    Autopolyploid inheritance and a heterozygous reciprocal translocation shape chromosome genetic behavior in tetraploid blueberry (Vaccinium corymbosum)

    Understanding chromosome recombination behavior in polyploidy species is key to advancing genetic discoveries. In blueberry, a tetraploid species, the line of evidences about its genetic behavior still remain poorly understood, owing to the inter-specific, and inter-ploidy admixture of its genome and lack of in depth genome-wide inheritance and comparative structural studies. Here we describe a new high-quality, phased, chromosome-scale genome of a diploid blueberry, clone W85. The genome was integrated with cytogenetics and high-density, genetic maps representing six tetraploid blueberry cultivars, harboring different levels of wild genome admixture, to uncover recombination behavior and structural genome divergence across tetraploid and wild diploid species. Analysis of chromosome inheritance and pairing demonstrated that tetraploid blueberry behaves as an autotetraploid with tetrasomic inheritance. Comparative analysis demonstrated the presence of a reciprocal, heterozygous, translocation spanning one homolog of chr-6 and one of chr-10 in the cultivar Draper. The translocation affects pairing and recombination of chromosomes 6 and 10. Besides the translocation detected in Draper, no other structural genomic divergences were detected across tetraploid cultivars and highly inter-crossable wild diploid species.     Chinese Workers’ Literature in the 20th and 21st Centuries

    “Chinese workers’ literature” is an umbrella term that comprises diverse writings by workers, for workers, and about workers. In the 1930s, roughly at the same time that an international proletarian arts movement was flourishing, a factory-based reportage literature—mostly written by leftist intellectuals and partly inspired by Russian and Japanese experiments— developed in semicolonial Shanghai. After the founding of the People’s Republic of China (PRC) in 1949, writings by workers themselves were officially promoted and published in state-supported venues. Largely consisting of edifying stories, poems, and plays, workers’ literature from the 1950s to the 1970s provided models of behavior and contributed to a shared sense of dignity among industrial workers; it was, however, severely limited in its expressive range. Along with the implementation of market reforms beginning in the 1980s and the privatization and contracting out of most state-owned industry, a new literature emerged in the Special Economic Zones of South China and has grown into a heterogeneous phenomenon encompassing poetry and prose written by countless rural-to-urban migrant workers, the mainstay of the country’s new workforce. These writings have been appreciated for their intimate portrayals of the human costs of economic development, for giving voice to the silent majority of precarious laborers who have made it possible, and for potentially restituting a measure of dignity to a social group whose members were once considered “masters of the country” but who, in the early 21st century, enjoy little job security and few rights. While it is possible to hear resonances across these disparate times and locations, much has changed along the way, including the social position of the worker and the groups associated with this term, the forms they have experimented with and the media through which their writings circulate, and the extent to which the workers have actively contributed to its production and circulation

    Myogenesis in the sea urchin embryo: The molecular fingerprint of the myoblast precursors

    Background: In sea urchin larvae the circumesophageal fibers form a prominent muscle system of mesodermal origin. Although the morphology and later development of this muscle system has been well-described, little is known about the molecular signature of these cells or their precise origin in the early embryo. As an invertebrate deuterostome that is more closely related to the vertebrates than other commonly used model systems in myogenesis, the sea urchin fills an important phylogenetic gap and provides a unique perspective on the evolution of muscle cell development.Results: Here, we present a comprehensive description of the development of the sea urchin larval circumesophageal muscle lineage beginning with its mesodermal origin using high-resolution localization of the expression of several myogenic transcriptional regulators and differentiation genes. A few myoblasts are bilaterally distributed at the oral vegetal side of the tip of the archenteron and first appear at the late gastrula stage. The expression of the differentiation genes Myosin Heavy Chain, Tropomyosin I and II, as well as the regulatory genes MyoD2, FoxF, FoxC, FoxL1, Myocardin, Twist, and Tbx6 uniquely identify these cells. Interestingly, evolutionarily conserved myogenic factors such as Mef2, MyoR and Six1/2 are not expressed in sea urchin myoblasts but are found in other mesodermal domains of the tip of the archenteron. The regulatory states of these domains were characterized in detail. Moreover, using a combinatorial analysis of gene expression we followed the development of the FoxF/FoxC positive cells from the onset of expression to the end of gastrulation. Our data allowed us to build a complete map of the Non-Skeletogenic Mesoderm at the very early gastrula stage, in which specific molecular signatures identify the precursors of different cell types. Among them, a small group of cells within the FoxY domain, which also express FoxC and SoxE, have been identified as plausible myoblast precursors. Together, these data support a very early gastrula stage segregation of the myogenic lineage.Conclusions: From this analysis, we are able to precisely define the regulatory and differentiation signatures of the circumesophageal muscle in the sea urchin embryo. Our findings have important implications in understanding the evolution of development of the muscle cell lineage at the molecular level. The data presented here suggest a high level of conservation of the myogenic specification mechanisms across wide phylogenetic distances, but also reveal clear cases of gene cooption. © 2013 Andrikou et al.; licensee BioMed Central Ltd

    Hyperproduction of fibrin and inefficacy of antithrombin III and alpha2 macroglobulin in the presence of bacterial porins.

