Existuje velké množství ledvinných transportérů, nicméně tato diplomová práce je zaměřena především na megalin. Konkrétně jsme se zabývali stanovením transportu peptidů přes tento renální transportér v buněčných liniích HK-2 a JEG-3. Megalin je glykoprotein, jehož hlavní funkcí je tubulární reabsorpce glomerulárně filtrovaného albuminu a dalších vysokomolekulárních látek. Mezi endogenní substráty megalinu dále patří např. transkobalamin - vitamin B12, inzulin nebo lehké řetězce imunoglobulinů. Je také schopen transportovat některá léčiva, jako jsou aminoglykosidová a peptidová antibiotika. V rámci této práce jsme zkoumali vliv snížení exprese megalinu na transport albuminu a anti-VEGFR2 aptameru. Snížení exprese bylo dosaženo transfekcí buněk pomocí siRNA proti LRP2 genu. Nižší exprese LRP2 vede ke snížení transportu albuminu a anti- VEGFR2 aptameru, což bylo následně stanoveno pomocí akumulačních studií. Plenty of kidney transporters have been identified, however this diploma thesis is focused on megalin. Specifically, we dealed with determination of transport of peptides via this renal transporter in HK-2 and JEG-3 cell lines. Megalin is a glycoprotein whose main function is tubular reabsorption of albumin filtered in glomerules and other high molecular weight substances. Another endogenous substrates of megalin are e.g. transcoblamine - vitamin B12, insulin or light chains of immunoglobulins. It is also able to transport some drugs like aminoglycoside and polypeptide antibiotics. Within this work, we investigated the influence of reduced expression of megalin on the transport of albumin and anti-VEGFR2 aptamer. Geocaching and its contribution to the tourism industry in the West Bohemia

Diplomová práce se zabývá vlivem geocachingu na cestovní ruch v Západočeském kraji. Teoretická část rozebírá cestovní ruch a poté geocaching, jeho historii, pravidla a hlavní principy. Následně je rozebrán geocaching z pohledu cestovního ruchu. V praktické části je proveden smíšený výzkum, který zkoumá vliv geocachingu na účastníky cestovního ruchu. Cílem práce je analyzovat geocaching jako možný nástroj pro rozvoj cestovního ruchu v Západočeském kraji prostřednictvím kvantitativního a kvalitativního výzkumu. Perception of the moral acceptability and riskiness of selected addictive substances among the adult population

The topic of this bachelor thesis is the perception of moral acceptability and riskiness of selected addictive substances in the adult population. The aim of this thesis is to describe the relationship between perception of moral acceptability and perception of riskiness of using selected substances between adult respondents based on a questionnaire survey. The research also attempts to map how attitudes differ in relation to gender, education, respondents' region of residence and their personal experience with addictive substances. The research sample consisted of 172 respondents, 128 women and 43 men. A minimum age of 18 years and voluntary participation were the prerequisites for entering the study. Respondents were selected by self-selection and snowball sampling methods. The thesis is based on the findings obtained through quantitative research, and the research instrument was an online questionnaire. The questionnaire was divided into four sections: basic socio-demographic data of the respondents, questions related to moral acceptability, questions related to the riskiness of substance use and additional questions. Data were then analyzed using Excel and Statistica 14. Mann-Whitney U test, Kruskal- Wallis test and Spearman correlation coefficient were used for analysis. The results show that...Tématem této bakalářské práce je vnímání morální přijatelnosti a rizikovosti vybraných návykových látek u dospělé populace. Cílem práce je popsat vztah mezi vnímáním morální přijatelnosti a vnímáním rizikovosti užívání vybraných návykových látek u dospělých respondentů na základě dotazníkového šetření. Výzkum se též pokouší mapovat, jak se postoje liší v souvislosti s pohlavím, vzděláním, krajem bydliště respondentů a jejich osobní zkušeností s návykovými látkami. Výzkumný soubor tvořilo 172 respondentů, z toho 128 žen a 43 mužů. Podmínkou pro vstup do výzkumu byl minimální věk 18 let a dobrovolná účast. Respondenti byli vybíráni metodou samovýběru a metodou sněhové koule. Práce vychází z poznatků získaných prostřednictvím kvantitativního výzkumu, přičemž výzkumným nástrojem byl online dotazník. Dotazník byl rozdělen do čtyř sekcí: základní sociodemografické údaje respondentů, otázky týkající se morální přijatelnosti, otázky týkající se rizikovosti užívání návykových látek a doplňující otázky. Data byla následně analyzována pomocí programů Excel a Statistica 14. K analýze byl využit Mann- Whitneyův U test, Kruskalův-Wallisův test a Spearmanův korelační koeficient. 1 Defence by the defence counsel in the trial Abstract The aim of this thesis is not only to analyse the legal regulation concerning the defence counsel and the defence in the main trial, and to provide the reader with a de lege lata view on this issue, but also to provide de lege ferenda considerations for some legal institutes. The thesis also provides the reader with a comparison of the current legal regulation and the current version of the draft of the new Criminal Procedure Code (as of 14 October 2022) on selected criminal procedure issues. The content of the thesis is divided into an introduction, four parts and a conclusion. The introduction briefly justifies the choice of this topic and sets out the objectives to be achieved in the thesis. The importance of the right to a defence and the importance of the person of the defence counsel is emphasised. The first part of the thesis defines the right of defence, emphasises the fundamental human rights and freedoms in criminal proceedings and the basic principles of criminal procedure, among which the principle of ensuring the right of defence is emphasised. Diplomová práce dále poskytuje čtenáři komparaci současné právní úpravy a aktuální verze návrhu nového trestního řádu (stav ke dni 14. 10. 2022) u vybraných trestněprocesních otázek. Analysis of Virtual Team Management

