New method for non-standard invisible particle searches in tau lepton decays
Motivated by models proposed to explain the Standard Model anomalies, and the
unprecedented data to be collected by the Belle~II
experiment during the next years, we study the kinematics of tau pair decays
and propose a new method to search for lepton flavor violating processes in tau
lepton decays to invisible beyond Standard Model particles, such as , where is either an electron or a muon, and is a
massive particle that escapes undetected. The new method improves by one order
of magnitude the expected upper limit on the production
in 3x1 prong tau decays and establishes the possibility of performing this
search in 1x1 prong tau decays which has not been previously considered.Comment: 6 pages, 7 figure
The enzymes LSD1 and Set1A cooperate with the viral protein HBx to establish an active hepatitis B viral chromatin state
Indexación: Web of ScienceWith about 350 million people chronically infected around the world hepatitis B is a major health problem. Template for progeny HBV synthesis is the viral genome, organized as a minichromosome (cccDNA) inside the hepatocyte nucleus. How viral cccDNA gene expression is regulated by its chromatin structure; more importantly, how the modulation of this structure impacts on viral gene expression remains elusive. Here, we found that the enzyme SetDB1 contributes to setting up a repressed cccDNA chromatin state. This repressive state is activated by the histone lysine demethylase-1 (LSD1). Consistently, inhibiting or reducing LSD1 levels led to repression of viral gene expression. This correlates with the transcriptionally repressive mark H3K9 methylation and reduction on the activating marks H3 acetylation and H3K4 methylation on viral promoters. Investigating the importance of viral proteins we found that LSD1 recruitment to viral promoters was dependent on the viral transactivator protein HBx. Moreover, the histone methyltransferase Set1A and HBx are simultaneously bound to the core promoter, and Set1A expression correlates with cccDNA H3K4 methylation. Our results shed light on the mechanisms of HBV regulation mediated by the cccDNA chromatin structure, offering new therapeutic targets to develop drugs for the treatment of chronically infected HBV patients.http://www.nature.com/articles/srep2590
Transcription factors Sp1 and p73 control the expression of the proapoptotic protein NOXA in the response of testicular embryonal carcinoma cells to cisplatin
Testicular germ cell tumors (TGCTs) are highly responsive to and curable by cisplatin-based chemotherapy even in advanced stages. We have studied the molecular mechanisms involved in the induction of apoptosis in response to cisplatin, and found that proapoptotic Noxa is transcriptionally up-regulated following cisplatin exposure, even in the absence of p53, in NTERA2 cisplatin-sensitive cells but not in 1411HP-resistant cells. Blockade of Noxa reduced the apoptotic response of embryonal carcinoma (EC) NTERA2 cells to cisplatin. A detailed analysis of the Noxa promoter revealed that p73 and Sp1-like factors, Sp1 and KLF6, played key roles in the transcriptional control of this gene. Overexpression of TAp73 induced Noxa whereas the dominant negative isoform ΔNp73, reduced the levels of Noxa after cisplatin exposure in NTERA2 and 2102EP. Interestingly, down-regulation of Sp1 increased Noxa expression in response to cisplatin. However, blockade of KLF6 decreased cisplatin-induced up-regulation of Noxa in EC cell lines. In addition, tissue microarray analyses of TGCTs revealed that expression of Noxa correlates with good clinical prognosis in patients with embryonal carcinoma. Thus, our data show the transcriptional network that regulates Noxa in EC cells, which is key for their apoptotic response to cisplatin-based chemotherapy, and propose Noxa as a predictive factor of therapeutic response
Multicenter phase II study of matured dendritic cells pulsed with melanoma cell line lysates in patients with advanced melanoma

Abstract

Background

Several single center studies have provided evidence of immune activation and antitumor activity of therapeutic vaccination with dendritic cells (DC) in patients with metastatic melanoma. The efficacy of this approach in patients with favorable prognosis metastatic melanoma limited to the skin, subcutaneous tissues and lung (stages IIIc, M1a, M1b) was tested in a multicenter two stage phase 2 study with centralized DC manufacturing.

Methods

The vaccine (IDD-3) consisted 8 doses of autologous monocyte-derived matured DC generated in serum-free medium with granulocyte macrophage colony stimulating factor (GM-CSF) and interleukin-13 (IL-13), pulsed with lysates of three allogeneic melanoma cell lines, and matured with interferon gamma. The primary endpoint was antitumor activity.

Results

Among 33 patients who received IDD-3 there was one complete response (CR), two partial responses (PR), and six patients had stable disease (SD) lasting more than eight weeks. The overall prospectively defined tumor growth control rate was 27% (90% confidence interval of 13-46%). IDD-3 administration had minimal toxicity and it resulted in a high frequency of immune activation to immunizing melanoma antigens as assessed by <it>in vitro </it>immune monitoring assays.

Conclusions

The administration of matured DC loaded with tumor lysates has significant immunogenicity and antitumor activity in patients with limited metastatic melanoma.

Clinical trial registration

NCT00107159.</p
Training and Onboarding initiatives in High Energy Physics experiments
In this paper we document the current analysis software training and
onboarding activities in several High Energy Physics (HEP) experiments: ATLAS,
CMS, LHCb, Belle II and DUNE. Fast and efficient onboarding of new
collaboration members is increasingly important for HEP experiments as analyses
and the related software become ever more complex with growing datasets. A
meeting series was held by the HEP Software Foundation (HSF) in 2022 for
experiments to showcase their initiatives. Here we document and analyse these
in an attempt to determine a set of key considerations for future experiments
First observations of oblique ionospheric sounding chirp signal in Mexico
The results of the first experiment of oblique ionospheric sounding (OIS) chirp signal reception in Mexico are
reported. Maximal and Lowest Observed Frequencies variations were studied under the quiet Space Weather
conditions. The diurnal ionospheric variations by OIS signal confirm the results based on GNSS data in the
Mexican region. The best HF radio propagation conditions along the considered path are during morning and
daytime hours. The multi-hop propagation is frequent. The interlayer propagation modes are present at nighttime
Aedes albopictus in northeast Mexico: An update on adult distribution and first report of parasitism by Ascogregarina taiwanensis
ABSTRACT Background & objectives: Aedes albopictus has been known as efficient vector of dengue in Asian countries and its wide displacement of Ae. aegypti has been documented in many parts of the world. The present survey was carried out to update the distribution of Ae. albopictus in northeast Mexico and to report the first record of parasitism of mosquitoes by Ascogregarina taiwanensis in Mexico
Interim Design Report
The International Design Study for the Neutrino Factory (the IDS-NF) was
established by the community at the ninth "International Workshop on Neutrino
Factories, super-beams, and beta- beams" which was held in Okayama in August
2007. The IDS-NF mandate is to deliver the Reference Design Report (RDR) for
the facility on the timescale of 2012/13. In addition, the mandate for the
study [3] requires an Interim Design Report to be delivered midway through the
project as a step on the way to the RDR. This document, the IDR, has two
functions: it marks the point in the IDS-NF at which the emphasis turns to the
engineering studies required to deliver the RDR and it documents baseline
concepts for the accelerator complex, the neutrino detectors, and the
instrumentation systems. The IDS-NF is, in essence, a site-independent study.
Example sites, CERN, FNAL, and RAL, have been identified to allow site-specific
issues to be addressed in the cost analysis that will be presented in the RDR.
The choice of example sites should not be interpreted as implying a preferred
choice of site for the facility
