    Practical guidelines for modelling post-entry spread in invasion ecology

    In this article we review a variety of methods to enable understanding and modelling the spread of a pest or pathogen post-entry. Building upon our experience of multidisciplinary research in this area, we propose practical guidelines and a framework for model development, to help with the application of mathematical modelling in the field of invasion ecology for post-entry spread. We evaluate the pros and cons of a range of methods, including references to examples of the methods in practice. We also show how issues of data deficiency and uncertainty can be addressed. The aim is to provide guidance to the reader on the most suitable elements to include in a model of post-entry dispersal in a risk assessment, under differing circumstances. We identify both the strengths and weaknesses of different methods and their application as part of a holistic, multidisciplinary approach to biosecurity research

    Genome wide expression profiling reveals suppression of host defence responses during colonisation by Neisseria meningitides but not N. lactamica.

    Both Neisseria meningitidis and the closely related bacterium Neisseria lactamica colonise human nasopharyngeal mucosal surface, but only N. meningitidis invades the bloodstream to cause potentially life-threatening meningitis and septicaemia. We have hypothesised that the two neisserial species differentially modulate host respiratory epithelial cell gene expression reflecting their disease potential. Confluent monolayers of 16HBE14 human bronchial epithelial cells were exposed to live and/or dead N. meningitidis (including capsule and pili mutants) and N. lactamica, and their transcriptomes were compared using whole genome microarrays. Changes in expression of selected genes were subsequently validated using Q-RT-PCR and ELISAs. Live N. meningitidis and N. lactamica induced genes involved in host energy production processes suggesting that both bacterial species utilise host resources. N. meningitidis infection was associated with down-regulation of host defence genes. N. lactamica, relative to N. meningitidis, initiates up-regulation of proinflammatory genes. Bacterial secreted proteins alone induced some of the changes observed. The results suggest N. meningitidis and N. lactamica differentially regulate host respiratory epithelial cell gene expression through colonisation and/or protein secretion, and that this may contribute to subsequent clinical outcomes associated with these bacteria

    Hydrogen-powered road vehicles : the health benfits and drawbacks of a new fuel

    Because of the political, social and environmental problems associated with dependency on fossil fuels, there is considerable interest in alternative energy sources. Hydrogen is regarded as a promising option, particularly as a fuel for road vehicles. The Dutch Energy Research Centre (ECN) recently published a vision of the future, in which it suggested that by 2050 more than half of all cars in the Netherlands could be running on hydrogen. A switch to using hydrogen as the primary energy source for road vehicles would have far-reaching social consequences. As with all technological developments, opportunities would be created, but drawbacks would inevitably be encountered as well. Some of the disadvantages associated with hydrogen are already known, and are to some degree manageable. It is likely, however, that other drawbacks would come to light only once hydrogen-powered cars were actually in use. With that thought in mind, and in view of the social significance of a possible transition to hydrogen, it was decided that the Health Council should assess the positive and negative effects that hydrogen use could have on public health. It is particularly important to make such an assessment at the present early stage in the development of hydrogen technologies, so that gaps in existing scientific knowledge may be identified and appropriate strategies may be developed for addressing such gaps.     Effective professional development remains a major focus for universities, educational agencies, school districts and educators. Successful staff development projects for educators provide long-range effects in which administrators, communities, universities, students and even parents eventually receive benefits. Quality professional development encompasses a wide range of opportunities for the purpose of enhancing educator performance and excellence. This article discusses the organization and implementation of a summit designed to target an audience of school media personnel. Additionally, the paper presents the organization and implementation of the summit. In the planning of any professional development activity, two equally important tasks emerge (1) developing the program and (2) selecting the targeted participants. The development of the program is based on the needs of the targeted audience; therefore, the targeted audience must be determined prior to the beginning of the planning process. Several additional factors are critical in the success of a staff development event. It is equally essential to have clear library administrative support of the project and have a wide-range of professional contacts for identifying and recruiting experts to facilitate sessions

    NADPH as a potential intrinsic probe for tumour margin estimation

    The fluorescent properties of the reduced coenzyme NADH and its phosphorylated derivative (NADPH) have been explored in order to assess their potential as an intrinsic probe for cancer surgery. NADPH production is increased in cancer cells to quench reactive oxygen species and meet higher demands for biosynthesis, and has attractive fluorescent properties such as emission towards the visible part of the spectrum and a relatively long fluorescence lifetime upon binding to enzymes (~ 1 – 6.5 ns) that helps discriminate against other endogenous species. Different environmental effects on NAD(P)H fluorescence are reported here, including an increase in lifetime upon oxygen removal, an ability to retain its fluorescent properties in a complex medium (a silica phantom) and its fluorescence lifetime also being distinguishable in a cell environment. In addition, the development of a miniaturized liquid light guide filter-based timecorrelated single photon counting fluorescence lifetime system is reported as a step towards time-resolved visual imaging in cancer surgery. This system has been demonstrated as being capable of accurately measuring NAD(P)H fluorescence lifetimes in both simple solvent and cellular environments
