    The Inventory of Personality Organization: A valid instrument to detect the severity of personality dysfunction

    Background and aims In the eleventh revision of the International Classification of Diseases (ICD-11), the severity of personality dysfunction became the central dimension of personality disorder’s (PDs) definition, besides the trait domain qualifiers. Personality functioning, also known as personality organization (PO), is becoming an increasingly important concept in administering, predicting, and measuring severity and nature of personality disturbance. Otto Kernberg and his team developed several tools to measure personality impairment. The Inventory of Personality Organization (IPO) is a self-report rating scale for the measurement of PO. Aim of this study was to identify severity groups according to the level of PO and to explore their validity. Materials and methods A clinical sample of 118 patients was recruited from a 4-weeks in-patient cognitive psychotherapy program. Beside the IPO, Structured Clinical Interview for the fourth edition of the Diagnostic and Statistical Manual of Mental Disorders, (DSM-IV.) Axis I and II, Symptom Check List-90 (SCL-90), State-Trait Anger Expression Inventory and Dissociative Experience scale (DES). Two types of analyses were conducted: a person-centered (latent profile) analysis and various variable-centered tests to confirm the factor structure of IPO and calculate group differences. Results The three-factor (CFI = 0.990, TLI = 0.990, RMSEA = 0.022, SRMR = 0.089) and the five-factor (CFI = 0.995, TLI = 0.995, RMSEA = 0.014, SRMR = 0.090) models of the IPO was supported. Latent class analysis identified three subgroups of PO: “Well-integrated,” “Moderately integrated,” and “Disintegrated” classes. There were no significant differences between the three classes in the number of Axis 1 diagnoses (p = 0.354; η2 = 0.01). Group differences in the number of PDs, the number of PD symptoms as well as in the presence of borderline and depressive PD were significant (all p < 0.001; V = 0.35–0.42; η2 = 0.15–0.26). Persons with more severe PO problem level had higher rates of psychopathological symptoms, state and trait anger, and dissociative characteristics (all p < 0.001; η2 = 0.13–0.36). Conclusion The IPO can be an appropriate instrument to measure the severity of personality disorganization and to classify participants along a continuum of severity in this regard. Our results present further evidence that the severity of personality dysfunction, the central dimension of the ICD-11 and the Alternative Model for PDs is detectable with an instrument, the IPO, that was initially developed to detect the disturbances in PO.Peer Reviewe

    Relating Compulsivity and Impulsivity With Severity of Behavioral Addictions: A Dynamic Interpretation of Large-Scale Cross-Sectional Findings

    Background and Aim: Impulsivity and compulsivity are two key temperament traits involved in behavior regulation. The aim of this study was to test several existing theories in explaining the role of impulsivity and compulsivity in symptom severity in various behavioral addictions. Methods: Data were collected from a (representative) general population sample (N = 2,710, mean age:39.8 years (SD:13.6), 51% woman), and from people who are at increased risk of having a behavioral addiction (N = 9,528 in total, mean age: 28.11 (SD:8.3), 34.3% woman), including people with problematic gaming and internet use, pathological gambling, exercise dependence, compulsive buying and work addiction. Symptom severity, reward driven impulsivity and relief driven compulsivity were assessed. Results: For non-problematic groups, impulsivity is present to about the same extent as compulsivity, whereas for problematic groups, compulsivity dominates over impulsivity in all groups (except for gambling). The strength of the correlation between impulsivity and compulsivity is higher in more severe forms of the disorders (from r = 0.18 to r = 0.59 in the representative population). Discussion: Based on these data, it appears that relief-driven behavior (negative reinforcement) dominates over reward-driven behavior (positive reinforcement) in more severe cases of a behavioral addiction. Conclusion: This is the first large-scale study to find empirical support for the neuroscientific theory on the dominance of compulsivity (“needing”) over impulsivity (“wanting”) in more severe cases of a behavioral addiction. Although longitudinal research is needed, a possible shift from impulsivity to compulsivity takes place, similar to substance use addictions, which maintains the circle of addiction.Peer Reviewe

    Human mobility in a Bronze Age Vatya ‘urnfield’ and the life history of a high-status woman

