    Peroxisome proliferator-activated receptor α (PPARα) mRNA expression in human hepatocellular carcinoma tissue and non-cancerous liver tissue

    <p>Abstract</p> <p>Background</p> <p>Peroxisome proliferator-activated receptor α (PPARα) regulates lipid metabolism in the liver. It is unclear, however, how this receptor changes in liver cancer tissue. On the other hand, mouse carcinogenicity studies showed that PPARα is necessary for the development of liver cancer induced by peroxisome proliferators, and the relationship between PPARα and the development of liver cancer have been the focus of considerable attention. There have been no reports, however, demonstrating that PPARα is involved in the development of human liver cancer.</p> <p>Methods</p> <p>The subjects were 10 patients who underwent hepatectomy for hepatocellular carcinoma. We assessed the expression of PPARα mRNA in human hepatocellular carcinoma tissue and non-cancerous tissue, as well as the expression of target genes of PPARα, carnitine palmitoyltransferase 1A and cyclin D1 mRNAs. We also evaluated glyceraldehyde 3-phosphate dehydrogenase, a key enzyme in the glycolytic system.</p> <p>Results</p> <p>The amounts of PPARα, carnitine palmitoyltransferase 1A and glyceraldehyde 3-phosphate dehydrogenase mRNA in cancerous sections were significantly increased compared to those in non-cancerous sections. The level of cyclin D1 mRNA tends to be higher in cancerous than non-cancerous sections. Although there was a significant correlation between the levels of PPARα mRNA and cyclin D1 mRNA in both sections, however the correlation was higher in cancerous sections.</p> <p>Conclusion</p> <p>The present investigation indicated increased expression of PPARα mRNA and mRNAs for PPARα target genes in human hepatocellular carcinoma. These results might be associated with its carcinogenesis and characteristic features of energy production.</p

    Staying Cool Across the First Year of Middle School

    As students transition into middle school they must successfully negotiate a new, larger peer context to attain or maintain high social standing. The goal of this study was to examine the extent to which the maintenance, attainment, and loss of a cool status over the course of the sixth grade is associated with student and classroom levels of physical, verbal, and relational aggression. To address this goal, we studied a sample of 1985 (55% girls) ethnically diverse adolescents from 99 sixth grade classrooms in the United States. Attaining a cool status at any point across the school year was associated with stronger aggressive reputations. Additionally, classroom norms for aggressive behavior moderated the association between changes in aggression over the school year and the stability of coolness such that students who maintained their coolness across the school year showed greater increases in their verbally aggressive reputations from fall to spring when they were in classrooms with higher levels of aggression. The findings illustrate the importance of fitting in with social norms for maintaining a high social status among a new set of peers in middle school

    Regulation of normal or abnormal behaviour is critically controlled by the central serotonergic systems. Recent evidence has suggested that serotonin (5-HT) neurotransmission dysfunction contributes to a variety of pathological conditions, including depression, anxiety, schizophrenia and Parkinson’s disorders. There is also a great amount of evidence indicating that 5-HT signalling may affect the reinforcing properties of drugs of abuse by the interaction and modulation of dopamine (DA) function. This chapter is focused on one of the more addictive drugs, nicotine. It is widely recognised that the effects of nicotine are strongly associated with the stimulatory action it exhibits on mesolimbic DAergic function. We outline the role of 5-HT and its plethora of receptors, focusing on 5-HT2 subtypes with relation to their involvement in the neurobiology of nicotine addiction. We also explore the novel pharmacological approaches using 5-HT agents for the treatment of nicotine dependence. Compelling evidence shows that 5-HT2C receptor agonists may be possible therapeutic targets for smoking cessation, although further investigation is required.peer-reviewe

    Targeting cancer metabolism: a therapeutic window opens

    Genetic events in cancer activate signalling pathways that alter cell metabolism. Clinical evidence has linked cell metabolism with cancer outcomes. Together, these observations have raised interest in targeting metabolic enzymes for cancer therapy, but they have also raised concerns that these therapies would have unacceptable effects on normal cells. However, some of the first cancer therapies that were developed target the specific metabolic needs of cancer cells and remain effective agents in the clinic today. Research into how changes in cell metabolism promote tumour growth has accelerated in recent years.     South Africa’s coastal-breeding white-breasted cormorants: population trends, breeding season and movements, and diet

    White-breasted cormorants Phalacrocorax [carbo] lucidus  breed around South Africa’s coast and at inland localities. Along the coasts of the Northern, Western and Eastern Cape provinces, numbers breeding were similar during  the periods 1977–1981 (1 116 pairs at 41 localities) and 2008–2012 (1 280 pairs at 41 localities). Along the coast of KwaZulu-Natal (not counted in 1977–1981), 197 pairs bred at nine localities in 2008–2012, when the overall number breeding around South Africa’s coastline was about 1 477 pairs. Between the two study periods, numbers decreased in the Northern and Western Cape provinces following the loss of several breeding localities, but they increased in the Eastern Cape. In the Western Cape, however, numbers were stable east of Cape Agulhas and at nine well-monitored West Coast localities that were surveyed from 1978 to 2012. White-breasted cormorants breed throughout the year, with breeding at some localities more seasonal than at others and the timing of peaks in breeding varying at and between localities. In the vicinity of Saldanha Bay/Langebaan Lagoon (Western Cape), in Algoa Bay (Eastern Cape) and in northern KwaZulu-Natal, it is likely that birds moved between breeding localities in different years, although breeding often occurred at the same locality over several years. Human disturbance, presence of predators, competition for breeding space and occurrence of breeding by other waterbirds may influence movements between colonies. Securing sufficient good habitat at which white-breasted cormorants may breed will be important for conservation of the species. The species may breed at an age of 4 years, possibly younger. The bulk of their diet around South Africa’s coast consists of inshore marine and estuarine fish species that are not intensively exploited by humans.African Journal of Marine Science 2013, 35(4): 473–49