    Bacterial porins enhance the thrombin activity upon chromogen substrate chromozym. Should porin-dependent enhancement of thrombin activity take place also upon fibrinogen in vivo, this might greatly increase the fibrin production which, in turn, might lead to blood vessel obstruction. In this study, we demonstrate fibrin hyperproduction in a simplified coagulative system, consisting of fibrinogen and thrombin-pure molecules, in the presence of bacterial porins. In particular, bacterial porins, in the presence of thrombin, significantly increased the fibrin production compared with thrombin alone. Also, fibrin hyperproduction took place even in the presence of the thrombin inhibitors antithrombin III (AT III) or alpha2 macroglobulin (alpha2M). However, the thrombin-fibrinogen reaction in the presence of AT III or alpha2M did not generate fibrin, unless porins were present. In conclusion, porins not only enhance thrombin activity but also inhibit the antithrombin activity exerted by AT III or alpha2M. We hypothesize that, because of porins activity, fibrin is largely generated due to thrombin hyperactivation. Moreover, further fibrin is produced by thrombin, which is not blocked by two serpins for the presence of porins. These results might be relevant as to the occurrence of disseminated intravascular coagulation in sepsis by gram-negative bacteria, which are known to produce porins

    Road toward a new model of care for idiopathic pulmonary fibrosis in the Lazio Region

    A timely, confirmed diagnosis of Idiopathic Pulmonary Fibrosis (IPF) has a significant impact on the evolution of the disease. The current model of care in the Lazio region (in Italy) was assessed on the basis of real-world data provided by the four reference centers responsible for diagnosing and treating IPF. The 5-year, population-based, retrospective longitudinal study provided the data that is at the basis of the current proposal for a new clinical and therapeutic pathway (DTCP) and has been shared with regional decision makers. A DTCP must be defined and based on four pillars: GPs, pulmonologists, IPF centers, and telemedicine. Each must play a role within a sort of hub-and-spoke model. IPF centers remain the hubs, while spokes are identified in trained GPs and pulmonologists

    Gamma spectrometer based on CeBr3 scintillator with Compton suppression for identification of trace activities in water

    The performance of a spectrometry system (SPEC) for identification of trace activities in water is presented. The SPEC detector is a part of the TAWARA-RTM platform designed for monitoring of water processed for distribution by the Waterworks Company. The SPEC system is dedicated for identification of radioisotopes emitting \u3b3-rays of energies between 50 keV and 2 MeV. The SPEC detector comprises a CeBr3 scintillator grown from specially purified raw material with reduced content of actinides that produce unwanted alpha particle background. In order to reduce the Compton scattering background, an active anti-Compton shield (ACS) made of BGO is used. In addition, a passive lead shield around the ACS is used for further suppression of the external background. The SPEC detector is designed for use with a dedicated concentrator system, that will be used for absorption of a selected group of radioactive elements that may be present in water processed at the waterworks. The system is tested upon capability to detect the \u3b3-ray emitters with activity according to the guidance levels published by the EU in the Council Directive. \ua9 2015 IEEE

    The ATLAS Fast Tracker Processing Units - track finding and fitting

    The Fast Tracker is a hardware upgrade to the ATLAS trigger and data-acquisition system, with the goal of providing global track reconstruction by the start of the High Level Trigger starts. The Fast Tracker can process incoming data from the whole inner detector at full first level trigger rate, up to 100 kHz, using custom electronic boards. At the core of the system is a Processing Unit installed in a VMEbus crate, formed by two sets of boards: the Associative Memory Board and a powerful rear transition module called the Auxiliary card, while the second set is the Second Stage board. The associative memories perform the pattern matching looking for correlations within the incoming data, compatible with track candidates at coarse resolution. The pattern matching task is performed using custom application specific integrated circuits, called associative memory chips. The auxiliary card prepares the input and reject bad track candidates obtained from from the Associative Memory Board using the full precision and a linearized fit. The track candidates from the auxiliary card use only 8 of 12 silicon layers, the track segments are extended to the additional layers by the Second Stage Board. During the first half of 2016, the first Fast Tracker VMEbus Processing Units will be installed in the ATLAS cavern. This talk will summarize the experience with newer associative memory chips and the boards; monitoring/debugging tools, including input/output data rates, track finding efficiency and track fitting results. Comparisons of the different metrics with offline simulation will also be shown
