applied in field sampling. The results showed 97% of the lines wereopaque2 while 3% were non-opaque2. Both methods of conventional breeding using light table and marker assisted selection (MAS) were comparable. However, the application of SSR markers and theFTA technology offers the breeder a fast, time saving, reliable and less labour intensive method of screening QPM maize during the early growing stages instead of having to wait to screen the kernels onthe light table after harvesting. Moreover, the routine biochemical analysis for high lysine and tryptophan levels need not be carried out at each backcross since the presence of the opaque2 gene is confirmed with markers

    Résultats fonctionnels de la chirurgie de la cataracte par phacoalternative avec implantation en chambre postérieure: à propos de 300 cas à Bobo Dioulasso (Burkina Faso)

    La cataracte est la premiÚre cause de cécité curable dans le monde. Son traitement est chirurgical. Le but de notre travail a été d'évaluer les résultats de la phacoalternative ou la chirurgie de la cataracte à petite incision. Il s'est agi d'une étude transversale descriptive à collecte prospective allant du 1er janvier au 31 septembre 2014, chez des patients ùgés d'au moins 40 ans. Les données socio-démographiques, l'acuitévisuelle, l'astigmatisme et les complications ont été évalués.Nous avons inclus 300 yeux de 286 patients. L'ùge moyen était de 66 ans (écart type9,93) avec une prédominance masculine de 57,7%. Les co-morbidités étaient dominées par l'hypertension artérielle 30,33% des cas. L'acuitévisuelle pré-opératoire était de moins de 1/20Ú dans 70, 7% des cas. En biométrie, la puissance moyenne était de 21,50 dioptries. L'implant posé aété adéquat dans 60%. Les principales complications per-opératoires  étaient le chémosis post-anesthésie 4,67% et l'issue de vitrée moins de 2% des cas. Les complications post opératoires précoces ont été dominées par l'oedÚme de cornée 26,33%, et les complications tardives par la cataracte secondaire. L'astigmatisme induit était de 1, 12 dioptrie en  moyenne (écart type 1,26). Sans correction, les résultats visuels étaientmauvais dans moins de 1%, limites dans 31%, et bons 68% suivants les normes de l'Organisation Mondiale de la Santé. La phacoalternative donne des résultats satisfaisants, avec peu de complications. L'amélioration du plateau technique et la disponibilité d'implants adéquats pourraient les améliorer

    Consistent model of magnetism in ferropnictides

    The discovery of superconductivity in LaFeAsO introduced the ferropnictides as a major new class of superconducting compounds with critical temperatures second only to cuprates. The presence of magnetic iron makes ferropnictides radically different from cuprates. Antiferromagnetism of the parent compounds strongly suggests that superconductivity and magnetism are closely related. However, the character of magnetic interactions and spin fluctuations in ferropnictides, in spite of vigorous efforts, has until now resisted understanding within any conventional model of magnetism. Here we show that the most puzzling features can be naturally reconciled within a rather simple effective spin model with biquadratic interactions, which is consistent with electronic structure calculations. By going beyond the Heisenberg model, this description explains numerous experimentally observed properties, including the peculiarities of the spin wave spectrum, thin domain walls, crossover from first to second order phase transition under doping in some compounds, and offers new insight in the occurrence of the nematic phase above the antiferromagnetic phase transition.Comment: 5 pages, 3 figures, revtex

    Spatial distribution and male mating success of Anopheles gambiae swarms

    <p>Abstract</p> <p>Background</p> <p><it>Anopheles gambiae </it>mates in flight at particular mating sites over specific landmarks known as swarm markers. The swarms are composed of males; females typically approach a swarm, and leave <it>in copula</it>. This mating aggregation looks like a lek, but appears to lack the component of female choice. To investigate the possible mechanisms promoting the evolution of swarming in this mosquito species, we looked at the variation in mating success between swarms and discussed the factors that structure it in light of the three major lekking models, known as the female preference model, the hotspot model, and the hotshot model.</p> <p>Results</p> <p>We found substantial variation in swarm size and in mating success between swarms. A strong correlation between swarm size and mating success was observed, and consistent with the hotspot model of lek formation, the <it>per capita </it>mating success of individual males did not increase with swarm size. For the spatial distribution of swarms, our results revealed that some display sites were more attractive to both males and females and that females were more attracted to large swarms. While the swarm markers we recognize help us in localizing swarms, they did not account for the variation in swarm size or in the swarm mating success, suggesting that mosquitoes probably are attracted to these markers, but also perceive and respond to other aspects of the swarming site.</p> <p>Conclusions</p> <p>Characterizing the mating system of a species helps understand how this species has evolved and how selective pressures operate on male and female traits. The current study looked at male mating success of <it>An. gambiae </it>and discussed possible factors that account for its variation. We found that swarms of <it>An. gambiae </it>conform to the hotspot model of lek formation. But because swarms may lack the female choice component, we propose that the <it>An. gambiae </it>mating system is a lek-like system that incorporates characteristics pertaining to other mating systems such as scramble mating competition.</p

