The ADAXIALIZED LEAF1 gene functions in leaf and embryonic pattern formation in rice
AbstractThe adaxial–abaxial axis in leaf primordia is thought to be established first and is necessary for the expansion of the leaf lamina along the mediolateral axis. To understand axis information in leaf development, we isolated the adaxialized leaf1 (adl1) mutant in rice, which forms abaxially rolled leaves. adl1 leaves are covered with bulliform-like cells, which are normally distributed only on the adaxial surface. An adl1 double mutant with the adaxially snowy leaf mutant, which has albino cells that specifically appear in the abaxial mesophyll tissue, indicated that adl1 leaves show adaxialization in both epidermal and mesophyll tissues. The expression of HD-ZIPIII genes in adl1 mutant increased in mature leaves, but not in the young primordia or the SAM. This indicated that ADL1 may not be directly involved in determining initial leaf polarity, but rather is associated with the maintenance of axis information. ADL1 encodes a plant-specific calpain-like cysteine proteinase orthologous to maize DEFECTIVE KERNEL1. Furthermore, we identified intermediate and strong alleles of the adl1 mutant that generate shootless embryos and globular-arrested embryos with aleurone layer loss, respectively. We propose that ADL1 plays an important role in pattern formation of the leaf and embryo by promoting proper epidermal development

Signal transmission from the mechanical forces to the various intracellular activities is a fundamental process during tissue development. Despite their critical role, the mechanism of mechanical forces in the biological process is poorly understood. In this study, we demonstrated that in the response to hydrostatic pressure (HP), the piezo type mechanosensitive ion channel component 1 (PIEZO1) is a primary mechanosensing receptor for odontoblast differentiation through coordination of the WNT expression and ciliogenesis. In stem cells from human exfoliated deciduous teeth (SHED), HP significantly promoted calcium deposition as well as the expression of odontogenic marker genes, PANX3 and DSPP, and WNT related-genes including WNT5b and WNT16, whereas HP inhibited cell proliferation and enhanced primary cilia expression. WNT signaling inhibitor XAV939 and primary cilia inhibitor chloral hydrate blocked the HP-induced calcium deposition. The PIEZO1 activator Yoda1 inhibited cell proliferation but induced ciliogenesis and WNT16 expression. Interestingly, HP and Yoda1 promoted nuclear translocation of RUNX2, whereas siRNA-mediated silencing of PIEZO1 decreased HP-induced nuclear translocation of RUNX2. Taken together, these results suggest that PIEZO1 functions as a mechanotransducer that connects HP signal to the intracellular signalings during odontoblast differentiation
Effect of drought stress on flowering characteristics in rice (Oryza sativa L.): A study using genotypes contrasting in drought tolerance and flower opening time
Rice is most susceptible to heat and drought stress at flowering stage, but flowering characteristics under drought stress are not well characterized. This study investigated flowering characteristics of rice genotypes contrasting in their flower opening time (FOT) and level of drought tolerance. Near-isogenic lines for the early-morning flowering trait (IR64+ qEMF3) and for drought tolerance (IR87707-445-B-B-B), and their recurrent parent cultivar (IR64) were used. IR64+ qEMF3 had stable earlier FOT than IR64 and IR87707-445-B-B-B under drought stress conditions. Drought stress occasionally affects FOT depending on genotype. The number of open spikelets was higher in IR87707-445-B-B-B than in IR64 and IR64+ qEMF3, and the difference among genotypes increased as the rice plants were subjected to more severe stress levels. Panicle temperature increased under drought stress conditions and was similar among genotypes when it was measured at the same time of day, demonstrating that earlier FOT in IR64+ qEMF3 must be beneficial to avoid heat stress at flowering under drought stress conditions. However, IR64+ qEMF3 did not exhibit drought avoidance, as evidenced by the root mass at depth. To assess the potential for the EMF trait to complement ongoing drought breeding efforts, heat tolerance among 13 advanced drought breeding lines and released cultivars was tested. Wide variation in heat tolerance at flowering was observed and, notably, none of the 13 lines possessed the EMF trait. This study therefore proposes that a breeding strategy that transfers the EMF trait into drought tolerant lines could enhance the resilience of rice spikelets to the combined stresses of heat and drought at flowering
Iroquois homeobox 3 regulates odontoblast proliferation and differentiation mediated by Wnt5a expression
Iroquois homeobox (Irx) genes are TALE-class homeobox genes that are evolutionarily conserved across species and have multiple critical cellular functions in fundamental tissue development processes. Previous studies have shown that Irxs genes are expressed during tooth development. However, the precise roles of genes in teeth remain unclear. Here, we demonstrated for the first time that Irx3 is an essential molecule for the proliferation and differentiation of odontoblasts. Using cDNA synthesized from postnatal day 1 (P1) tooth germs, we examined the expression of all Irx genes (Irx1-Irx6) by RT-PCR and found that all genes except Irx4 were expressed in the tooth tissue. Irx1-Irx3 a were expressed in the dental epithelial cell line M3H1 cells, while Irx3 and Irx5 were expressed in the dental mesenchymal cell line mDP cells. Only Irx3 was expressed in both undifferentiated cell lines. Immunostaining also revealed the presence of IRX3 in the dental epithelial cells and mesenchymal condensation. Inhibition of endogenous Irx3 by siRNA blocks the proliferation and differentiation of mDP cells. Wnt3a, Wnt5a, and Bmp4 are factors involved in odontoblast differentiation and were highly expressed in mDP cells by quantitative PCR analysis. Interestingly, the expression of Wnt5a (but not Wnt3a or Bmp4) was suppressed by Irx3 siRNA. These results suggest that Irx3 plays an essential role in part through the regulation of Wnt5a expression during odontoblast proliferation and differentiation
