    Comparison of the Effect of Cold Plasma with Conventional Preservation Methods on Red Wine Quality Using Chemometrics Analysis

    In this study, the effect of cold plasma (CP) on the physicochemical and biological properties of red wine was investigated in comparison with the effects of the conventional preservation method and the combined method. In addition, the effect of storage time after the application of each of the analyzed methods was evaluated. The study examined the effects of the different preservation methods on the pH, color, phenolic content, antioxidant activity and microbiological purity of the red wine. Chemometric analysis was used to discover the relationship between the preservation method used and wine quality. In the wine samples tested, a reduction in phenolic compounds and a decrease in antioxidant activity were noted after storage. This effect was mildest for preservation methods with the addition of potassium metabisulphite and those in which a mixture of helium and nitrogen was used as the working gas. On a positive note, the CP treatment did not affect the color of the wine in a way perceptible to the consumer: ∆E*—1.12 (He/N2; 5 min). In addition, the lowest growth of microorganisms was detected in the CP-treated samples. This indicates the potential of cold plasma as an alternative method to the use of potassium metabisulfite in wine production, which may contribute to its wider use in the alcohol industry in the future

    Variability of high rainfalls and related synoptic situations causing heavy floods at the northern foothills of the Tatra Mountains

    This contribution provides the basics of the climatology of the Polish Tatra Mountains in a nutshell, with particular reference to intense precipitation and its relation to atmospheric circulation. Variability of various precipitation characteristics, including selected indices of intense precipitation in Zakopane and at Kasprowy Wierch, is illustrated in this paper. None of the trends in these characteristics and indices calculated for the entire time interval exhibit a statistical significance, but short-time fluctuations are evident. The occurrence of intense precipitation in the Tatra Mountains is strongly related to three circulation types. These situations (Nc, NEc, Bc) are associated with cyclones following track Vb after van Bebber. In addition to changing frequencies of circulation, this study also reveals an increase in the frequency of the circulation types associated with extreme precipitation

    Biological Activity of an Epilobium angustifolium L. (Fireweed) Infusion after In Vitro Digestion

    The biological activity of an in vitro digested infusion of Epilobium angustifolium (fireweed) was examined in a model system of intestinal epithelial and colon cancer tissues. The content of selected phenolic compounds in the digested aqueous extract of fireweed was determined using HPLC-ESI-QTOF-MS/MS. Biological activity was examined using the human colon adenocarcinoma cell lines HT-29 and CaCo-2 and the human colon epithelial cell line CCD 841 CoTr. Cytotoxicity was assessed by an MTT assay, a Neutral Red uptake assay, May-Grünwald-Giemsa staining, and a label-free Electric Cell-Substrate Impedance Sensing cytotoxicity assay. The effect of the infusion on the growth of selected intestinal bacteria was also examined. The extract inhibited the growth of intestinal cancer cells HT-29. This effect can be attributed to the activity of quercetin and kaempferol, which were the most abundant phenolic compounds found in the extract after in vitro digestion. The cytotoxicity of the fireweed infusion was dose-dependent. The highest decrease in proliferation (by almost 80%) compared to the control was observed in HT-29 line treated with the extract at a concentration of 250 μg/mL. The fireweed infusion did not affect the growth of beneficial intestinal bacteria, but it did significantly inhibit E. coli. The cytotoxic effect of the fireweed extract indicates that it does not lose its biological activity after in vitro digestion. It can be concluded that the fireweed infusion has the potential to be used as a supporting agent in colon cancer therapy

    CXCL8, CCL2, and CMV Seropositivity as New Prognostic Factors for a Severe COVID-19 Course

    The exact pathophysiology of severe COVID-19 is not entirely elucidated, but it has been established that hyperinflammatory responses and cytokine storms play important roles. The aim of this study was to examine CMV status, select chemokines, and complement components in COVID-19, and how concentrations of given molecules differ over time at both molecular and proteomic levels. A total of 210 COVID-19 patients (50 ICU and 160 non-ICU patients) and 80 healthy controls were enrolled in this study. Concentrations of select chemokines (CXCL8, CXCL10, CCL2, CCL3, CCR1) and complement factors (C2, C9, CFD, C4BPA, C5AR1, CR1) were examined at mRNA and protein levels with regard to a COVID-19 course (ICU vs. non-ICU group) and CMV status at different time intervals. We detected several significant differences in chemokines and complement profiles between ICU and non-ICU groups. Pro-inflammatory chemokines and the complement system appeared to greatly contribute to the pathogenesis and development of severe COVID-19. Higher concentrations of CXCL8 and CCL2 in the plasma, with reduced mRNA expression presumably through negative feedback mechanisms, as well as CMV-positive status, correlated with more severe courses of COVID-19. Therefore, CXCL8, CCL2, and CMV seropositivity should be considered as new prognostic factors for severe COVID-19 courses. However, more in-depth research is needed

    Floods at the northern foothills of the Tatra Mountains — A Polish-Swiss research project

    The present paper introduces the topical area of the Polish-Swiss research project FLORIST (Flood risk on the northern foothills of the Tatra Mountains), informs on its objectives, and reports on initial results. The Tatra Mountains are the area of the highest precipitation in Poland and largely contribute to flood generation. The project is focused around four competence clusters: observation-based climatology, model-based climate change projections and impact assessment, dendrogeomorphology, and impact of large wood debris on fluvial processes. The knowledge generated in the FLORIST project is likely to have impact on understanding and interpretation of flood risk on the northern foothills of the Tatra Mountains, in the past, present, and future. It can help solving important practical problems related to flood risk reduction strategies and flood preparedness