Cílem práce je pojmenovat a popsat specifika a možná rizika práce virtuálních týmů a navrhnout opatření, jimiž by se dala eliminovat nebo omezit. Práce je rozdělena na část teoretickou a část praktickou. Teoretická část je zaměřena na charakteristiku virtuální týmové práce a zasazuje ji do kontextu řízení lidských zdrojů. V praktické části je pomocí několika výzkumných metod provedena analýza současného stavu týmové spolupráce v konkrétním virtuálním týmu. V závěru je představen soubor doporučení, jež povedou ke zlepšení efektivnosti virtuálního týmu.The aim of the thesis is to identify and describe the specifics and possible risks of the work of virtual teams and to propose measures that could eliminate or reduce there risks. The work is divided into a theoretical part and a practical part. The theoretical part focuses on the characteristics of virtual teamwork and places it in the context of human resource management. In the practical part, an analysis of the current state of teamwork in a particular virtual team is performed using several research methods. Description, Action, or Attitude? A Translator’s Remark on Two Novels by Romesh Gunesekera and Chitra Banerjee Divakaruni

Combining translation and linguistic studies, the article introduces two less-known immigrant writers Romesh Gunesekera and Chitra Banerjee Divakaruni and their novels Reef and The Mistress of Spices, which were both translated by the author. The novels reflect the social and political problems in Sri Lanka and India which make their citizens flee abroad. The stories are well narrated by poetic language and brought their authors international recognition. Especially the descriptive passages in both novels are very impressive and are considered here as specific language structures. A partial analysis of typical sentence elements of such passages in comparison with their Czech equivalents is the main aim of this article. THE ROLE OF SYNTACTIC STYLISTIC MEANS IN EXPRESSING THE EMOTION TERM LOVE

Love as one of important feelings in human emotional, cognitive and social life has always attracted attention of the researchers: psychologists, linguists, philosophers, ethnologists, etc. We may speak about extralinguistic and linguistic ways of love manifestation. To linguistic ones belong, of course, stylistic means, which include lexical, syntactic, phonetic, and semasiological level. The author focuses on lexical-syntactical means of expressing love in two Slavic languages, Czech and Slovak, using linguocognitive and cultural approach. Congenital Diarrhea and Cholestatic Liver Disease: Phenotypic Spectrum Associated with MYO5B Mutations

Myosin Vb (MYO5B) is a motor protein that facilitates protein trafficking and recycling in polarized cells by RAB11- and RAB8-dependent mechanisms. Biallelic MYO5B mutations are identified in the majority of patients with microvillus inclusion disease (MVID). MVID is an intractable diarrhea of infantile onset with characteristic histopathologic findings that requires life-long parenteral nutrition or intestinal transplantation. A large number of such patients eventually develop cholestatic liver disease. Bi-allelic MYO5B mutations are also identified in a subset of patients with predominant early-onset cholestatic liver disease. We present here the compilation of 114 patients with disease-causing MYO5B genotypes, including 44 novel patients as well as 35 novel MYO5B mutations, and an analysis of MYO5B mutations with regard to functional consequences. Our data support the concept that (1) a complete lack of MYO5B protein or early MYO5B truncation causes predominant intestinal disease (MYO5B-MVID), (2) the expression of full-length mutant MYO5B proteins with residual function causes predominant cholestatic liver disease (MYO5B-PFIC), and (3) the expression of mutant MYO5B proteins without residual function causes both intestinal and hepatic disease (MYO5B-MIXED).
