Branched Chain Amino Acids Promote ATP Production Via Translocation of Glucose Transporters
Purpose: We have previously shown that maintenance of ATP levels is a promising strategy for preventing neuronal cell death, and that branched chain amino acids (BCAAs) enhanced cellular ATP levels in cultured cells and antagonized cell death. BCAAs attenuated photoreceptor degeneration and retinal ganglion cell death in rodent models of retinal degeneration or glaucoma. This study aimed to elucidate the mechanisms through which BCAAs enhance ATP production. Methods: Intracellular ATP concentration was measured in HeLa cells under glycolysis and citric acid cycle inhibited conditions. Next, glucose uptake was quantified in HeLa cells and in 661W retinal photoreceptor-derived cells under glycolysis inhibition, endoplasmic reticulum stress, and glucose transporters (GLUTs) inhibited conditions, by measuring the fluorescence of fluorescently labeled deoxy-glucose analog using flow cytometry. Then, the intracellular behavior of GLUT1 and GLUT3 were observed in HeLa or 661W cells transfected with enhanced green fluorescent protein-GLUTs. Results: BCAAs recovered intracellular ATP levels during glycolysis inhibition and during citric acid cycle inhibition. BCAAs significantly increased glucose uptake and recovered decreased glucose uptake induced by endoplasmic reticulum stress or glycolysis inhibition. However, BCAAs were unable to increase intracellular ATP levels or glucose uptake when GLUTs were inhibited. Fluorescence microscopy revealed that supplementation of BCAAs enhanced the translocation of GLUTs proteins to the plasma membrane over time. Conclusions: BCAAs increase ATP production by promoting glucose uptake through promotion of glucose transporters translocation to the plasma membrane. These results may help expand the clinical application of BCAAs in retinal neurodegenerative diseases, such as glaucoma and retinal degeneration

Simulating second-generation herbaceous bioenergy crop yield using the global hydrological model H08 (v.bio1)
Large-scale deployment of bioenergy plantations would have adverse effects on water resources. There is an increasing need to ensure the appropriate inclusion of the bioenergy crops in global hydrological models. Here, through parameter calibration and algorithm improvement, we enhanced the global hydrological model H08 to simulate the bioenergy yield from two dedicated herbaceous bioenergy crops: Miscanthus and switchgrass. Site-specific evaluations showed that the enhanced model had the ability to simulate yield for both Miscanthus and switchgrass, with the calibrated yields being well within the ranges of the observed yield. Independent country-specific evaluations further confirmed the performance of the H08 (v.bio1). Using this improved model, we found that unconstrained irrigation more than doubled the yield under rainfed condition, but reduced the water use efficiency (WUE) by 32â% globally. With irrigation, the yield in dry climate zones can exceed the rainfed yields in tropical climate zones. Nevertheless, due to the low water consumption in tropical areas, the highest WUE was found in tropical climate zones, regardless of whether the crop was irrigated. Systemic Lymphadenectomy Cannot Be Recommended for Low-Risk Corpus Cancer
Objective. The objective of this study is to ascertain whether omission of lymphadenectomy could be possible when uterine corpus cancer is considered low-risk based on intraoperative pathologic indicators. Patient and Methods. Between 1998 and 2007, a total of 83 patients with low risk corpus cancer (endometrioid type, grade 1 or 2, myometrial invasion âŠ50%, and no intraoperative evidence of macroscopic extrauterine spread, including pelvic and paraaortic lymph node swelling and adnexal metastasis) underwent the total abdominal hysterectomy and bilateral salpingo-oophorectomy without lymphadenectomy. A retrospective review of the medical records was performed, and the disease-free survival (DFS), overall survival (OS), peri- and postoperative morbidities and complications were evaluated. Results. The 5-year DFS rates and the 5-year OS rates were 97.6% and 98.8%, respectively. No patient presented postoperative leg lymphedema and deep venous thrombosis. Conclusion. Omission of lymphadenectomy did not worsen the DFS or OS. The present findings suggest that systemic lymphadenectomy could be omitted in low-risk endometrial carcinoma
How many hot days and heavy precipitation days will grandchildren experience that break the records set in their grandparentsâ lives?