    In this study, we present osteological and strontium isotope data of 29 individuals (26 cremations and 3 inhumations) from Szigetszentmiklós-Ürgehegy, one of the largest Middle Bronze Age cemeteries in Hungary. The site is located in the northern part of the Csepel Island (a few kilometres south of Budapest) and was in use between c. 2150 and 1500 BC, a period that saw the rise, the apogee, and, ultimately, the collapse of the Vatya culture in the plains of Central Hungary. The main aim of our study was to identify variation in mobility patterns among individuals of different sex/age/social status and among individuals treated with different burial rites using strontium isotope analysis. Changes in funerary rituals in Hungary have traditionally been associated with the crises of the tell cultures and the introgression of newcomers from the area of the Tumulus Culture in Central Europe around 1500 BC. Our results show only slight discrepancies between inhumations and cremations, as well as differences between adult males and females. The case of the richly furnished grave n. 241 is of particular interest. The urn contains the cremated bones of an adult woman and two 7 to 8-month-old foetuses, as well as remarkably prestigious goods. Using 87Sr/86Sr analysis of different dental and skeletal remains, which form in different life stages, we were able to reconstruct the potential movements of this high-status woman over almost her entire lifetime, from birth to her final days. Our study confirms the informative potential of strontium isotopes analyses performed on different cremated tissues. From a more general, historical perspective, our results reinforce the idea that exogamic practices were common in Bronze Age Central Europe and that kinship ties among high-rank individuals were probably functional in establishing or strengthening interconnections, alliances, and economic partnerships

    Lipid core peptide (LCP)-based group A streptococcal (GAS) vaccines incorporating numerous GAS M-protein epitopes have been synthesized in high purity. The LCP system incorporates adjuvant, carrier, and antigens into a single well-characterized construct. A tetra-epitopic vaccine, incorporating a highly conserved GAS M-protein C-terminal epitope and three strain-specific epitopes from the variable GAS M-protein N-terminal region, was synthesized using stepwise solid-phase peptide synthesis. Immunization of mice with this construct yielded high IgG antibody responses without the use of traditional adjuvants

    Potential of Lipid Core Peptide Technology as a Novel Self-Adjuvanting Vaccine Delivery System for Multiple Different Synthetic Peptide Immunogens

    This study demonstrates the effectiveness of a novel self-adjuvanting vaccine delivery system for multiple different synthetic peptide immunogens by use of lipid core peptide (LCP) technology. An LCP formulation incorporating two different protective epitopes of the surface antiphagocytic M protein of group A streptococci (GAS)—the causative agents of rheumatic fever and subsequent rheumatic heart disease—was tested in a murine parenteral immunization and GAS challenge model. Mice were immunized with the LCP-GAS formulation, which contains an M protein amino-terminal type-specific peptide sequence (8830) in combination with a conserved non-host-cross-reactive carboxy-terminal C-region peptide sequence (J8) of the M protein. Our data demonstrated immunogenicity of the LCP-8830-J8 formulation in B10.BR mice when coadministered in complete Freund's adjuvant and in the absence of a conventional adjuvant. In both cases, immunization led to induction of high-titer GAS peptide-specific serum immunoglobulin G antibody responses and induction of highly opsonic antibodies that did not cross-react with human heart tissue proteins. Moreover, mice were completely protected from GAS infection when immunized with LCP-8830-J8 in the presence or absence of a conventional adjuvant. Mice were not protected, however, following immunization with an LCP formulation containing a control peptide from a Schistosoma sp. These data support the potential of LCP technology in the development of novel self-adjuvanting multi-antigen component vaccines and point to the potential application of this system in the development of human vaccines against infectious diseases

    Increased Tau Phosphorylation and Impaired Presynaptic Function in Hypertriglyceridemic ApoB-100 Transgenic Mice

    AIMS: ApoB-100 is the major protein component of cholesterol- and triglyceride-rich LDL and VLDL lipoproteins in the serum. Previously, we generated and partially described transgenic mice overexpressing the human ApoB-100 protein. Here, we further characterize this transgenic strain in order to reveal a possible link between hypeprlipidemia and neurodegeneration. METHODS AND RESULTS: We analyzed the serum and cerebral lipid profiles, tau phosphorylation patterns, amyloid plaque-formation, neuronal apoptosis and synaptic plasticity of young (3 month old), adult (6 month old) and aging (10–11 month old) transgenic mice. We show that ApoB-100 transgenic animals present i) elevated serum and cerebral levels of triglycerides and ApoB-100, ii) increased cerebral tau phosphorylation at phosphosites Ser(199), Ser(199/202), Ser(396) and Ser(404). Furthermore, we demonstrate, that tau hyperphosphorylation is accompanied by impaired presynaptic function, long-term potentiation and widespread hippocampal neuronal apoptosis. CONCLUSIONS: The results presented here indicate that elevated ApoB-100 level and the consequent chronic hypertriglyceridemia may lead to impaired neuronal function and neurodegeneration, possibly via hyperphosphorylation of tau protein. On account of their specific phenotype, ApoB-100 transgenic mice may be considered a versatile model of hyperlipidemia-induced age-related neurodegeneration