    Windborne long-distance migration of malaria mosquitoes in the Sahel

    Over the past two decades efforts to control malaria have halved the number of cases globally, yet burdens remain high in much of Africa and the elimination of malaria has not been achieved even in areas where extreme reductions have been sustained, such as South Africa1,2. Studies seeking to understand the paradoxical persistence of malaria in areas in which surface water is absent for 3–8 months of the year have suggested that some species of Anopheles mosquito use long-distance migration3. Here we confirm this hypothesis through aerial sampling of mosquitoes at 40–290 m above ground level and provide—to our knowledge—the first evidence of windborne migration of African malaria vectors, and consequently of the pathogens that they transmit. Ten species, including the primary malaria vector Anopheles coluzzii, were identified among 235 anopheline mosquitoes that were captured during 617 nocturnal aerial collections in the Sahel of Mali. Notably, females accounted for more than 80% of all of the mosquitoes that we collected. Of these, 90% had taken a blood meal before their migration, which implies that pathogens are probably transported over long distances by migrating females. The likelihood of capturing Anopheles species increased with altitude (the height of the sampling panel above ground level) and during the wet seasons, but variation between years and localities was minimal. Simulated trajectories of mosquito flights indicated that there would be mean nightly displacements of up to 300 km for 9-h flight durations. Annually, the estimated numbers of mosquitoes at altitude that cross a 100-km line perpendicular to the prevailing wind direction included 81,000 Anopheles gambiae sensu stricto, 6 million A. coluzzii and 44 million Anopheles squamosus. These results provide compelling evidence that millions of malaria vectors that have previously fed on blood frequently migrate over hundreds of kilometres, and thus almost certainly spread malaria over these distances. The successful elimination of malaria may therefore depend on whether the sources of migrant vectors can be identified and controlled

    Diversity, dynamics, direction, and magnitude of high-altitude migrating insects in the Sahel

Available from Nature Research via the DOI in this record. Long-distance migration of insects impacts food security, public health, and conservation–issues that are especially significant in Africa. Windborne migration is a key strategy enabling exploitation of ephemeral havens such as the Sahel, however, its knowledge remains sparse. In this first cross-season investigation (3 years) of the aerial fauna over Africa, we sampled insects flying 40–290 m above ground in Mali, using nets mounted on tethered helium-filled balloons. Nearly half a million insects were caught, representing at least 100 families from thirteen orders. Control nets confirmed that the insects were captured at altitude. Thirteen ecologically and phylogenetically diverse species were studied in detail. Migration of all species peaked during the wet season every year across localities, suggesting regular migrations. Species differed in flight altitude, seasonality, and associated weather conditions. All taxa exhibited frequent flights on southerly winds, accounting for the recolonization of the Sahel from southern source populations. “Return” southward movement occurred in most taxa. Estimates of the seasonal number of migrants per species crossing Mali at latitude 14°N were in the trillions, and the nightly distances traversed reached hundreds of kilometers.     Trends in Pretreatment HIV-1 Drug Resistance in Antiretroviral Therapy-naive Adults in South Africa, 2000–2016: A Pooled Sequence Analysis

    Background: South Africa has the largest public antiretroviral therapy (ART) programme in the world. We assessed temporal trends in pretreatment HIV-1 drug resistance (PDR) in ART-naĂŻve adults from South Africa. Methods: We included datasets from studies conducted between 2000 and 2016, with HIV-1 pol sequences from more than ten ART-naĂŻve adults. We analysed sequences for the presence of 101 drug resistance mutations. We pooled sequences by sampling year and performed a sequence-level analysis using a generalized linear mixed model, including the dataset as a random effect. Findings: We identified 38 datasets, and retrieved 6880 HIV-1 pol sequences for analysis. The pooled annual prevalence of PDR remained below 5% until 2009, then increased to a peak of 11·9% (95% confidence interval (CI) 9·2-15·0) in 2015. The pooled annual prevalence of non-nucleoside reverse-transcriptase inhibitor (NNRTI) PDR remained below 5% until 2011, then increased to 10.0% (95% CI 8.4–11.8) by 2014. Between 2000 and 2016, there was a 1.18-fold (95% CI 1.13–1.23) annual increase in NNRTI PDR (p < 0.001), and a 1.10-fold (95% CI 1.05–1.16) annual increase in nucleoside reverse-transcriptase inhibitor PDR (p = 0.001). Interpretation: Increasing PDR in South Africa presents a threat to the efforts to end the HIV/AIDS epidemic. These findings support the recent decision to modify the standard first-line ART regimen, but also highlights the need for broader public health action to prevent the further emergence and transmission of drug-resistant HIV.     © 2014, Orsi. Humic substances (HS) are complex mixtures of natural organic material which are found almost everywhere in the environment, and particularly in soils, sediments, and natural water. HS play key roles in many processes of paramount importance, such as plant growth, carbon storage, and the fate of contaminants in the environment. While most of the research on HS has been traditionally carried out by conventional experimental approaches, over the past 20 years complementary investigations have emerged from the application of computer modeling and simulation techniques. This paper reviews the literature regarding computational studies of HS, with a specific focus on molecular dynamics simulations. Significant achievements, outstanding issues, and future prospects are summarized and discussed
