    A Bayesian space-time model for discrete spread processes on a lattice

We formulate a model where the spread of a disease over a heterogeneous landscape through time is represented as a probabilistic function of two processes: local diffusion and random-jump dispersal. This formulation represents two mechanisms of spread which result in highly peaked and long-tailed distributions of dispersal distances (i.e., local and long-distance spread), commonly observed in the spread of infectious diseases and biological invasions. We demonstrate the properties of this model using a simulation experiment and an empirical case study - the spread of mountain pine beetle in western Canada. Posterior predictive checking was used to validate the number of newly inhabited regions in each time period. The model performed well in the simulation study in which a goodness-of-fit statistic measuring the number of newly inhabited regions in each time interval fell within the 95% posterior predictive credible interval in over 97% of simulations. The case study of a mountain pine beetle infestation in western Canada (1999-2009) extended the base model in two ways. First, spatial covariates thought to impact the local diffusion parameters, elevation and forest cover, were included in the model. Second, a refined definition for translocation or jump-dispersal based on mountain pine beetle ecology was incorporated improving the fit of the model. Posterior predictive checks on the mountain pine beetle model found that the observed goodness-of-fit test statistic fell within the 95% posterior predictive credible interval for 8 out of 10. years. The simulation study and case study provide evidence that the model presented here is both robust and flexible; and is therefore appropriate for a wide range of spread processes in epidemiology and ecology.PostprintPeer reviewe

    L^{2}-restriction bounds for eigenfunctions along curves in the quantum completely integrable case

    We show that for a quantum completely integrable system in two dimensions,the L2L^{2}-normalized joint eigenfunctions of the commuting semiclassical pseudodifferential operators satisfy restriction bounds ofthe form γϕj2ds=O(log) \int_{\gamma} |\phi_{j}^{\hbar}|^2 ds = {\mathcal O}(|\log \hbar|) for generic curves γ\gamma on the surface.     Progressing the care, husbandry and management of ageing mice used in scientific studies

    Driven by the longer lifespans of humans, particularly in Westernised societies, and the need to know more about ‘healthy ageing’, ageing mice are being used increasingly in scientific research. Many departments and institutes involved with ageing research have developed their own systems to determine intervention points for potential refinements and to identify humane end points. Several good systems are in use, but variations between them could contribute to poor reproducibility of the science achieved. Working with scientific and regulatory communities in the UK, we have reviewed the clinical signs observed in ageing mice and developed recommendations for enhanced monitoring, behaviour assessment, husbandry and veterinary interventions. We advocate that the default time point for enhanced monitoring should be 15 months of age, unless prior information is available. Importantly, the enhanced monitoring should cause no additional harms to the animals. Where a mouse strain is well characterised, the onset of age-related enhanced monitoring may be modified based on knowledge of the onset of an expected age-related clinical sign. In progeroid models where ageing is accelerated, enhanced monitoring may need to be brought forward. Information on the background strain must be considered, as it influences the onset of age-related clinical signs. The range of ageing models currently used means that there will be no ‘one-size fits all’ solution. Increased awareness of the issues will lead to more refined and consistent husbandry of ageing mice, and application of humane end points will help to reduce the numbers of animals maintained for longer than is scientifically justified

To isolate this effect from other processes that occur in the transistor structure when under illumination, we focus attention on the photo-response of metal-insulator-semiconductor (MIS) capacitors - the core structure of transistors. The capacitors comprised poly(3-hexylthiophene), (P3HT), as the active semiconductor in combination with one of three insulators, namely, poly(amide-imide), (PAI), SU-8 photoresist and polysilsesquioxane (PSQ). Following initial characterization in the dark, the capacitor response was measured both during and after irradiation with light in the wavelength range 400–700 nm. Three different approaches were employed to study the photo-response, each providing a different insight into the processes occurring. Capacitance-voltage sweeps before, during and after illumination provided direct evidence supporting the view that the photovoltaic effect occurred as a result of electron trapping in interface states of density up to ∼2 × 1012 cm−2 in the P3HT/PAI combination but lower for SU-8 and PSQ. The dynamic photo-response, in which device capacitance was held constant by changing the applied bias, showed a fast component related to optically induced photoconduction in the semiconductor and a slower component reflecting the dynamics of interface electron trapping.     Selective coupling of type 6 adenylyl cyclase with type 2 IP3 receptors mediates direct sensitization of IP3 receptors by cAMP

    Interactions between cyclic adenosine monophosphate (cAMP) and Ca2+ are widespread, and for both intracellular messengers, their spatial organization is important. Parathyroid hormone (PTH) stimulates formation of cAMP and sensitizes inositol 1,4,5-trisphosphate receptors (IP3R) to IP3. We show that PTH communicates with IP3R via “cAMP junctions” that allow local delivery of a supramaximal concentration of cAMP to IP3R, directly increasing their sensitivity to IP3. These junctions are robust binary switches that are digitally recruited by increasing concentrations of PTH. Human embryonic kidney cells express several isoforms of adenylyl cyclase (AC) and IP3R, but IP3R2 and AC6 are specifically associated, and inhibition of AC6 or IP3R2 expression by small interfering RNA selectively attenuates potentiation of Ca2+ signals by PTH. We define two modes of cAMP signaling: binary, where cAMP passes directly from AC6 to IP3R2; and analogue, where local gradients of cAMP concentration regulate cAMP effectors more remote from AC. Binary signaling requires localized delivery of cAMP, whereas analogue signaling is more dependent on localized cAMP degradation

    The cytotoxic domain of colicin E9 is a channel-forming endonuclease

    Bacterial toxins commonly translocate cytotoxic enzymes into cells using dedicated channelforming subunits or domains as conduits. We demonstrate that the small cytotoxic endonuclease domain from the bacterial toxin colicin E9 (the E9 DNase) exhibits nonvoltage- gated, channel-forming activity in planar lipid bilayers and that this activity is linked to toxin translocation into cells. A disulfide bond engineered into the DNase abolished channel activity and colicin toxicity but left endonuclease activity unaffected, with NMR experiments suggesting decreased conformational flexibility as the likely reason for these alterations. Concomitant with the reduction of the disulfide bond was the restoration of conformational flexibility, DNase channel activity and colicin toxicity. Our data suggest that endonuclease domains of colicins may mediate their own translocation across the bacterial inner membrane through an intrinsic channel activity that is dependent on structural plasticity in the protein

    Constraining warm dark matter with cosmic shear power spectra

    We investigate potential constraints from cosmic shear on the dark matter particle mass, assuming all dark matter is made up of light thermal relic particles. Given the theoretical uncertainties involved in making cosmological predictions in such warm dark matter scenarios we use analytical fits to linear warm dark matter power spectra and compare (i) the halo model using a mass function evaluated from these linear power spectra and (ii) an analytical fit to the non-linear evolution of the linear power spectra. We optimistically ignore the competing effect of baryons for this work. We find approach (ii) to be conservative compared to approach (i). We evaluate cosmological constraints using these methods, marginalising over four other cosmological parameters. Using the more conservative method we find that a Euclid-like weak lensing survey together with constraints from the Planck cosmic microwave background mission primary anisotropies could achieve a lower limit on the particle mass of 2.5 keV.Comment: 26 pages, 9 figures, minor changes to match the version accepted for publication in JCA

    Fermionic partner of Quintessence field as candidate for dark matter

    Quintessence is a possible candidate for dark energy. In this paper we study the phenomenologies of the fermionic partner of Quintessence, the Quintessino. Our results show that, for suitable choices of the model parameters, the Quintessino is a good candidate for cold or warm dark matter.
