    Cigarette ignition propensity, smoking behavior, and toxicant exposure: A natural experiment in Canada

    <p>Abstract</p> <p>Background</p> <p>This study used a 'pre-post' research design to measure the impact of the Canadian reduced ignition propensity law on cigarette toxicity and smoking behavior among Canadian smokers.</p> <p>Method</p> <p>The study was conducted in Ontario, Canada over a ten-month period in 2005-2006, consisting of 4 laboratory visits (baseline N = 61, final N = 42). At Visit 1, questionnaire data and biospecimens were collected. During the following 24 hours, participants smoked 5 cigarettes <it>ad libitum </it>through a topography recording device and collected their cigarette butts. Visit 2 consisted of a questionnaire and smoking one cigarette to measure laboratory topography values. After ten months, these procedures were repeated.</p> <p>Results</p> <p>Generalized estimating equations, with law status (pre and post) as a fixed within-subject factor, were used to determine changes in behavior and biomarker exposure. Overall, there were no significant differences in smoking topography, breath carbon monoxide, and saliva cotinine pre-post law (<it>p</it>>0.1). However, analyses revealed a significant increase in the summed concentrations of hydroxyfluorene metabolites (N = 3),, and 1-hydroxypyrene in urine, with at notable increase in hydroxyphenanthrene metabolites (N = 3) (<it>p</it><sub>Σhydroxyfluorene </sub>= 0.013, 22% increase; <it>p<sub>1-hydroxypyrene </sub></it>= 0.018, 24% increase; <it>p</it><sub>Σhydroxyphenanthrene </sub>= 0.061, 17% increase).</p> <p>Conclusion</p> <p>While the results suggest no change in topography variables, data showed increases in exposure to three PAH biomarkers following reduced ignition propensity implementation in Canada. These findings suggest that human studies should be considered to evaluate policy impacts.</p

    Absorption spectrum of a weakly n-doped semiconductor quantum well

    We calculate, as a function of temperature and conduction band electron density, the optical absorption of a weakly n-doped, idealized semiconductor quantum well. In particular, we focus on the absorption band due to the formation of a charged exciton. We conceptualize the charged exciton as an itinerant excitation intimately linked to the dynamical response of itinerant conduction band electrons to the appearance of the photo-generated valence band hole. Numerical results for the absorption in the vicinity of the exciton line are presented and the spectral weights associated with, respectively, the charged exciton band and the exciton line are analyzed in detail. We find, in qualitative agreement with experimental data, that the spectral weight of the charged exciton grows with increasing conduction band electron density and/or decreasing temperature at the expense of the exciton.Comment: 5 pages, 4 figure

    Intraprocedural MRI-based dosimetry during transarterial radioembolization of liver tumours with holmium-166 microspheres (EMERITUS-1):a phase I trial towards adaptive, image-controlled treatment delivery

    PURPOSE: Transarterial radioembolization (TARE) is a treatment for liver tumours based on injection of radioactive microspheres in the hepatic arterial system. It is crucial to achieve a maximum tumour dose for an optimal treatment response, while minimizing healthy liver dose to prevent toxicity. There is, however, no intraprocedural feedback on the dose distribution, as nuclear imaging can only be performed after treatment. As holmium-166 ((166)Ho) microspheres can be quantified with MRI, we investigate the feasibility and safety of performing (166)Ho TARE within an MRI scanner and explore the potential of intraprocedural MRI-based dosimetry. METHODS: Six patients were treated with (166)Ho TARE in a hybrid operating room. Per injection position, a microcatheter was placed under angiography guidance, after which patients were transported to an adjacent 3-T MRI system. After MRI confirmation of unchanged catheter location, (166)Ho microspheres were injected in four fractions, consisting of 10%, 30%, 30% and 30% of the planned activity, alternated with holmium-sensitive MRI acquisition to assess the microsphere distribution. After the procedures, MRI-based dose maps were calculated from each intraprocedural image series using a dedicated dosimetry software package for (166)Ho TARE. RESULTS: Administration of (166)Ho microspheres within the MRI scanner was feasible in 9/11 (82%) injection positions. Intraprocedural holmium-sensitive MRI allowed for tumour dosimetry in 18/19 (95%) of treated tumours. Two CTCAE grade 3–4 toxicities were observed, and no adverse events were attributed to treatment in the MRI. Towards the last fraction, 4/18 tumours exhibited signs of saturation, while in 14/18 tumours, the microsphere uptake patterns did not deviate from the linear trend. CONCLUSION: This study demonstrated feasibility and preliminary safety of a first in-human application of TARE within a clinical MRI system. Intraprocedural MRI-based dosimetry enabled dynamic insight in the microsphere distribution during TARE. This proof of concept yields unique possibilities to better understand microsphere distribution in vivo and to potentially optimize treatment efficacy through treatment personalization.     Micro- and macro-habitat selection of Atlantic salmon, (Salmo salar), post-smolts in relation to marine environmental cues

