    Determinants of inspiratory muscle strength in healthy humans

    We investigated 1) the relationship between the baseline and inspiratory muscle training (IMT) induced increase in maximal inspiratory pressure (PI,max) and 2) the relative contributions of the inspiratory chest wall muscles and the diaphragm (Poes/Pdi) to PI,max prior to and following-IMT. Experiment 1: PI,max was assessed during a Müeller manoeuvre before and after 4-wk IMT (n=30). Experiment 2: PI,max and the relative contribution of the inspiratory chest wall muscles to the diaphragm (Poes/Pdi) were assessed during a Müeller manoeuvre before and after 4-wk IMT (n=20). Experiment 1: PI,max increased 19% (P<0.01) post-IMT and was correlated with baseline PI,max (r=−0.373, P<0.05). Experiment 2: baseline PI,max was correlated with Poe/Pdi (r=0.582, P<0.05) and after IMT PI,max increased 22% and Poe/Pdi increased 5% (P<0.05). In conclusion, baseline PI,max and the contribution of the chest wall inspiratory muscles relative to the diaphragm affect, in part, baseline and IMT-induced ΔPI,max. Great care should be taken when designing future IMT studies to ensure parity in the between-subject baseline PI,max

    Inspiratory muscle training abolishes the blood lactate increase associated with volitional hyperpnoea superimposed on exercise and accelerates lactate and oxygen uptake kinetics at the onset of exercise

    We examined the effects of inspiratory muscle training (IMT) upon volitional hyperpnoea-mediated increases in blood lactate ([lac−]B) during cycling at maximal lactate steady state (MLSS) power, and blood lactate and oxygen uptake kinetics at the onset of exercise. Twenty males formed either an IMT (n=10) or control group (n=10). Before and after a 6-wk intervention two 30 min trials were performed at MLSS (207 ± 28 W), which was determined using repeated 30 min constant power trials. The first was a reference trial, whereas during the second trial from 20-28 min participants mimicked the breathing pattern commensurate with 90% of the maximal minute ventilation (V ˙ E) measured during maximal incremental exercise. Before the intervention the MLSS [lac−]B was 3.7 ± 1.8 and 3.9 ± 1.6 mmol·L-1 in the IMT and control group, respectively

    Glacial thermohaline circulation states of the northern Atlantic: The compatibility of modelling and observations

    We present 3He data froma repeat section across Drake Passage, fromthree sections off the South American continent in the Pacific, at 28?S, 35?S, and 43?S, and fromthree sections in the Atlantic, eastward of the Malvinas, close to 35?W, and near the Greenwich Meridian. In Drake Passage, a distinct high-3He signal is observed that is centered just above the boundary of the Lower and the Upper Circumpolar Deep Water (LCDW, UCDW), and is concentrated towards the northern continental slope. 3He concentrations in the Antarctic Circumpolar Current (ACC) upstream of Drake Passage (World Ocean Circulation Experiment section P19 at 88?W) are markedly lower than those found in Drake Passage, and a regional source of primordial helium in the path of the ACC that might cause the high-3He feature can be ruled out. We explain the feature by addition of high-3He waters present at the 43?S Pacific section. This supports a previous, similar interpretation of a low-salinity anomaly in Drake Passage (Naveira Garabato et al., Deep- Sea Research I 49 (2002) 681), that is strongly related to the high-3He feature. Employing multiparameter water mass analysis (including 3He as a parameter), we find that deep waters as met at the 43?S Pacific section, flowing south along the South American continental slope, contribute substantially to the ACC waters in Drake Passage (fractions exceed 50% locally). Lesser, but laterally more extended contributions are found east of the Malvinas, and still smaller ones are present at 35?W and at the Greenwich Meridian. Using velocity measurements from one of the two Drake Passage sections, we estimate the volume transport of these waters to be 7.071.2 Sv, but the average transport may be somewhat lower as the other realization had a less pronounced signal. The enhanced 3He signature in Drake Passage is essentially confined north of the Polar Front. Further downstreamthe signature crosses this front, to the extent that at 35?W the contributions south and north of it are of similar magnitude. At the same time, the 3He levels north of the front are reduced due to a substantial admixture of low-3He North Atlantic Deep Water, such that 3He becomes highest south of the front. The flow of Southeast Pacific deep slope waters entering the ACC constitutes the predominant exit pathway of the primordial helium released in the deep Pacific, and represents a considerable fraction of the deep water return flow fromthe Pacific into the ACC. Therefore and also because the density range of the added deep slope waters is intermediate between those of UCDW and LCDW, they must be considered a distinct water mass. r 2003 Elsevier Ltd.     Reproducibility of the bronchoconstrictive response to eucapnic voluntary hyperpnoea

