West Nile virus in overwintering mosquitoes, central Europe
Background: West Nile virus (WNV) is currently the most important mosquito-borne pathogen spreading in Europe. Data on overwintering of WNV in mosquitoes are crucial for understanding WNV circulation in Europe; nonetheless, such data were not available so far.Results: A total of 28,287 hibernating mosquitoes [27,872 Culex pipiens, 73 Anopheles maculipennis (sensu lato), and 342 Culiseta annulata], caught in February or March between 2011 and 2017 in a WNV-endemic region of South Moravia, Czech Republic, were screened for the presence of WNV RNA. No WNV positive pools were found from 2011 to 2016, while lineage 2 WNV RNA was detected in three pools of Culex pipens mosquitoes collected in 2017 at two study sites.Conclusions: To the best of our knowledge, this is the first record of WNV RNA in overwintering mosquitoes in Europe. The data support the hypothesis of WNV persistence in mosquitoes throughout the winter season in Europe
Ixodes ricinus and Its Transmitted Pathogens in Urban and Peri-Urban Areas in Europe: New Hazards and Relevance for Public Health.
Tick-borne diseases represent major public and animal health issues worldwide. Ixodes ricinus, primarily associated with deciduous and mixed forests, is the principal vector of causative agents of viral, bacterial, and protozoan zoonotic diseases in Europe. Recently, abundant tick populations have been observed in European urban green areas, which are of public health relevance due to the exposure of humans and domesticated animals to potentially infected ticks. In urban habitats, small and medium-sized mammals, birds, companion animals (dogs and cats), and larger mammals (roe deer and wild boar) play a role in maintenance of tick populations and as reservoirs of tick-borne pathogens. Presence of ticks infected with tick-borne encephalitis virus and high prevalence of ticks infected with Borrelia burgdorferi s.l., causing Lyme borreliosis, have been reported from urbanized areas in Europe. Emerging pathogens, including bacteria of the order Rickettsiales (Anaplasma phagocytophilum, "Candidatus Neoehrlichia mikurensis," Rickettsia helvetica, and R. monacensis), Borrelia miyamotoi, and protozoans (Babesia divergens, B. venatorum, and B. microti) have also been detected in urban tick populations. Understanding the ecology of ticks and their associations with hosts in a European urbanized environment is crucial to quantify parameters necessary for risk pre-assessment and identification of public health strategies for control and prevention of tick-borne diseases
Preparation and Characterisation of Highly Stable Iron Oxide Nanoparticles for Magnetic Resonance Imaging
Magnetic nanoparticles produced using aqueous coprecipitation usually exhibit wide particle size distribution. Synthesis of small and uniform magnetic nanoparticles has been the subject of extensive research over recent years. Sufficiently small superparamagnetic iron oxide nanoparticles easily permeate tissues and may enhance the contrast in magnetic resonance imaging. Furthermore, their unique small size also allows them to migrate into cells and other body compartments. To better control their synthesis, a chemical coprecipitation protocol was carefully optimised regarding the influence of the injection rate of base and incubation times. The citrate-stabilised particles were produced with a narrow average size range below 2nm and excellent stability. The stability of nanoparticles was monitored by long-term measurement of zeta potentials and relaxivity. Biocompatibility was tested on the Caki-2 cells with good tolerance. The application of nanoparticles for magnetic resonance imaging (MRI) was then evaluated. Increasing Incidence of Geomyces destructans Fungus in Bats from the Czech Republic and Slovakia
BACKGROUND: White-nose syndrome is a disease of hibernating insectivorous bats associated with the fungus Geomyces destructans. It first appeared in North America in 2006, where over a million bats died since then. In Europe, G. destructans was first identified in France in 2009. Its distribution, infection dynamics, and effects on hibernating bats in Europe are largely unknown. METHODOLOGY/PRINCIPAL FINDINGS: We screened hibernacula in the Czech Republic and Slovakia for the presence of the fungus during the winter seasons of 2008/2009 and 2009/2010. In winter 2009/2010, we found infected bats in 76 out of 98 surveyed sites, in which the majority had been previously negative. A photographic record of over 6000 hibernating bats, taken since 1994, revealed bats with fungal growths since 1995; however, the incidence of such bats increased in Myotis myotis from 2% in 2007 to 14% by 2010. Microscopic, cultivation and molecular genetic evaluations confirmed the identity of the recently sampled fungus as G. destructans, and demonstrated its continuous distribution in the studied area. At the end of the hibernation season we recorded pathologic changes in the skin of the affected bats, from which the fungus was isolated. We registered no mass mortality caused by the fungus, and the recorded population decline in the last two years of the most affected species, M. myotis, is within the population trend prediction interval. CONCLUSIONS/SIGNIFICANCE: G. destructans was found to be widespread in the Czech Republic and Slovakia, with an epizootic incidence in bats during the most recent years. Further development of the situation urgently requires a detailed pan-European monitoring scheme
Investigations of diseases with natural focality in Biospheric Protection Area Pálava
Long-term studies of the Medical Zoology Laboratory at the Institute of Vertebrate Biology of the Academy of Sciences of the Czech Republic concentrate on natural foci of diseases transmitted by haematophagous arthropods in this area, e.g., Valtice fever (the etiologic agent is mosquito-borne ahyňa virus), Lyme borreliosis (the agent is tick-borne Borrelia burgdorferi sensu lato) or tularemia (the agent is Francisella tularensis that is occasionally transmitted also by ixodid ticks). We detected newly the activity of mosquito-borne West Nile virus in this area, and additional tick-borne pathogens might be expected (Anaplasma phagocytophilum, Babesia microti). In addition, our laboratory has investigated the ecology of some non-tramissible diseases, e.g. avian botulism, salmonellosis in gulls and emmonsiosis in rodents; the latter two diseases also occur in humans
Detection of antibodies against tick-borne encephalitis in human sera by screening in neutralization test
Using plaque-reduction neutralization test, we examined blood sera of 704 persons for antibodies against tick-borne encephalitis virus. Specific antibodies were detected in 121 persons. Detailed results were delivered to National Public Health Institute in Prague according to the agreement
Contribution to canine babesiosis in the Czech Republic
From March to November 2010, a total of 68 samples of blood from 41 hunting and working dogs that never left the Czech Republic were examined. Some dogs were sampled repeatedly. Blood samples were examined by polymerase chain reaction for the presence of DNA of piroplasms with negative results. Specific IgG antibodies against Babesia canis were detected by indirect immunofluorescence test, and five dogs (12.21%) were seropositive. Titres ranged from 50 to 200. One dog was positive in two samplings within 3 months. The highest number of positive samples was taken in June. The results of this study suggest a likely contact of the examined dogs with the parasite; although in 2005, a total of 340 adult unfed Dermacentor reticulatus ticks in 34 pools screened by PCR for babesiae were negative