    Atlantic salmon is an economically and culturally important species. The species encounters several natural and man-made threats during its migration between fresh water and the ocean, which in combination may explain its ongoing decline. With the aim to better understand whether post-smolt behaviour is influenced by physical oceanographic conditions, the migratory behaviour of 173 post-smolts in a high-latitude Norwegian fjord was investigated, combining acoustic telemetry with site- and time-specific environmental variables from an oceanographic model. Most post-smolts (94%) performed a unidirectional migration out the fjord. Progression rates were relatively high (0.42–2.41 km h−1; 0.84–3.78 BL s−1) and increased with distance from the river. While post-smolts had an affinity for lower salinities in the inner fjord, statistical models failed to detect any significant relationship between the small-scale (within arrays) migratory behaviour and salinity, temperature, or coastal surface currents within the fjord. In the outer part, the post-smolts predominantly exited the fjord system through the strait with the highest surface salinities and lowest temperatures, independently of the current direction. Our findings indicate that the macro-habitat selection of the Atlantic salmon post-smolts was influenced by environmental factors: the post-smolts directed their migration towards “ocean cues.” However, this was not confirmed on the micro-habitat level.publishedVersio

    The skeletal phenotype of chondroadherin deficient mice

    Chondroadherin, a leucine rich repeat extracellular matrix protein with functions in cell to matrix interactions, binds cells via their a2b1 integrin as well as via cell surface proteoglycans, providing for different sets of signals to the cell. Additionally, the protein acts as an anchor to the matrix by binding tightly to collagens type I and II as well as type VI. We generated mice with inactivated chondroadherin gene to provide integrated studies of the role of the protein. The null mice presented distinct phenotypes with affected cartilage as well as bone. At 3–6 weeks of age the epiphyseal growth plate was widened most pronounced in the proliferative zone. The proteome of the femoral head articular cartilage at 4 months of age showed some distinct differences, with increased deposition of cartilage intermediate layer protein 1 and fibronectin in the chondroadherin deficient mice, more pronounced in the female. Other proteins show decreased levels in the deficient mice, particularly pronounced for matrilin-1, thrombospondin-1 and notably the members of the a1-antitrypsin family of proteinase inhibitors as well as for a member of the bone morphogenetic protein growth factor family. Thus, cartilage homeostasis is distinctly altered. The bone phenotype was expressed in several ways. The number of bone sialoprotein mRNA expressing cells in the proximal tibial metaphysic was decreased and the osteoid surface was increased possibly indicating a change in mineral metabolism. Micro-CT revealed lower cortical thickness and increased structure model index, i.e. the amount of plates and rods composing the bone trabeculas. The structural changes were paralleled by loss of function, where the null mice showed lower femoral neck failure load and tibial strength during mechanical testing at 4 months of age. The skeletal phenotype points at a role for chondroadherin in both bone and cartilage homeostasis, however, without leading to altered longitudinal growth

    The role of human leukocyte antigen (HLA) class I and killer immunoglobulin-like receptor molecules in mediating acute retroviral syndrome (ARS) during human immunodeficiency virus type 1 (HIV-1) infection is unclear. Among 72 sub-Saharan African adults, HLA-A*23 was associated with lower odds of ARS (adjusted odds ratio, 0.10 [95% confidence interval, .01–.48]; P = .009), which warrants further studies to explore its role on HIV-1–specific immunopathogenesis