    Background: Eucapnic voluntary hyperpnoea (EVH) is considered an effective bronchoprovocation challenge for identifying exercise-induced bronchoconstriction (EIB). However, the reproducibility of the hyperpnoea-induced bronchoconstriction (HIB) response elicited by EVH remains unknown and was therefore the focus of this study. Methods: Two cohorts of 16 physically active males (each cohort comprised 8 controls and 8 with physician diagnosis of asthma) participated in two studies of the short- and long-term reproducibility of the bronchoconstrictive response to an EVH test with dry air. EVH was performed on days 0, 7, 14, and 21 (short-term study), and 0, 35, and 70 (long-term study). HIB was diagnosed by a ≥10% fall in forced expiratory volume in 1 s (FEV1) after EVH. Results: On day 0 of the short-term study, FEV1 fell by 2 ± 1% (P < 0.05) and 27 ± 18% (P < 0.01) from pre-to post-EVH in control and HIB-positive groups respectively. The post-EVH fall in FEV1 did not differ across the short-term study test days. In the HIB-positive group, the day-to-day coefficient of variation, reproducibility, and smallest meaningful change for the fall in FEV1 were 12%, 328 mL, and 164 mL, respectively. On day 0 of the long-term study, FEV1 fell by 2 ± 2% and 25 ± 18% (P < 0.01) after EVH in control and HIB-positive groups respectively. The post-EVH fall in FEV1 did not differ across the long-term study test days. In the HIB-positive group, the day-to-day coefficient of variation, reproducibility, and smallest meaningful change for the fall in FEV1 were 10%, 196 mL, and 98 mL respectively. Conclusion: The EVH test elicits a reproducible bronchoconstrictive response in physically active males with physician diagnosed asthma. These data thus support the clinical utility of the EVH test for EIB screening and monitoring

    Monoclonal origin of B lymphocyte colony-forming cells in spleen colonies formed by multipotential hemopoietic stem cells

    Spleen colonies produced by transplanting lethally irradiated mice with either 12 day fetal liver or adult bone marrow cells were found to contain B- lymphocyte colony-forming cells (BL-CFC) . The proportion of BL-CFC positive spleen colonies did not increase substantially between 8 and 14 days after transplantation, the range being 18-45 percent. However, the absolute number of BL-CFC per spleen colony varied considerably (between 1 and 10,318), although the majority of colonies contained less than 200 BL-CFC. Irrespective of the time after transplantation, smaller spleen colonies were found to have a higher frequency of BL-CFC than larger spleen colonies. To determine the possible clonal origin of BL-CFC from spleen colony- forming unit (CFU-S), CBA mice were injected with equal numbers of CBA and CBA T(6)/T(6) fetal liver or adult bone marrow cells. Analysis of 7-15-day spleen colonies demonstrated that 90 percent were either exclusively T(6) positive or T(6) negative and approximately equal numbers ofboth colony types were observed. B-lymphocyte colonies were grown and successfully karyotyped from 19 spleen colonies. When compared with the original spleen colony karyotype the B-lymphocyte colony cells karyotype was identical in all 19 cases. In 3 of the 19 colonies analyzed a mixture of T(6) positive and T(6) negative karyotypes was present and identical proportions of the karyotypes were present in the pooled B-lymphocyte colony cells and spleen colony cells. The data indicate that the B-lymphocyte colony-forming cells detected in spleen colonies are genuine members of the hemopoietic clone derived from the initiating hemopoietic stem cell (CFU-S)

    We thank Chiappa et al. (4,5) for commending our work (2,3,7), which we reciprocate in light of their thought provoking research that sparked the ensuing trans-Atlantic debate on the effects of inspiratory muscle loading on lactate clearance after exercise. Specifically, despite using similar methodologies, Chiappa et al. (4,5) have twice shown accelerated lactate clearance with inspiratory loading, whereas we have twice shown no effect (2,7). We hypothesized that these discrepancies may be due to interstudy differences in participant endurance training status, as evidenced by higher V˙O2peak and faster blood lactate recovery kinetics in our participants. We were thus intrigued by the authors’ unpublished data showing, in sedentary individuals, no effect of inspiratory loading on lactate clearance. In their accompanying figure, the authors also present novel data showing a significant correlation between maximal inspiratory pressure (MIP) and changes in the area under the blood [Laj] curve with inspiratory loading. This observation informed their hypothesis that the efficacy of inspiratory loading is influenced by inspiratory muscle mass rather than training status

    Plasma electrons above Saturn's main rings: CAPS observations

    We present observations of thermal ( similar to 0.6 - 100eV) electrons observed near Saturn's main rings during Cassini's Saturn Orbit Insertion (SOI) on 1 July 2004. We find that the intensity of electrons is broadly anticorrelated with the ring optical depth at the magnetic footprint of the field line joining the spacecraft to the rings. We see enhancements corresponding to the Cassini division and Encke gap. We suggest that some of the electrons are generated by photoemission from ring particle surfaces on the illuminated side of the rings, the far side from the spacecraft. Structure in the energy spectrum over the Cassini division and A-ring may be related to photoelectron emission followed by acceleration, or, more likely, due to photoelectron production in the ring atmosphere or ionosphere
