    Differential expression of gga-miR-142-3p microRNA of haemopoeitic origin during immune organ development and functional stages in chicken embryos creates new opportunities for understanding its pivotal role during embryonic developmental stages. To decipher the role of gga-miR-142-3p in-ovo knockdown was carried out with LNA modified anti-miR-gga-miR-142-3p via intravenous route at developmental and functional stages of the immune organs and other organs. Bioinformatic analysis of the genes targeted by gga-miRNA-142-3p revealed that the predicted gene XPO1 conserved binding sites at 3’UTR. The target gene XPO1 was evaluated as the validated target of gga-miR-142-3p by employing qPCR SYBR green based technology, which was evidenced by its increased expression in the tissues of gga-miR-142-3p knockdown chicken embryos. Histopathological alterations in the immune organs and visceral organs indicated that the systemic knockdown of gga-miR-124-3p led to over expression of the XPO1 gene during the embryonic stages, and changed the environment of the immune organs related to structural integrity, immune response, signal transduction and migration of B and T cells during the embryonic developmental stage in the chicken embryos. The results clearly indicated that these changes could alter the postnatal development and functions of these immune organs, and may lead to development of immuno-compromised chickens.Različita ekspresija gga-miR-142-3p mikroRNA hemopoetskog podrijetla daje nove mogućnosti razumijevanja njezine ključne uloge u embrionalnom razvoju limfnih organa i funkcionalnih zbivanja u pilećem zametku. Za otkrivanje uloge gga-miR-142-3p in ovo provedeno je utišavanje zaključanom nukleinskom kiselinom što preko anti-miR-gga-miR-142-3p preinačuje razvojne i funkcionalne sposobnosti limfnih i drugih organa. Bioinformatička analiza ciljnih gena za gga-miRNA-142-3p otkriva da gen XPO1 ima konzervirana mjesta vezanja na 3’UTR. Gen XPO1 bio je potvrđen kao cilj za gga-miR-142-3p uporabom tehnologije temeljene na qPCR SYBR zelenilu, što je bilo dokazano njegovom povećanom ekspresijom u tkivima pilećih zametaka s utišanim gga-miR-142-3p. Patohistološke promjene u imunosnim i unutarnjim organima pokazuju da sustavno utišavanje gga-miR-124-3p vodi do prevelike ekspresije gena XPO1 tijekom embrionalnog razvoja. To mijenja zadaću imunosnih organa s obzirom na strukturni integritet, imunosni odgovor, prijenos poruka te migraciju B i T limfocita tijekom razvoja pilećih zametaka. Rezultati jasno naznačuju da te promjene mogu preinačiti postnatalni razvoj i funkcije imunosnih organa te mogu dovesti do razvoja imunološki oslabljenih pilića

    Doping dependence of the coupling of electrons to bosonic modes in the single-layer high-temperature Bi2Sr2CuO6 superconductor

    A recent highlight in the study of high-Tc superconductors is the observation of band renormalization / self-energy effects on the quasiparticles. This is seen in the form of kinks in the quasiparticle dispersions as measured by photoemission and interpreted as signatures of collective bosonic modes coupling to the electrons. Here we compare for the first time the self-energies in an optimally doped and strongly overdoped, non-superconducting single-layer Bi-cuprate (Bi2Sr2CuO6). Besides the appearance of a strong overall weakening, we also find that weight of the self-energy in the overdoped system shifts to higher energies. We present evidence that this is related to a change in the coupling to c-axis phonons due to the rapid change of the c-axis screening in this doping range.Comment: 4 pages, 3 figure

    Ultraviolet photochemistry of 2-bromothiophene explored using universal ionization detection and multi-mass velocity-map imaging with a PImMS2 sensor