    Campi Flegrei active seismic experiments waveforms compilation

    A new experiment called SERAPIS (SEismic Reflection/Refraction Acquisition Project for Imaging complex volcanic Structures) has been planned and carried out, based on off-shore seismic energization and data acquisition on land and on sea-bottom. The experiment was performed in September, 2001 during which the vessel NADIR of IFREMER (equipped with 12, 16-liters airgun) produced more than 5000 air gun shots recorded at a sea-bottom seismograph array of 72 OBS and 62 stations installed on-land. Active seismic refraction DSS (Deep Seismic Soundings) acquired during the surveys conducted in 1980 and 1985 were recovered jointly with seismic data acquired in the Campi Flegrei area in the framework of the MareVes97 (an experiment devoted to the definition of the structure of the Somma-Vesuvio complex) offshore survey. The data set acquired during the SERAPIS experiment has been successfully used to infer 3D images of the volcanic structures of Campi Flegrei and Neapolitan bay. Active seismic waveforms and related P-picks (more than 90000 data) from the SERAPIS experiment are also available in the project data server

    Binding Energy of Charged Excitons in ZnSe-based Quantum Wells

    Excitons and charged excitons (trions) are investigated in ZnSe-based quantum well structures with (Zn,Be,Mg)Se and (Zn,Mg)(S,Se) barriers by means of magneto-optical spectroscopy. Binding energies of negatively () and positively (X+) charged excitons are measured as functions of quantum well width, free carrier density and in external magnetic fields up to 47 T. The binding energy of shows a strong increase from 1.4 to 8.9 meV with decreasing quantum well width from 190 to 29 A. The binding energies of X+ are about 25% smaller than the binding energy in the same structures. The magnetic field behavior of and X+ binding energies differ qualitatively. With growing magnetic field strength, increases its binding energy by 35-150%, while for X+ it decreases by 25%. Zeeman spin splittings and oscillator strengths of excitons and trions are measured and discussed

    Amino Acid Signatures to Evaluate the Beneficial Effects of Weight Loss

    Aims. We investigated the relationship between circulating amino acid levels and obesity; to what extent weight loss followed by weight maintenance can correct amino acid abnormalities; and whether amino acids are related to weight loss. Methods:. Amino acids associated with waist circumference (WC) and BMI were studied in 804 participants from the Malmö Diet and Cancer Cardiovascular Cohort (MDC-CC). Changes in amino acid levels were analyzed after weight loss and weight maintenance in 12 obese subjects and evaluated in a replication cohort (n = 83). Results:. Out of the eight identified BMI-associated amino acids from the MDC-CC, alanine, isoleucine, tyrosine, phenylalanine, and glutamate decreased after weight loss, while asparagine increased after weight maintenance. These changes were validated in the replication cohort. Scores that were constructed based on obesity-associated amino acids and known risk factors decreased in the ≥10% weight loss group with an associated change in BMI (R2 = 0.16–0.22, p < 0.002), whereas the scores increased in the <10% weight loss group (p < 0.0004). Conclusions:. Weight loss followed by weight maintenance leads to differential changes in amino acid levels associated with obesity. Treatment modifiable scores based on epidemiological and interventional data may be used to evaluate the potential metabolic benefit of weight loss

    Genetic Variance in the Adiponutrin Gene Family and Childhood Obesity

    AIM: The adiponutrin gene family consists of five genes (PNPLA1-5) coding for proteins with both lipolytic and lipogenic properties. PNPLA3 has previously been associated with adult obesity. Here we investigated the possible association between genetic variants in these genes and childhood and adolescent obesity. METHODS/RESULTS: Polymorphisms in the five genes of the adiponutrin gene family were selected and genotyped using the Sequenom platform in a childhood and adolescent obesity case-control study. Six variants in PNPLA1 showed association with obesity (rs9380559, rs12212459, rs1467912, rs4713951, rs10947600, and rs12199580, p0.05). When analyzing these SNPs in relation to phenotypes, two SNPs in the PNPLA3 gene showed association with insulin sensitivity (rs12483959: beta = -0.053, p = 0.016, and rs2072907: beta = -0.049, p = 0.024). No associations were seen for PNPLA2, PNPLA4, and PNPLA5. CONCLUSIONS: Genetic variation in the adiponutrin gene family does not seem to contribute strongly to obesity in children and adolescents. PNPLA1 exhibited a modest effect on obesity and PNPLA3 on insulin sensitivity. These data, however, require confirmation in other cohorts and ethnic groups