To improve how climate change resonates with people, we propose a simple indicator: how many extreme events (hot days and heavy precipitation days) are grandchildren projected to experience that their grandparents will not experience in their lives? We analyse the Coupled Model Intercomparison Project Phase 6 ensemble. During grandchildren's lifetime (2020â2100) under the shared socioeconomic pathway 5â8.5 (SSP5-8.5), in some tropical regions, they are projected to experience >1000 hot days and >5 heavy precipitation days breaking records set in their grandparents' lifetime until 2040. These numbers of unprecedented hot days and heavy precipitation days under SSP5-8.5 are greater in countries with lower COâ emissions and income per capita than in countries with higher COâ emissions and income per capita. We show that not only the numbers of unprecedented hot days and heavy precipitation days but also their unevenness across countries can be significantly lowered in the SSP1-2.6 scenario, which is consistent with the 2 °C goal of the Paris Agreement. This new approach would help adults easily understand how their climate change mitigation efforts could decrease the unprecedented extreme events during youths' lifetime and reduce the intergenerational and intragenerational inequalities regarding extreme events
Carbon prices, climate change mitigation & food security: How to avoid trade-offs?
Carbon price policies deliver cost-efficient mitigation across sectors, but can result in tradeoffs with food security and other sustainable development goals. Scenarios for a 1.5 °C world based on carbon prices could increase the undernourished population by 80 - 300 million in 2050. Applying a uniform carbon price across geographic regions and economic sectors has inequitable effects on countriesâ agricultural competitiveness and food availability. Under higher carbon prices, regions with poor productivity â and consequently higher GHG emissions per unit of output â would experience increased agricultural commodity prices

Burnout of Healthcare Workers Amid the COVID-19 Pandemic: A Follow-Up Study
The coronavirus disease 2019 (COVID-19) pandemic has posed a significant challenge to the modern healthcare system and led to increased burnout among healthcare workers (HCWs). We previously reported that HCWs who engaged in COVID-19 patient care had a significantly higher prevalence of burnout (50.0%) than those who did not in November 2020 (period 1). We performed follow-up surveys in HCWs in a Japanese national university hospital, including basic demographics, whether a participant engaged in care of COVID-19 patients in the past 2 weeks, and the Maslach Burnout Inventory in February 2021 (period 2) and May 2021 (period 3). Periods 1 and 3 were amid the surges of COVID-19 cases, and period 2 was a post-surge period with a comparatively small number of COVID-19 patients requiring hospitalization. Response rates to the surveys were 33/130 (25.4%) in period 1, 36/130 (27.7%) in period 2, and 56/162 (34.6%) in period 3, respectively. While no consistent tendency in the prevalence of burnout based on variables was observed throughout the periods, the prevalence of burnout tends to be higher in periods 1 and 3 in those who engaged in COVID-19 patient care in the last 2 weeks (50.0%, 30.8%, 43.1% in period 1, 2, and 3, respectively). Given the prolonged pandemic causing stigmatization and hatred against HCWs leading to increased prevalence of burnout, high-level interventions and supports are warranted
Involvement of endothelins in neuroprotection of valosin-containing protein modulators against retinal ganglion cell damage
We have previously shown that Kyoto University Substances (KUSs), valosin-containing protein (VCP) modulators, suppress cell death in retinal ganglion cells of glaucoma mouse models through alterations of various genes expressions. In this study, among the genes whose expression in retinal ganglion cells was altered by KUS treatment in the N-methyl-D-aspartic acid (NMDA) injury model, we focused on two genes, endothelin-1 (Edn1) and endothelin receptor type B (Ednrb), whose expression was up-regulated by NMDA and down-regulated by KUS treatment. First, we confirmed that the expression of Edn1 and Ednrb was upregulated by NMDA and suppressed by KUS administration in mice retinae. Next, to clarify the influence of KUSs on cell viability in relation to the endothelin signaling, cell viability was examined with or without antagonists or agonists of endothelin and with or without KUS in 661W retinal cells under stress conditions. KUS showed a significant protective effect under glucose-free conditions and tunicamycin-induced stress. This protective effect was partially attenuated in the presence of an endothelin antagonist or agonist under glucose-free conditions. These results suggest that KUSs protect cells partially by suppressing the upregulated endothelin signaling under stress conditions