The raw ion events data and calculation log files can be retrieved from the University of Bristol’s research data repository and can be accessed using the following DOI:10.5523/bris.k35bi3pqsdbh2b5moo2e3puxf.The ultraviolet photochemistry of 2-bromothiophene (C4H3SBr) has been studied across the wavelength range 265-245 nm using a velocity-map imaging (VMI) apparatus recently modified for multi-mass imaging and vacuum ultraviolet (VUV, 118.2 nm) universal ionization. At all wavelengths, molecular products arising from the loss of atomic bromine were found to exhibit recoil velocities and anisotropies consistent with those reported elsewhere for the Br fragment [J. Chem. Phys. 142, 224303 (2015)]. Comparison between the momentum distributions of the Br and C4H3S fragments suggests that bromine is formed primarily in its ground (2P3/2) spin-orbit state. These distributions match well at high momentum, but relatively fewer slow moving molecular fragments were detected. This is explained by the observation of a second substantial ionic product, C3H3+. Analysis of ion images recorded simultaneously for several ion masses and the results of high-level ab initio calculations suggest that this fragment ion arises from dissociative ionization (by the VUV probe laser) of the most internally excited C4H3S fragments. This study provides an excellent benchmark for the recently modified VMI instrumentation and offers a powerful demonstration of the emerging field of multi-mass VMI using event-triggered, high frame-rate sensors, and universal ionization.Publisher PDFPeer reviewe

    Positive adjacency effects mediated by seed disperser birds in pine plantations

    This study examines the consequences of adjacent elements for a given patch, through their effects on zoochorous dispersion by frugivorous birds. The case study consists of pine plantations (the focal patch) adjacent to other patches of native vegetation (mixed patches of native forest and shrublands), and/or pine plantations. Our hypothesis is that input of native woody species propagules generated by frugivorous birds within plantations strongly depends on the nature of the surrounding vegetation. To test this hypothesis, we studied frugivorous-bird abundance, seed dispersion, and seedling establishment in nine pine plantation plots in contact with patches of native vegetation. To quantify adjacency arrangement effects, we used the percentage of common border between a patch and each of its adjacent elements. Frugivorous bird occurrence in pine plantations is influenced by the adjacent vegetation: the greater the contact with native vegetation patches, the more abundant were the frugivorous birds within pine plantations. Furthermore, frugivorous birds introduce into plantations the seeds of a large sample of native fleshy-fruited species. The results confirm the hypothesis that zoochorous seed rain is strongly determined by the kind of vegetation surrounding a given plantation. This finding underlines the importance of the composition of the mosaic surrounding plantations and the availability of mobile link species as key landscape features conditioning passive restoration processes

    Routine exercise-based cardiac rehabilitation does not increase aerobic fitness: A CARE CR study

    Background Recent evidence suggests that routine exercise-based cardiac rehabilitation (CR) may not lead to a substantial increase in estimated peak oxygen uptake (V̇O2peak). This could reduce the potential benefits of CR and explain why CR no longer improves patient survival in recent studies. We aimed to determine whether routine exercise-based CR increases V̇O2peak using gold-standard maximal cardiopulmonary exercise testing (CPET), and to quantify the exercise training stimulus which might be insufficient in patients undertaking CR. Methods We studied the effects of a routine, twice weekly, exercise-based CR programme for eight weeks (intervention group) compared with abstention from supervised exercise training (control group) in patients with coronary heart disease. The primary outcome was V̇O2peak measured using CPET. We also measured changes in body composition using dual X-ray absorptiometry, carotid intima-media thickness, hs-CRP and N-terminal pro B-type natriuretic peptide at baseline, 10 weeks and one year. We also calculated the Calibre 5-year all-cause mortality risk score. Results Seventy patients (age 63.1 SD10.0 years; BMI 29.2 SD4.0 kg·m−2; 86% male) were recruited (n = 48 intervention; n = 22 controls). The mean aerobic exercise training duration was 23 min per training session, and the mean exercise training intensity was 45.9% of heart rate reserve. V̇O2peak was 23·3 ml·kg-1·min−1 at baseline, and there were no changes in V̇O2peak between groups at any time point. The intervention had no effect on any of the secondary endpoints. Conclusion Routine CR does not lead to an increase in V̇O2peak and is unlikely to improve long-term outcomes
